For diseases that involve the immune system, the alleles of the human leukocyte antigen (HLA) complex can play a major role. For example, if responsiveness to therapy is immunologically mediated, one would think that responders and non-responders might tend to have different HLA alleles. However, comparing the frequencies between the two groups of patients at each allele can introduce a substantial multiple comparisons problem as the number of alleles is large. This paper proposes an efficient two-stage procedure for identifying alleles that may mediate response. In the first-stage, the distri

In association studies searching for genes underlying complex traits, the results are often inconsistent, and population admixture has been recognized qualitatively as one major potential cause. Hardy-Weinberg equilibrium (HWE) is often employed to test for population admixture; however, its power is generally unknown. Through analytical and simulation approaches, we quantify the power of the HWE test for population admixture and the effects of population admixture on increasing the type I error rate of association studies under various scenarios of population differentiation and admixture. We

The human and animal trypanosomiases of Africa provide unique challenges to epidemiologists because of the spatial and temporal scales over which variation in transmission takes place. This chapter describes how our descriptions of the different components of transmission, from the parasites to the affected hosts, eventually developed to include geographical dimensions. It then briefly mentions two key analytical techniques used in the application of multi-temporal remotely sensed imagery to the interpretation of field data; temporal Fourier analysis for data reduction, and a variety of discri

For the analysis of interval-censored data, we propose a deep generalized accelerated hazards model. This model is designed to facilitate a detailed exploration of the relationship between various risk factors and the hazard associated with failure time. We develop a sieve maximum likelihood estimation procedure that combines deep neural networks and monotonic splines. By employing deep neural networks, we can effectively capture nonparametric effects, enabling a flexible and adaptive modeling approach for complex relationships. Under certain regularity conditions, we derive a nonasymptotic er

Accurate prediction of the breast cancer patient's life expectancy is essential for treatment decisions. This study aims to develop a novel model estimation and variable selection method for the partially linear additive quantile regression model when the survival times are subject to right censoring. Rather than most of the existing methods using the formulation of synthetic data points or weighting schemes to tackle censoring, we use an adapted loss function to solve censoring. Moreover, we adopt the B-spline to approximate the nonparametric additive components. To further improve the predic

Interval-censored competing risks data frequently arise in medical and clinical studies among others and furthermore, the cause of failure may be missing in some situations. In this paper, we consider regression analysis of such data under the framework of an additive subdistribution hazard model and propose a two-step sieve and weighted maximum likelihood estimation procedure. The method explicitly imposes constraints on the cumulative incidence functions to ensure valid survival function estimation and adopts an augmented inverse probability weighting strategy to address the issue of missing

It is common practice in randomized clinical trials with the standard survival outcome to follow patients until a prespecified number of events have been observed, a type of trial known as the event-driven trial. The event-driven design ensures that the target power for a specified Type I error rate is achieved to detect the target hazard ratio, regardless of the specification of other quantities. To understand the treatment effect for chronic conditions, the analysis of recurrent events has gained popularity in randomized controlled trials, particularly large-scale confirmatory trials. In the

Accurate forecasts of disease outbreaks are critical for effective public health responses, management of healthcare surge capacity, and communication of public risk. There are a growing number of powerful forecasting methods that fall into two broad categories-empirical models that extrapolate from historical data, and mechanistic models based on fixed epidemiological assumptions. However, these methods often underperform precisely when reliable predictions are most urgently needed-during periods of rapid epidemic escalation. Here, we introduce epimodulation, a hybrid approach that integrates

AIMS: Non-linear models by machine learning may identify different risk factors with different weighting in comparison to conventional linear models.
METHODS AND RESULTS: The analyses were performed in 15,933 patients included in the Shinken Database (SD) 2004-2014 (n = 22,022) for whom baseline data of blood sampling and ultrasound cardiogram and follow-up data at 2 years were available. Using non-linear models with machine learning software, 118 risk factors and their weighting of risk for all-cause mortality, heart failure (HF), acute coronary syndrome (ACS), ischemic stroke (IS), and intr

OBJECTIVES: To access medical specialists by health specialists working in the regional health field, the possibility of utilizing the voice approach for dissociative identity disorder (DID) patients as a health assessment for medical access (HAMA) was investigated. The first step is to investigate whether the plural personae in a single DID patient can be discriminated by voice analysis.
METHODS: Voices of DID patients including these with different personae were extracted from YouTube and were analysed using the software PRAAT with basic frequency, oral factors, chin factors and tongue fact

