Chemogenetic activation of hippocampal area CA2 promotes acute and chronic seizures in a mouse model of epilepsy.

Pyramidal cells (PCs) of hippocampal area CA2 exhibit increased excitability in temporal lobe epilepsy (TLE) and in mouse models of TLE. In epileptic mice, selective inhibition of CA2 PCs reduces chronic seizures. Here we asked if activating CA2 PCs increases seizures. Mice expressing Cre recombinase in CA2 PCs (Amigo2-Cre mice) were injected with the convulsant pilocarpine to induce a period of severe seizures (status epilepticus, SE), which leads to chronic seizures after 3-4 weeks (epilepsy). Epileptic mice were injected with a Cre-dependent adeno-associated virus (AAV) to express an excita

Rapid on-site detection of Pasteurella multocida in cattle using an LFS-based RPA-CRISPR/Cas12a assay.

BACKGROUND: Pasteurella multocida is an important bacterial pathogen associated with bovine respiratory disease (BRD), causing substantial economic losses in cattle production worldwide. Rapid field detection of P. multocida remains challenging because conventional diagnostic methods are often time-consuming and require specialized laboratory equipment. This study aimed to establish a rapid visual assay for P. multocida to support field diagnosis and disease management in cattle. METHODS: A recombinase polymerase amplification (RPA)-CRISPR/Cas12a assay targeting the species-specific kmt1 gene

CRISPR-Cas12a-assisted RT-RPA-LFA for rapid cluster IV-specific detection of cardamom mosaic virus in crude plant extracts.

Cardamom mosaic virus (CdMV), associated with mosaic/katte disease in cardamom, is a highly destructive virus that leads to severe reductions in plant growth and yield. High levels of genetic variability approaching species-level divergence exist among CdMV isolates from different geographical regions. Development of a cluster-specific diagnostic assay can enable reliable detection of locally prevalent virus populations and differentiate them from divergent ones. Therefore, in the present study, a CRISPR-Cas12a-coupled reverse transcription-recombinase polymerase amplification-lateral flow ass

Development and preliminary validation of a rapid on-site detection method for Schmallenberg virus using RT-RAA-LFD.

Schmallenberg virus (SBV) is an emerging arbovirus that causes fever, diarrhea, abortion, and congenital malformations in ruminants. The disease spreads rapidly and poses significant control challenges. Upon its identification, SBV disseminated across Europe, causing major economic losses in livestock production and international trade. Although vaccines exist in certain regions, effective treatment options remain limited, highlighting the need for rapid, early, and accurate detection. In this study, we report the development and preliminary validation of a rapid point-of-care detection method

A Streamlined Workflow for Purkinje Cell Labeling and High-Resolution Analyses of Dendrites and Spines in Mice.

Cerebellar Purkinje cells (PCs) exhibit a unique and highly complex dendritic architecture, which plays a crucial role in integrating synaptic inputs and shaping their physiological properties. However, fine morphological investigation of PCs at the single-cell level is challenging due to the densely packed soma and extensive dendritic arborization. Here, we present stepwise procedures for the sparse labeling of PCs via a viral-mediated approach, enabling detailed two-dimensional dendritic Sholl analysis and three-dimensional spine quantification. We demonstrated high-resolution labeling of PC

Suboptimal PAM Enables One-Pot RPA-CRISPR/Cas12a Detection of Clostridium perfringens in Food Samples.

The strict requirement for a canonical TTTV protospacer adjacent motif (PAM) restricts target-site selection in clustered regularly interspaced short palindromic repeats/CRISPR-associated protein 12a (CRISPR/Cas12a)-based diagnostics and limits the flexibility of assay design. Whether suboptimal PAMs can be effectively incorporated into one-pot detection systems for Clostridium perfringens has not been systematically examined. Here, we developed two one-pot recombinase polymerase amplification (RPA)-CRISPR/Cas12a assays targeting the plc and cpe toxin genes of C. perfringens and systematically

MDM2 deletion and its associated p53 activation in subcutaneous white adipose tissue induce hepatic fibrosis via galectin-3 in a mouse model.

BACKGROUND: White adipose tissue (WAT) communicates with distal metabolic organs and regulates their functions by secreting adipokines. Subcutaneous WAT (sWAT) is thought to be metabolically protective but prone to dysfunction with aging and obesity. Chronic activation of p53 is commonly observed in dysfunctional WAT at different anatomical locations in metabolic diseases. However, the impact of MDM2 deletion and its associated p53 activation in sWAT on adipokine profile and systemic metabolism is unknown. METHODS AND RESULTS: To investigate the effects of selective MDM2 deletion and its asso

The CLAMP-Linked Invasion Protein (CLIP) plays an essential role in Plasmodium berghei zoites.

Apicomplexan parasites such as Toxoplasma and Plasmodium spp. rely on the sequential secretion of parasite apical organelles, called micronemes and rhoptries, to invade host cells. The claudin-like apicomplexan microneme protein (CLAMP) is a conserved protein that plays an essential role during host cell invasion in Toxoplasma and Plasmodium zoites. Previous studies have shown that CLAMP is essential in Plasmodium merozoites for erythrocyte invasion and also in sporozoites for the invasion of the mosquito vector salivary glands and of mammalian host hepatocytes. In Toxoplasma gondii tachyzoite

Transsynaptic neural circuit mapping of ventral hippocampus motivational control systems.

The ventral hippocampus contributes to food intake regulation and a range of motivational and memory processes, and its dysfunction is associated with several cognitive and behavioral disorders. However, its circuit-level organization remains incompletely understood. Ventral CA1 (CA1v) neurons send projections to several regions involved in motivational control. Here we focus on three major forebrain targets: the nucleus accumbens shell (ACBsh), medial prefrontal cortex (mPFC), and lateral hypothalamic area (LHA). We mapped the upstream and downstream circuitry of CA1v neurons defined by their

Development of an HPV 16 rapid test founded in user-centered design with primary care clinicians.

