Thus far, intermediate filaments (IFs) have been the least understood of the three cytoskeletal filament systems with regard to their structure, assembly, network formation, and dynamics. This picture is now slowly but definitely changing, as recent in vivo and in vitro experiments, including generation of transgenic animals, have yielded important new data shedding light on the following areas: the molecular architecture of IFs; the role of the highly variable end domains during IF assembly and network formation; the factors that govern whether IF proteins are involved in de novo filament for

A substrain of Namalwa cells, denoted substrain B, was grown in fermentors up to the 100-L scale, and was induced with Sendai virus to produce interferon (IFN). The titer of the crude IFN varied extensively between different batches; part of the variation was caused by a differential expression of IFN-alpha and IFN-beta. More than 80% of the IFN activity was IFN-beta by several criteria. A two-step purification procedure was developed and the resulting preparation had a specific activity of approximately 10(6) U/mg protein. The IFN-beta type was found to be heterogeneous, and could be separate

The nucleotide sequence of the two minor SV40 DNA restriction fragments Hind L and Hind M is here reported. For this purpose we used 5'-32p-labeled DNA fragments either partially digested with snake venom diesterase for analysis by the wandering spot method or partially degraded with the base specific reagents dimethylsulphate or hydrazine for direct sequence analysis on gel. In the former procedure the strands were separated before degradation, while in the latter the strands were separated after modification but before the degradation. Due to the presence of several termination codons, the r

O-linked N-acetylglucosamine (O-GlcNAc) is a post-translational modification of serine and threonine residues on nuclear and cytoplasmic proteins. The identity of O-GlcNAcylated proteins during ageing and neurodegenerative disease remains incompletely defined. Thus, animal models play a crucial role for the systematic characterization and cataloguing of O-GlcNAc-modified proteins. In this study, proteomic analysis was performed to identify O-GlcNAc-modified proteins in both L1 larval and adult stages of wild-type N2 and of aex-3p::Tau(V337M) transgenic Caenorhabditis elegans, a nematode model

Apoptosis consists of highly regulated pathways involving post-translational modifications and cleavage of proteins leading to sequential inactivation of the main cellular processes. Here, we focused on the apoptotic processing of one of the essential components of the mRNA splicing machinery, the U1-70K snRNP protein. We found that at an early stage of apoptosis, before the cleavage of the C-terminal part of the protein by caspase-3, the basal phosphorylation of the Ser140 residue located within the RNA recognition motif, increases very significantly. A caspase-dependent, PP1-mediated dephosp

Transfer RNA-derived small RNAs (tsRNAs), previously considered inactive tRNA degradation products, have now been shown to be functional small non-coding RNAs. They may play important roles within the CNS and in brain-body interactions, both during normal developmental stages as well as in diverse brain pathologies. Among the cell types found in the CNS, tsRNAs are particularly abundant in neurons. Correspondingly, neurons show cell type specific tRNA expression profiles when compared to other cells of the CNS under homeostatic conditions and defects in tRNA processing may lead to neurological

BACKGROUND: Myopia, a global health burden, is associated with choroidal dysfunction, but the role of post-translational modifications, specifically S-nitrosylation (SNO), in its pathogenesis remains unclear. This study aimed to characterize choroidal S-nitrosylation and proteomic changes in lens-induced myopia (LIM) to identify SNO-driven regulatory pathways.
METHODS: Myopia was induced in guinea pigs by unilateral -10 D lens wear (LIM group), with contralateral eyes as normal controls (NC). Ocular changes were assessed by axial length (AL) and refractive error measurements. Choroidal thickn

Insoluble, hyperubiquitylated TAR DNA-binding protein of 43 kDa (TDP-43) in the central nervous system characterizes frontotemporal dementia and ALS in many individuals with these neurodegenerative diseases. The causes for neuropathological TDP-43 aggregation are unknown, but it has been suggested that stress granule (SG) formation is important in this process. Indeed, in human embryonic kidney HEK293E cells, various SG-forming conditions induced very strong TDP-43 ubiquitylation, insolubility, and reduced splicing activity. Osmotic stress-induced SG formation and TDP-43 ubiquitylation occurre

The process of aging is an integral but complex component of life that has been intensely studied for decades, from the molecular level to whole organisms. At the tissue level, bone is one of the most difficult to study due to its composite nature of inorganic and organic phases, but advancements in proteomics are enhancing our understanding of the latter, improving our understanding of skeletal aging through identifying temporal changes across the bone proteome. The relative longevity of extracellular matrix (ECM) proteins can make them more susceptible to accumulating damage modifications ov

