Do children born after embryo vitrification differ from siblings born after fresh embryo transfer?

OBJECTIVE: To assess neonatal outcomes in siblings born after frozen embryo transfer (FET) in comparison with siblings born after fresh embryo transfer. DESIGN: Prospective cohort study. Linear mixed models and generalized linear mixed models were used to study the association between outcomes after FET vs. fresh embryo transfer, considering the correlation of observations from siblings and controlling for treatment and maternal characteristics. Subgroup analysis according to the day of fresh embryo transfer and according to the FET protocol was also performed. SUBJECTS: Sibling pairs of sin…

Late Diagnosis of NBEAL2-related Gray Platelet Syndrome in Finnish Siblings with Lifelong Thrombocytopenia.

In the case of familial thrombocytopenia, a congenital defect should be considered; however, in particular older patients who have had thrombocytopenia for a long time have often not yet been genetically analyzed using modern sequencing methods. Remarkably, sometimes they are still suspected of suffering from chronic immune thrombocytopenia until genetic testing reveals a congenital defect. We report on elderly Finnish siblings (both older than 60 years) with lifelong thrombocytopenia. The lifelong bleeding tendency in both the siblings was usually treated with tranexamic acid and platelet tra…

DBR1 Gene Mutation: Pathogenicity in the Homozygous State and Its Phenotype in Two Siblings.

The RNA lariat debranching enzyme (DBR1) facilitates the hydrolysis of 2'-5' prime branched phosphodiester bonds. It acts on the bonds at the junction of excised lariat intron RNA, converting them into linear molecules for degradation. Mutations in the gene result in the accumulation of lariat introns, leading to multiple system dysfunction. This case involves two siblings who exhibited a homozygous gene mutation in DBR1, identified as the variant c.200A>G p.(Tyr67Cys). Both showed similar manifestations, including premature birth, intrauterine growth restriction, growth deficiency, ichthyosis…

Rare C1q deficiency presenting as pediatric SLE: A case study of two consanguineous siblings.

C1q deficiency is a rare autosomal recessive genetic condition characterized by mutations in genes C1qA, C1qB, or C1qC which can cause a SLE-like disease. Here, we report the cases of two siblings with C1q deficiency, both of whom had homozygous mutations in the C1QA gene. Both of our patients had NP involvement, and the brother had chilblain lesions. Diagnosis of C1q deficiency was delayed, highlighting the importance of clinical suspicion and genetic testing. This is especially crucial in cases with atypical presentations of SLE and a family history of consanguinity.

POLR3B-Related Hypomyelinating Leukodystrophy Type 8 (4H Syndrome): A Case Series of Two Siblings.

We present a case series of two siblings from a consanguineous family with genetically confirmed POLR3B-related hypomyelinating leukodystrophy type 8 (4H syndrome), a rare autosomal recessive disorder characterized by hypomyelination, hypodontia, and hypogonadotropic hypogonadism. Despite sharing the same homozygous mutation, the siblings exhibited distinct clinical phenotypes, with the older child presenting with severe motor dysfunction, optic disc pallor, and requiring orthopedic surgery, while the younger maintained independent ambulation with milder neurological symptoms. Both exhibited h…

Comparative study of enriched dopaminergic neurons from siblings with Gaucher disease discordant for parkinsonism.

Inducible pluripotent stem cells (iPSCs) derived from patient samples have significantly enhanced our ability to model neurological diseases. Comparative studies of dopaminergic (DA) neurons differentiated from iPSCs derived from siblings with Gaucher disease discordant for parkinsonism provides a valuable avenue to explore genetic modifiers contributing to GBA1-associated parkinsonism in disease-relevant cells. However, such studies are often complicated by the inherent heterogeneity in differentiation efficiency among iPSC lines derived from different individuals. To address this technical c…

Serum free light chains among twin siblings: is the kappa/lambda ratio genetically determined?

BACKGROUND: Serum kappa, lambda, the K/λ light chain concentrations are used for screening, diagnosis, and monitoring of patients with multiple myeloma and other plasma cell disorders. Biological variation studies conducted on healthy subjects showed that free light chains have a low within and high between-individual variation. We determined if this variation were genetically linked. METHODS: We obtained a single serum sample from 16 pairs of identical twins, 8 neonate twins, and 19 presumed directly-related siblings children, measured Κ and λ light chains and computed the Κ/λ ratio. RESULT…

Three siblings with self-limited familial infantile epilepsy with PRRT2 mutation: A case series.

We report three sisters with self-limited familial infantile epilepsy, caused by a mutation in proline-rich transmembrane protein2. Self-limited familial infantile epilepsy has been established as a distinct epileptic syndrome characterized by focal seizures in clusters of infantile-onset. The seizure types of our cases were focal with or without secondary generalization. The seizures manifested at 3-5 months of age, and each lasted 1-2 min. All three sisters fulfilled the criteria for self-limited familial infantile epilepsy, except in one case who showed interictal spikes in the right centra

Musculocontractural type of Ehlers-Danlos syndrome with novel CHST14 pathogenic variant in two siblings.

Musculocontractural Ehlers-Danlos syndrome (MC-EDS) is a rare entity worldwide with underlying pathogenic variant in the carbohydrate sulfotransferase 14 (CHST14) gene. Previous reports of the same entity from India were of two unrelated cases. Ours is the first report of two siblings in an Indian family with craniofacial dysmorphism and distal arthrogryposis with a clinical diagnosis of EDS, where an underlying pathogenic variant in CHST14 was detected by exome sequencing.

Resting state functional connectivity in adolescents with substance use disorder and their unaffected siblings.

We aimed to examine resting-state functional connectivity (rsFC) in adolescents with substance use disorder (SUD) and their unaffected biological siblings (SIB), relative to typically-developing controls (TDC) in order to identify alterations in functional network organization that may be associated with the familial risk for SUD. Resting-state functional magnetic resonance imaging analysis included 20 adolescents with SUD, 20 SIB, and 18 TDC. Network-based analysis revealed that adolescents with SUD had significantly both weaker and higher rsFC compared to TDC mainly within the default-mode n…

Understanding the novel MCM8 gene mutation: primary ovarian insufficiency and uterine hypoplasia in siblings.

This case report elucidates a scenario involving two sibling sisters born out of consanguineous marriage-one initially presenting with lower respiratory infection, concurrently exhibiting short stature and primary amenorrhoea. Investigation into the primary amenorrhoea unveiled hypergonadotropic hypogonadism, confirmed by the absence of ovaries and a hypoplastic uterus on pelvic MRI. Genetic analysis via whole exome sequencing identified a homozygous variant NM_001282717.2: c.808C>T in the MCM8 gene, located on exon 8 of chromosome 20, inherited in an autosomal recessive manner. The scarcity o

Long-term outcomes in two adult siblings with Fucosidosis - Diagnostic odyssey and clinical manifestations.

Fucosidosis (OMIN# 230000) is a rare lysosomal storage disorder (LSDs) caused by mutations in the FUCA1 gene, leading to alpha-L-fucosidase deficiency; it is inherited as an autosomal recessive trait. Fucosidosis represents a disease spectrum with a wide variety of clinical features, but most affected patients have slow neurologic deterioration. Many patients die young and the long-term clinical outcomes in adult patients are poorly documented. Here, we report the long-term follow up of two Caucasian siblings, a 31-year-old man and 25-year-old woman. We describe the clinical, biochemical, radi…
