Age-related differences in the cutaneous vascular response to exogenous angiotensin II.

Angiotensin II (ANG II) is locally produced in human skin and contributes to the reflex vasoconstriction (VC) response in aged but not young skin. We hypothesized that the exogenous ANG II-mediated VC response would be greater in older adults and would be affected by inhibition of adrenoreceptor or ANG II type II receptor (AT2R) pathways. Three microdialysis (MD) fibers were placed in the forearm skin of 11 young (26 ± 3 yr) and 11 older (68 ± 4 yr) individuals for perfusion of 1) Ringer solution (control), 2) adrenoreceptor blockade with yohimbine + propranolol, and 3) AT2R inhibition with PD

Beneficial effect of cyclosporine A on traumatic hemorrhagic shock.

BACKGROUND: Vascular hyporeactivity plays an important role in severe trauma and shock. We investigated the beneficial effect of cyclosporine A (CsA) on traumatic shock and its relationship to vascular reactivity improvement and mitochondrial permeability transition pore (MPTP). MATERIALS AND METHODS: Sodium pentobarbital-anesthetized rats were used to induce traumatic hemorrhagic shock by left femur fracture and hemorrhage, the beneficial effects of CsA (1, 5, and 10 mg/kg, intravenously) on animal survival, cardiovascular function, tissue blood perfusion, and mitochondrial function of vital

The role of free radicals and nitric oxide in the ischemia-reperfusion injury mediated by acute bladder outlet obstruction.

Free-radical generation and nitric oxide (NO) generation were detected in the rat bladder following acute bladder outlet obstruction (BOO), and the results were compared with those for vascular ischemia and reperfusion (I-R). Forty male Sprague Dawley rats were used. In the acute BOO plus I-R group (group 1), rats were catheterized with a 3-Fr catheter and an inflated balloon was positioned at the bladder neck. The bladder was overdistended after administration of Ringer solution and furosemide (12 mg/kg, each) for 60 min, and was then drained to allow reperfusion for 30 min. In the acute BOO

Modification of sodium channel gating in frog myelinated nerve fibres by Centruroides sculpturatus scorpion venom.

1. The effect of Centruroides sculpturatus scorpion venom on single frog myelinated nerve fibres was studied. Sodium currents through the nodal membrane were measured under voltage-clamp conditions before and after exposure to venom in Ringer solution 1-5 mug/ml. for 1-3 min. 2. Centruroides venom brings about repetitive firing and increased membrane potential noise. Spontaneous firing was also observed. Eventually the nodal membrane becomes inexcitable following venom treatment. 3. Under voltage clamp with a step depolarization of the membrane potential, activation and inactivation of sodium

Hemodynamics of respiratory failure in rabbit model: effect of surfactant supplementation.

In some infants, administration of surfactant has been associated with an acute decrease in blood pressure. We hypothesized that independent of patent ductus arteriosus, low blood volume sensitizes to a negative hemodynamic response to surfactant supplementation. Respiratory failure was induced by bronchoalveolar lavage (BAL) in 30 young rabbits that were paralyzed and ventilated using tidal volumes of 10 ml/kg. After BAL, 15 ml/kg blood was withdrawn while the same volume of one of the following was infused: Ringer's lactate (n = 12); 5% albumin (n = 5); or leukocyte-free red blood cells (RBC

Automated hemodynamic resuscitation system for computer-controlled norepinephrine, vasopressin, and fluid therapy in endotoxin-induced shock: a feasibility and physiological proof-of-concept study.

BACKGROUND: To support hemodynamic management in vasoplegic shock, we previously developed a closed-loop automated infusion system of norepinephrine (NE) and fluid to automatically restore arterial pressure (AP) and cardiac output (CO). To enhance the practical feasibility and efficacy of the system, we integrated a minimally invasive CO monitor and a closed-loop control of vasopressin (AVP) infusion to the system and validated the integrated system in dogs with endotoxin-induced shock. METHODS: Based on the circulatory equilibrium framework, the system directly controls systemic vascular res

Interventional Management of Acute Pancreatitis and Its Complications.

Acute pancreatitis (AP) is the most common cause of gastrointestinal-related hospitalizations in the United States, with gallstone disease and alcohol as the leading etiologies. Management is determined by disease severity, classified as interstitial edematous pancreatitis or necrotizing pancreatitis, with severity further stratified based on local complications and systemic organ dysfunction. Regardless of etiology, initial treatment involves aggressive intravenous fluid resuscitation with Lactated Ringer's solution, pain and nausea control, early oral feeding in 24 to 48 h, and etiology-dire

A 3-O-sulfated heparan sulfate dodecasaccharide (12-mer) suppresses thromboinflammation and attenuates early organ injury following trauma and hemorrhagic shock.

INTRODUCTION: Dysregulated inflammation and coagulation are underlying mechanisms driving organ injury after trauma and hemorrhagic shock. Heparan sulfates, cell surface glycosaminoglycans abundantly expressed on the endothelial surface, regulate a variety of cellular processes. Endothelial heparan sulfate containing a rare 3-O-sulfate modification on a glucosamine residue is anticoagulant and anti-inflammatory through high-affinity antithrombin binding and sequestering of circulating damage-associated molecular pattern molecules. Our goal was to evaluate therapeutic potential of a synthetic 3

Randomized, placebo-controlled trial of lazaroid effects on severe acute pancreatitis in rats.

OBJECTIVES: To evaluate the therapeutic potential of lazaroids in severe necrotizing acute pancreatitis and to investigate the association between oxidative stress, protease activation, and local production of proinflammatory cytokines and the severity and lethality of the disease. BACKGROUND: Oxidative stress is a crucial factor in the pathophysiology of acute pancreatitis and its systemic complications. Treatment with antioxidants, however, failed to improve survival in most studies performed so far. Lazaroids are a novel class of antioxidants that potently protect pancreatic acinar cells a

Impact of Pre-Hospital Intravenous Infusion on Physiological Parameters in Severe Trauma Patients.

Introduction The relationship between pre-hospital intravenous (IV) infusions administered by emergency life-saving technicians (ELSTs) to trauma patients in shock and the resulting variability in their vital signs before hospital arrival remains unclear. In 2014, Japan approved the use of lactated Ringer's solution via IV by ELSTs for patients aged 15 and older with non-cardiac arrest and shock symptoms not caused by cardiogenic factors. However, the impact of pre-hospital IV infusions on physiological parameters in severely injured trauma patients is still unknown. Aim The aim of this study

Skin pigmentation is negatively associated with circulating vitamin D concentration and cutaneous microvascular endothelial function.

Darkly pigmented individuals are at the greatest risk of hypovitaminosis D, which may result in microvascular endothelial dysfunction via reduced nitric oxide (NO) bioavailability and/or increased oxidative stress and inflammation. We investigated the associations among skin pigmentation (M-index; skin reflectance spectrophotometry), serum vitamin D concentration [25(OH)D], circulating inflammatory cytokine (TNF-α, IL-6, and IL-10) concentrations, and the NO contribution to local heating-induced cutaneous vasodilation (%NO-mediated vasodilation) in a diversely pigmented cohort of young adults.
