Age-Related Changes in Speech Recognition Performance-Intensity Functions and Auditory Brainstem Responses.

OBJECTIVES: While the mechanisms underlying age-related changes in speech recognition are thought to be multifactorial, cochlear synaptopathy has been proposed as a possible peripheral auditory contributor. Although evidence from animal and human temporal bone studies supporting the presence of age-related cochlear synaptopathy appears compelling, findings from prospective studies in humans have been less conclusive. The goals of this study were to investigate the effect of aging on measures historically used to assess retrocochlear function-speech recognition performance-intensity functions i

PRMT6 drives hearing loss by promoting asymmetric arginine dimethylation-mediated FOXG1 degradation and suppressing mitophagy.

OBJECTIVE: Age-related hearing loss (ARHL) is a progressive and irreversible sensorineural impairment with incompletely understood mechanisms. This study investigates the role of protein arginine methyltransferase 6 (PRMT6) and its regulatory mechanisms in ARHL. METHODS: In vivo, D-galactose (D-gal)-induced aging mouse models were established. Auditory brainstem response (ABR), cochlear β-galactosidase activity, and PRMT6 expression were measured, and the therapeutic effects of the PRMT6 inhibitor EPZ020411 were evaluated. In vitro, D-gal-treated HEI-OC1 cells were employed. PRMT6 was silence

Single-cell RNA sequencing analysis of mouse cochlea identifies a novel proinflammatory CD74+CD14+ macrophage subset in mice with age-related and noise-induced hearing loss.

Age-related hearing loss (ARHL) and noise-induced hearing loss (NIHL) are both associated with the activation of immune responses. However, the immune microenvironment within the cochlea in ARHL and NIHL remains poorly understood. Therefore, we profiled the immune cells in ARHL and NIHL using single-cell RNA sequencing analyses. We observed an increase in CD74+CD14+ macrophages (Mφ) in ARHL and NIHL, with this subset being associated with inflammatory pathways, immune cell chemotaxis and other processes that involved in inflammatory responses. ARHL and NIHL are linked to the degeneration of sp

Relationship between inner hair cell synaptopathy and outer hair cell loss in two mouse models of accelerated age-related hearing loss.

Hallmarks of sensorineural hearing loss are elevated hearing thresholds and defects in temporal auditory processing, the former being often caused by outer hair cell (OHC) damage, and the latter by the loss of synapses between inner hair cells (IHCs) and spiral ganglion neurons. In the well-studied CBA/CaJ mouse strain, these impairments are disconnected, IHC synaptopathy preceding OHC loss. We have investigated the relationship between IHC synaptopathy and OHC loss in the C57BL/6J (B6) and ICR mouse strains that model accelerated age-related hearing loss. Regression analysis revealed a strong

Primary Neural Degeneration in the Human Cochlea: Evidence for Hidden Hearing Loss in the Aging Ear.

The noise-induced and age-related loss of synaptic connections between auditory-nerve fibers and cochlear hair cells is well-established from histopathology in several mammalian species; however, its prevalence in humans, as inferred from electrophysiological measures, remains controversial. Here we look for cochlear neuropathy in a temporal-bone study of "normal-aging" humans, using autopsy material from 20 subjects aged 0-89 yrs, with no history of otologic disease. Cochleas were immunostained to allow accurate quantification of surviving hair cells in the organ Corti and peripheral axons of

A Large Genome-Wide Association Study of Age-Related Hearing Impairment Using Electronic Health Records.

Age-related hearing impairment (ARHI), one of the most common sensory disorders, can be mitigated, but not cured or eliminated. To identify genetic influences underlying ARHI, we conducted a genome-wide association study of ARHI in 6,527 cases and 45,882 controls among the non-Hispanic whites from the Genetic Epidemiology Research on Adult Health and Aging (GERA) cohort. We identified two novel genome-wide significant SNPs: rs4932196 (odds ratio = 1.185, p = 4.0x10-11), 52Kb 3' of ISG20, which replicated in a meta-analysis of the other GERA race/ethnicity groups (1,025 cases, 12,388 controls,

Effects of age and hearing loss on the relationship between discrimination of stochastic frequency modulation and speech perception.

OBJECTIVE: The frequency modulation (FM) of speech can convey linguistic information and also enhance speech-stream coherence and segmentation. The purpose of the present study was to use a clinically oriented approach to examine the effects of age and hearing loss on the ability to discriminate between stochastic patterns of low-rate FM and determine whether difficulties in speech perception experienced by older listeners relate to a deficit in this ability. DESIGN: Data were collected from 18 normal-hearing young adults, and 18 participants who were at least 60 years old, nine of whom had n

Association between cardiovascular health and hearing function: pure-tone and distortion product otoacoustic emission measures.

PURPOSE: A reduction in hearing sensitivity is often considered to be a normal age-related change. Recent studies have revisited prior ways of thinking about sensory changes over time, uncovering health variables other than age that play a significant role in sensory changes. METHOD: In this cross-sectional study, cardiovascular (CV) health, pure-tone thresholds at 1000 to 4000 Hz, and distortion product otoacoustic emissions (DPOAEs), with and without contralateral noise, were measured in 101 participants age 10-78 years. RESULTS: Persons in the "old" age category (49-78 years) had worse pu

Word-recognition performance in interrupted noise by young listeners with normal hearing and older listeners with hearing loss.