Chronic physical restraint stress increases oxidative stress in the brain, and dysregulation of oxidative stress can be one of the causes of major depressive disorder. To understand the underlying mechanisms, we undertook a systematic proteomic analysis of hippocampus in a chronic restraint stress mouse model of depression. Combining two-dimensional gel electrophoresis (2D-PAGE) for protein separation with nanoUPLC-ESI-q-TOF tandem mass spectrometry, we identified sixty-three protein spots that changed in the hippocampus of mice subjected to chronic restraint stress. We identified and classifi

Alpha-ketoglutarate-dependent dioxygenase, also known as fat mass and obesity-associated protein (FTO), is an RNA demethylase that mediates the demethylation of N6,2-O-dimethyladenosine (m6Am) and N6-methyladenosine (m6A). Both m6Am and m6A are prevalent modifications in mRNA and affect different aspects of transcript biology, including splicing, nuclear export, translation efficiency, and degradation. The role of FTO during (herpes) virus infection remains largely unexplored. In this study, we show that the UL13 protein kinase of the alphaherpesvirus pseudorabies virus (PRV) triggers phosphor

Cardiac fibrosis is a pathological scarring process that impairs cardiac function. N-acetyltransferase 10 (Nat10) is recently identified as the key enzyme for the N4-acetylcytidine (ac4C) modification of mRNAs. In this study, we investigated the role of Nat10 in cardiac fibrosis following myocardial infarction (MI) and the related mechanisms. MI was induced in mice by ligation of the left anterior descending coronary artery; cardiac function was assessed with echocardiography. We showed that both the mRNA and protein expression levels of Nat10 were significantly increased in the infarct zone a

PURPOSE: Anorexia nervosa (AN) is a neuropsychological public health concern with a socially disabling routine and affects a person's healthy relationship with food. The role of the NNAT (Neuronatin) gene in AN is well established. The impact of mutation at the protein's post-translational modification (PTM) site has been exclusively associated with the worsening of the protein's biochemical dynamics.
METHODS: To understand the relationship between genotype and phenotype, it is essential to investigate the appropriate molecular stability of protein required for proper biological functioning.

Olfactory dysfunction is observed in several neurological disorders, including Huntington disease (HD), and correlates with global cognitive performance, depression and degeneration of olfactory regions in the brain. Despite clear evidence demonstrating olfactory dysfunction in HD patients, only limited details are available in murine models and the underlying mechanisms are unknown. In order to determine if alterations in the olfactory bulb (OB) are observed in HD we assessed OB weight or area from 3 to 12 months of age in the BACHD transgenic lines (TG5 and TG9). O-GlcNAcylation is a ubiquitous post-translational modification across the mammalian proteome, yet determining how individual O-GlcNAcylation events control specific protein functions remains a central challenge. The O-GlcNAc cycle is governed by two enzymes: O-GlcNAc transferase (OGT), which installs O-GlcNAc, and O-GlcNAcase (OGA), which removes it. Because these enzymes act on thousands of proteins, conventional methods, such as pharmacological inhibition of OGT or OGA, produce broad cellular effects that are difficult to assign to individual substrates. This gap has motivated the developme

Circadian rhythms are generated by transcriptional-translational feedback loops of clock genes and proteins and are further regulated by post-translational modifications. Among these mechanisms, casein kinase I (CKI)-mediated phosphorylation of Period (PER) proteins plays a critical role in circadian period determination by regulating PER stability, degradation, and subcellular localization. Small-molecule CKI inhibitors have been shown to lengthen circadian period; however, the relationship between CKI isoform inhibition profiles and circadian period regulation remains incompletely understood

Post-translational modifications (PTMs) have been gradually elucidated in congenital malformations such as cleft palate. Among them, O-GlcNAcylation as a dynamic PTM of proteins regulates various critical biological processes including transcription, translation, and cell fate determination. In this study, a substantial decline in O-linked β-D-N-acetylglucosamine (O-GlcNAc) levels was detected within the palatine plates of all-trans retinoic acid (atRA)-induced cleft palate mice. The role of O-GlcNAc transferase (OGT), the sole enzyme responsible for catalyzing O-GlcNAcylation, was investigate

Despite significant advances in cancer therapy, the clinical efficacy of many anticancer agents remains limited by poor selectivity, systemic toxicity, and the emergence of drug resistance, highlighting the need for novel therapeutic strategies. Among privileged heterocyclic scaffolds, indole has emerged as a versatile pharmacophore for the development of targeted anticancer agents owing to its structural diversity and broad biological activity. This review critically evaluates recent advances in indole-based anticancer agents, with particular emphasis on structure-activity relationships (SAR)

RATIONALE: Protein citrullination (deimination) is a post-translational modification of proteins converting arginine(s) into citrulline(s). "Overcitrullination" could be associated with severe pathological conditions. Mass spectrometric analysis of modified proteins is hindered by several problems. A comprehensive study of the fragmentation of deiminated peptides is not yet available. In this paper we have made an attempt to describe the characteristics of these processes, based on the studies of epitope model oligopeptides derived from clinically relevant proteins.