Cardiovascular events, such as hospitalizations because of congestive heart failure, often occur repeatedly in patients with CKD. Many studies focus on analyses of the first occurrence of these events, and discard subsequent information. In this article, we review a number of statistical methods for analyzing ordered recurrent events of the same type, including Poisson regression and three commonly used survival models that are extensions of Cox proportional hazards regression. We illustrate the models by analyzing data from the Chronic Renal Insufficiency Cohort Study to identify risk factors

Misclassification of breast lesions can result in either cancer progression or unnecessary chemotherapy. Automated classification tools are seen as promising second opinion providers in reducing such errors. We have developed predictive algorithms that automate the categorization of breast lesions as either benign usual ductal hyperplasia (UDH) or malignant ductal carcinoma in situ (DCIS). From diagnosed breast biopsy images from two hospitals, we obtained 392 biomarkers using Dong et al.'s (2014) computational tools for nuclei identification and feature extraction. We implemented six machine

BACKGROUND: Pneumococcal conjugate vaccines (PCVs) reduce disease largely through their impact on nasopharyngeal (NP) carriage acquisition of Streptococcus pneumoniae, a precondition for developing any form of pneumococcal disease. We aimed to estimate the vaccine efficacy (VEC) and duration of protection of PCVs against S. pneumoniae carriage acquisition through meta-regression models.
METHODS: We identified intervention studies providing NP carriage estimates among vaccinated and unvaccinated children at any time after completion of a full vaccination schedule. We calculated VEC for PCV7 se

Making accurate prognoses in chronic, complex diseases is challenging due to the wide variation in expression across individuals. In many such diseases, the notion of subtypes-subpopulations that share similar symptoms and patterns of progression-have been proposed. We develop a probabilistic model that exploits the concept of subtypes to individualize prognoses of disease trajectories. These subtypes are learned automatically from data. On a new individual, our model incorporates static and time-varying markers to dynamically update predictions of subtype membership and provide individualized

In this paper, the dynamical behavior of a viral infection model with general incidence rate and two time delays is studied. By using the Lyapunov functional and LaSalle invariance principle, the global stabilities of the infection-free equilibrium and the endemic equilibrium are obtained. We obtain a threshold of the global stability for the uninfected equilibrium, which means the disease will be under control eventually. These results can be applied to a variety of viral infections of disease that would make it possible to devise optimal treatment strategies. Numerical simulations with appli

We address the problem of long-term dynamics of tuberculosis (TB) and latent tuberculosis (LTB) in semiclosed communities. These communities are congregate settings with the potential for sustained daily contact for weeks, months, and even years between their members. Basic examples of these communities are prisons, but certain urban/rural communities, some schools, among others could possibly fit well into this definition. These communities present a sort of ideal conditions for TB spread. In order to describe key relevant dynamics of the disease in these communities, we consider a five compa

Carcass searches are a common method for studying the risk of anthropogenic hazards to wildlife, including nontarget poisoning and collisions with anthropogenic structures. Typically, numbers of carcasses found must be corrected for scavenging rates and imperfect detection. Parameters for these processes (scavenging and detection) are often estimated using carcass distribution trials in which researchers place carcasses in the field at known times and locations. In this manuscript I develop a variety of estimators based on multi-event or hidden Markov models for use under different experimenta

Anthrax poses a community health risk due to accidental or intentional aerosol release. Reliable quantitative dose-response analyses are required to estimate the magnitude and timeline of potential consequences and the effect of public health intervention strategies under specific scenarios. Analyses of available data from exposures and infections of humans and non-human primates are often contradictory. We review existing quantitative inhalational anthrax dose-response models in light of criteria we propose for a model to be useful and defensible. To satisfy these criteria, we extend an exist

The authors propose a method for the diagnostics of the form of an external impact responsible for the fracture of long tubular bones based on the morphometric investigations and mathematical modeling. The study revealed specific morphological patterns of the fracture contour and fissures in the compact bone matter reflecting the character of the blow and the force of the accompanying pressure. A set of signs is formulated for the quantitative analysis of the fracture, and a morphometric study of the destroyed zone was carried out. The data obtained were used to develop a model for diagnostics
