Chalk Yeasts Cause Gluten-Free Bread Spoilage.

Four different yeast strains were isolated from industrial gluten-free bread (GFB) purchased from a local supermarket. These strains, including Hyphopichia burtonii, Wickerhamomyces anomalus, Saccharomycopsis fibuligera, and Cyberlindnera fabianii, are responsible for spoilage, which consists of white powdery and filamentous colonies due to the fragmentation of hyphae into short-length fragments (dust-type spots) that is typical of the spoilage produced by chalk yeasts. The isolated strains were identified using genomic analysis. Among them, C. fabianii was also isolated, which is a rare ascom

Partial replacement of high-fat diet with n-3 PUFAs enhanced beef tallow attenuates dyslipidemia and endoplasmic reticulum stress in tunicamycin-injected rats.

INTRODUCTION: Metabolic syndrome (MetS) is considered as a complex, intertwined multiple risk factors that directly increase the risk of various metabolic diseases, especially cardiovascular atherosclerotic diseases and diabetes mellitus type 2. While lifestyle changes, including dietary intervention are effective in mitigating or preventing MetS, there are no specific therapies against MetS. Typical western diets comprise of high saturated fatty acid, cholesterol, and simple sugar; consequently their consumption may increase the potential pathological developmental risk of MetS. Partial repla

An improved method for extraction of polar and charged metabolites from cyanobacteria.

A key requirement for 13C Metabolic flux analysis (13C-MFA), a widely used technique to estimate intracellular metabolic fluxes, is an efficient method for the extraction of intermediate metabolites for analysis via liquid chromatography mass spectrometry (LC/MS). The 13C isotopic labeling results in further distribution of an already sparse pool of intermediate metabolites into isotopologues, each appearing as a separate chromatographic feature. We examined some of the reported solvent systems for the extraction of polar intracellular metabolites from three strains of cyanobacteria of the gen

Glycosylation of erythrocyte spectrin and its modification in visceral leishmaniasis.

Using a lectin, Achatinin-H, having preferential specificity for glycoproteins with terminal 9-O-acetyl sialic acid derivatives linked in α2-6 linkages to subterminal N-acetylgalactosamine, eight distinct disease-associated 9-O-acetylated sialoglycoproteins was purified from erythrocytes of visceral leishmaniaisis (VL) patients (RBC(VL)). Analyses of tryptic fragments by mass spectrometry led to the identification of two high-molecular weight 9-O-acetylated sialoglycoproteins as human erythrocytic α- and β-spectrin. Total spectrin purified from erythrocytes of VL patients (spectrin(VL)) was re

Roles of unsaturated fatty acids (especially omega-3 fatty acids) in the brain at various ages and during ageing.

Among various organs, in the brain, the fatty acids most extensively studied are omega-3 fatty acids. Alpha-linolenic acid (18:3omega3) deficiency alters the structure and function of membranes and induces minor cerebral dysfunctions, as demonstrated in animal models and subsequently in human infants. Even though the brain is materially an organ like any other, that is to say elaborated from substances present in the diet (sometimes exclusively), for long it was not accepted that food can have an influence on brain structure, and thus on its function. Lipids, and especially omega-3 fatty acids

Comparative transcriptome analysis reveals aggravated damage on rice plants by brown planthoppers under elevated CO2.

The increase of atmospheric CO2 concentration will inevitably affect the photosynthesis of C3 plants, thus affecting herbivorous insects. Previous studies have shown that elevated CO2 (eCO2) may aggravate damage of brown planthopper (BPH) Nilaparvata lugens for rice plants. However, the molecular mechanism of this phenomenon is unclear. The comparative transcriptome analysis combined with corresponding phenotypic changes of rice plants and BPHs under ambient CO2 (aCO2) and eCO2 were studied to reveal the molecular mechanism of aggravated damage. The contents of soluble sugar and free fatty aci

Characterization and role of the 3-methylglutaconyl coenzyme A hidratase in Trypanosoma brucei.

Trypanosoma brucei, the agent of African Trypanosomiasis, is a flagellated protozoan parasite that develops in tsetse flies and in the blood of various mammals. The parasite acquires nutrients such as sugars, lipids and amino acids from their hosts. Amino acids are used to generate energy and for protein and lipid synthesis. However, it is still unknown how T. brucei catabolizes most of the acquired amino acids. Here we explored the role of an enzyme of the leucine catabolism, the 3-methylglutaconyl-Coenzyme A hydratase (3-MGCoA-H). It catalyzes the hydration of 3-methylglutaconyl-Coenzyme A (

Identification of primary and secondary metabolites with phosphorus status-dependent abundance in Arabidopsis, and of the transcription factor PHR1 as a major regulator of metabolic changes during phosphorus limitation.