BACKGROUND: The most common complaint of adults with hearing loss is understanding speech in noise. One class of masker that may be particularly useful in the assessment of speech-in-noise abilities is interrupted noise. Interrupted noise usually is a continuous noise that has been multiplied by a square wave that produces alternating intervals of noise and silence. Wilson and Carhart found that spondaic word thresholds for listeners with normal hearing were 28 dB lower in an interrupted noise than in a continuous noise, whereas listeners with hearing loss experienced only an 11 dB difference.

Tracking of the genetic deafness associated to the aging in Brazilian patients.

INTRODUCTION: Presbyacusis is the most common cause of auditory dysfunction that is generally associated with aging in industrialized societies. OBJECTIVE: To assess the presence of the mitochondrial 4977-bp deletion in Brazilian patients with presbyacusis. MATERIALS AND METHODS: One hundred unrelated patients of both genders were clinically examined to exclude syndromic forms of deafness. Specific oligonucleotide primers were designed to amplify the cytochrome b gene and the 4977-bp deletion of the mtDNA using the polymerase chain reaction. RESULTS: The mtDNA(4977) deletion was not identif

Effects of aging on hearing results in tympanoplasty.

The effects of aging on the preoperative and postoperative hearing results of tympanoplasty were assessed in 642 patients with chronic suppurative otitis media (COM) or cholesteatoma (CHL). Analysis focused on the correlation between hearing results and age for each disease and type of tympanoplasty. Data were evaluated by calculating the regression line, mainly using second order polynomial regression analysis. Averaged air and bone conduction thresholds (PTA) in patients were appreciably poorer in younger patients and increased with age, compared with physiological hearing impairment in old

Machine Learning Models Can Predict Tinnitus and Noise-Induced Hearing Loss.

OBJECTIVES: Despite the extensive use of machine learning (ML) models in health sciences for outcome prediction and condition classification, their application in differentiating various types of auditory disorders remains limited. This study aimed to address this gap by evaluating the efficacy of five ML models in distinguishing (a) individuals with tinnitus from those without tinnitus and (b) noise-induced hearing loss (NIHL) from age-related hearing loss (ARHL). DESIGN: We used data from a cross-sectional study of the Canadian population, which included audiologic and demographic informati

Morphological and neurochemical changes in GABAergic neurons of the aging human inferior colliculus.

It is well known that quality of hearing decreases with increasing age due to changes in the peripheral or central auditory pathway. Along with the decrease in the number of neurons the neurotransmitter profile is also affected in the various parts of the auditory system. Particularly, changes in the inhibitory neurons in the inferior colliculus (IC) are known to affect quality of hearing with aging. To date, there is no information about the status of the inhibitory neurotransmitter GABA in the human IC during aging. We have collected and processed inferior colliculi of persons aged 11-97 yea

Age effects in discrimination of intervals within rhythmic tone sequences.

This study measured listener sensitivity to increments of a target inter-onset interval (IOI) embedded within tone sequences that featured different rhythmic patterns. The sequences consisted of six 50-ms 1000-Hz tone bursts separated by silent intervals that were adjusted to create different timing patterns. Control sequences were isochronous, with all tonal IOIs fixed at either 200 or 400 ms, while other patterns featured combinations of the two IOIs arranged to create different sequential tonal groupings. Duration difference limens in milliseconds for increments of a single sequence IOI wer

Effects of age and hearing loss on stream segregation based on interaural time differences.

The effect of interaural time differences (ITDs) on obligatory stream segregation for successive tone bursts was investigated for older listeners with normal hearing (ONH) and hearing loss (OHL), by measuring the threshold for detecting a rhythmic irregularity in an otherwise isochronous sequence of interleaved "A" and "B" tones. The A and B tones had equal but opposite ITDs from 0 to 0.5 ms. For some of the ONH listeners, the threshold increased with increasing ITD, but no OHL listener showed an effect of ITD. It is concluded that hearing loss reduces the potency of ITDs in inducing obligator

SIRT1 expression in the cochlea and auditory cortex of a mouse model of age-related hearing loss.

SIRT1 is a highly conserved NAD(+)-dependent protein deacetylase known to have protective effects against a variety of age-related diseases. However, there is a lack of information concerning SIRT1 expression in the cochlea and auditory cortex of C57BL/6 mice, a mouse model of age-related hearing loss. Using RT-PCR and immunohistochemistry, we show that SIRT1 is abundantly expressed in the inner hair cells, strial marginal cells, strial intermediate cells, type I and type IV fibrocytes of the spiral ligament and spiral ganglion neurons. In addition, moderate SIRT1 is also detected in the outer

The effect of presentation level on memory performance.

OBJECTIVES: A loss of speech intelligibility at high presentation levels is called rollover. It is a phenomenon that increases in prevalence as people age. Whether the adverse effect of high presentation levels extends to processes subsequent to speech intelligibility, such as memory, is unknown. The present study examined this question on the basis of the previous finding that older but not younger adults showed memory impairment when acoustically distorted words were presented at 50 dB SL compared with an undistorted baseline presented at 65 dB SPL. One question investigated in the present s

The factors associated with a self-perceived hearing handicap in elderly people with hearing impairment--results from a community-based study.

OBJECTIVES: Elderly persons with a physiologic hearing deficit (hearing impairment) are not necessarily socially or emotionally disturbed by the deficit in everyday life (hearing handicap). The self-perception of a hearing handicap in elderly people is a key element in seeking consultation for a hearing impairment or using hearing aids. Thus, it is important to determine the factors associated with the self-perception of a hearing handicap. The aims of the present study were to report the relation between a hearing impairment and the self-perception of a hearing handicap, and the factors assoc