Despite effective screening modalities, cervical cancer remains a leading cause of cancer-related death among women in the United States aged 20 to 39 years old, and incidence is rising in women aged 30-44 years old. Up to 25% of patients who are screened for cervical cancer by testing for human papillomavirus (HPV) do not receive necessary follow-up care with current laboratory-based testing. Applying a user-centered design approach, we surveyed and interviewed practicing clinicians to establish the use case, value proposition, and user requirements of a cervical cancer screening test for use

Field-Deployable Paper-Based Detection of Vibrio vulnificus via Mechanically Automated Sequential RPA/CRISPR-Cas12a.

Vibrio vulnificus (V. vulnificus) is a highly virulent marine pathogen that causes severe infections, including sepsis and necrotizing fasciitis, with high fatality rates in susceptible individuals. Rapid and sensitive on-site detection is therefore essential, yet existing methods often depend on laboratory-based equipment and trained personnel, limiting field applicability. Here, we present an automated, paper-based RPA/CRISPR-Cas12a diagnostic platform that integrates a microfluidic paper pad with a spiral-spring-driven mechanical actuator. The device enables sequential execution of recombin

Mapping First In Vivo Ebolavirus Infection Events.

Orthoebolaviruses, including Ebola virus (EBOV) and Bundibugyo virus (BDBV), cause sporadic outbreaks and devastating hemorrhagic infections in humans. A significant challenge that impedes the study of filoviruses is that the infectious virus can only be handled in biosafety level 4 (BSL4) facilities due to their extreme virulence. Life cycle modeling systems that use minigenomes and transcription/replication-competent virus-like particles (trVLPs) are powerful tools for studying these virus' biology and to explore antiviral therapies under BSL2 conditions. To date, these powerful tools have o

Evaluating Apoptotic Gene Efficiency for CHO Culture Performance Using Targeted Integration.

Chinese hamster ovary (CHO) cells have long been the favored platform for producing complex biopharmaceuticals, such as monoclonal antibodies. Cell death is a critical factor in all CHO cultures, dictating the duration until harvest in batch cultures and viable cell density in perfusion. The programmed cell death, or apoptosis, pathway has been widely studied due to its relevance in affecting cell culture performance and the extensive knowledge about its protein-to-protein interaction network. However, clonal variation seen with random integration has confounded results, and it remains unclear

Cyp19a1-Cre-EGFP: A placenta-specific Cre transgenic mouse model for targeted gene recombination in trophoblast cells.

OBJECTIVE: The placenta plays a critical role in fetal development, yet placenta-specific gene manipulation remains a challenge due to widespread gene expression across maternal and embryonic tissues. Here, we describe the generation and characterization of Cyp19a1-Cre-EGFP transgenic mice, a novel Cre-loxP model for placenta-specific gene recombination. METHODS: A Cyp19a1-Cre-EGFP construct was generated using the Cyp19a1 promoter, driving Cre recombinase expression with an enhanced green fluorescent protein (EGFP) reporter. Transgenic mice were produced via pronuclear microinjection and bac

Tissue-specific clonal selection and differentiation of CD4+ T cells during infection.

Pathogen-specific CD4+ T cells undergo dynamic expansion and contraction during infection, ultimately generating memory clones that shape the subsequent immune responses. However, the influence of distinct tissue environments on the differentiation and clonal selection of polyclonal T cells remains unclear, primarily because of the technical challenges in tracking these cells in vivo. To address this question, we generated Tracking Recently Activated Cell Kinetics (TRACK) mice, a dual-recombinase fate-mapping system that enables precise spatial and temporal labeling of recently activated CD4+

A mouse model engineered to spatiotemporally control Cre expression in progesterone receptor positive cells†.

The Cre/loxP system is widely used for site-specific genetic manipulation in mice. The PgrCre mouse model, where Cre recombinase is driven by the progesterone receptor promoter, is commonly used for gene ablation in Pgr-positive uterine cells. However, the PgrCre is active in the neonatal uterus and does not allow temporal control. To enhance the functionality of the PgrCre mouse, we generated and characterized an inducible PgriCreERT2 mouse, in which iCreERT2 is inserted downstream of the endogenous Pgr promoter. PgriCreERT2 mice crossed with Rosa26-CAG-LSL-Sun1-sfGFP-myc reporter mice demons

Advancing Laboratory Diagnostics for Future Pandemics: Challenges and Innovations.

Since the beginning of the 21st century, major epidemics and pandemics such as SARS, H1N1pdm09, Ebola, and COVID-19 have repeatedly challenged global systems of disease diagnostics and control. These crises exposed the weaknesses of traditional diagnostic models, including long turnaround times, uneven resource distribution, and supply chain bottlenecks. As a result, there is an urgent need for more advanced diagnostic technologies and integrated diagnostics strategies. Our review summarizes key lessons learned from four recent major outbreaks and highlights advances in diagnostic technologies

Hepatic HSD17B6 is dispensable for diet-induced fatty liver disease in mice.

Metabolic dysfunction-associated fatty liver disease (MAFLD) affects up to a third of the global population, which causes huge both clinical and economic burdens. However, its therapeutic strategy is still limited. Steroid dysregulation plays a pivotal role in the homeostasis of lipid metabolism. 17-beta-hydroxysteroid dehydrogenase type 6 (HSD17B6)-one member of 17β-HSDs, encoded by the gene Hsd17b6, catalyzes the synthesis of androsterone and estrone-steroid hormones. However, whether the manipulation of HSD17B6 could ameliorate diet-induced fatty liver disease remains unknown. Here, we foun