Massive changes in gene expression occur when plants are subjected to phosphorus (P) limitation, but the breadth of metabolic changes in these conditions and their regulation is barely investigated. Nearly 350 primary and secondary metabolites were profiled in shoots and roots of P-replete and P-deprived Arabidopsis thaliana wild type and mutants of the central P-signalling components PHR1 and PHO2, and microRNA399 overexpresser. In the wild type, the levels of 87 primary metabolites, including phosphorylated metabolites but not 3-phosphoglycerate, decreased, whereas the concentrations of most

Reduction in the fitness of Bemisia tabaci fed on three previously infested tomato genotypes differing in the jasmonic acid pathway.

The effect of previous infestation (preconditioning) by the whitefly Bemisia tabaci (Gennadius) biotype B on the population fitness of subsequent infestations that fed on three isogenic tomato genotypes (wild-type [Wt], a jasmonic acid [JA] defense-enhanced genotype [35S], and a JA-deficient genotype [spr2]) was examined. We tested the hypotheses that whiteflies fed on preconditioned tomatoes (Solanum lycopersicum L.) would have reduced fitness and that the effect would be mediated via the JA-dependent systemic plant defense pathway. Preconditioning by the whitefly resulted in decreased levels

Clinical application of metabolomics in neonatology.

The youngest and more rapidly increasing "omic" discipline, called metabolomics, is the process of describing the phenotype of a cell, tissue or organism through the full complement of metabolites present. Metabolomics measure global sets of low molecular weight metabolites (including amino acids, organic acids, sugars, fatty acids, lipids, steroids, small peptides, vitamins, etc.), thus providing a "snapshot" of the metabolic status of a cell, tissue or organism in relation to genetic variations or external stimuli. The use of metabolomics appears to be a promising tool in neonatology. The ma

Extracellular invertase LIN6 of tomato: a pivotal enzyme for integration of metabolic, hormonal, and stress signals is regulated by a diurnal rhythm.

To elucidate the regulation pattern of extracellular invertase LIN6 of tomato, the corresponding promoter has been cloned and the sink-tissue specific expression and its regulation by sugars, stress stimuli, growth regulators, and the diurnal rhythm is shown. The in situ analysis of transgenic tobacco plants expressing a LIN6 promoter::beta-glucuronidase reporter gene fusion demonstrates LIN6 expression in sink tissues, such as pollen grains and vascular tissues of leaves and stems. LIN6 is up-regulated in close proximity to wounded tissue, and by methyl jasmonate and abscisic acid, global sig

Azide-Alkene Cycloaddition and Visible-Light-Mediated Wolff Rearrangement of Sugar Enones.

The synthesis of branched carbohydrate β-amino esters is described using the Wolff rearrangement of a diazoketone derived from the pyrolysis product (-)-levoglucosenone (LGO). An azide-alkene cycloaddition on LGO catalyzed by methanesulfonic acid directly afforded β-amino diazoketones via the triazoline. A subsequent visible-light-mediated Wolff rearrangement resulted in ring-contraction, giving a series of branched deoxyamino sugars with excellent diastereoselectivity (>20:1). The use of 2,2,2-trifluoroethanol as solvent at subambient temperature improved the yields of the ester relative to r

Diagnostic and therapeutic significance of selected antibodies against advanced glycation end-products (anti-AGEs).

The aim of this review is to summarize current knowledge on the diagnostic and therapeutic significance of antibodies against advanced glycation end-products (anti-AGEs). AGEs are formed through non-enzymatic reactions of reducing sugars with proteins, lipids, and nucleic acids, and their accumulation has been implicated in the pathogenesis of chronic diseases such as diabetes, atherosclerosis, cancer, and neurodegenerative disorders. This paper discusses anti-MGO modified proteins, anti-imidazolone, anti-pentosidine, anti-CML (including SIWA318H), anti-CEL, anti-CMA, and anti-GA-pyridine anti

Exploring the rich heritage and health benefits of diverse fruit wines and their production.

Wine is an alcoholic beverage made from fermented grapes or other fruits. The fermentation process occurs when yeast consumes sugars in the fruit and converts them into alcohol and carbon dioxide. Wine has been produced for thousands of years, with different regions developing distinct varieties based on the local grapes, climate, and production techniques. Key types of wine include red, white, rosé, sparkling, and fortified wines. In addition to its cultural and culinary significance, wine contains compounds like antioxidants, which have been studied for potential health benefits when consume
