Tetrachloroethylene: balance and tissue distribution in male Sprague-Dawley rats by drinking-water administration.

While low levels of organic contaminants have been reported to occur in drinking water, often little information is available concerning the toxicological significance of ingestion by this route. For tetrachloroethylene, considerable animal toxicity data exist for gavage or inhalation exposure. Since extrapolation of toxicity between routes of administration implies similar pharmacokinetic disposition, the pharmacokinetics of tetrachloroethylene was examined in rats following administration in the drinking water. Tetrachloro[14C]ethylene was administered to adult, male Sprague-Dawley rats in s…

Social anxiety, daily moods, and alcohol use over time among heavy social drinking men.

To investigate the tension reduction hypothesis of alcohol use in the natural environment and with several forms of tension, 36 heavy social drinking young men kept daily records of their drinking, anxiety, unhappiness, and anger for 3 months and for 1 month from 6 to 7 months from the start of the study. No significant correlations were found between the frequency and intensity of any of the daily moods and drinking rates or intoxication frequency, either concurrently or within a few days or weeks. Drinking was also unrelated to general trait anxiety, depression, stressful life experiences, a…

Increased alcohol selection in rats after alcohol drinking paired with recovery from thiamine deficiency.

Repeated pairings of novel alcohol solutions (5% or 7.5% w/v) with IP injections of 267 microgram/kg thiamine in thiamine-deficient male Sprague-Dawley rats resulted in the ingestion of pharmacologically active doses of alcohol in association with recovery. Mean alcohol intake in a postrecovery fluid choice situation exceeded metabolic capacity for 7--10 days; such intake was not observed in nondeficient pair-fed rats or in formely deficient rats whose recovery was not paired with alcohol drinking. Alcohol was self-selected in the presence of water under conditions where the rats had no experi…

Global trifluoroacetic acid hotspots in aquatic environments: Drinking-water risk and supply-chain responsibility.

Trifluoroacetic acid (TFA) is the most abundant per- and polyfluoroalkyl substance (PFAS) in aquatic environments, posing increasing threats to water quality and potential human exposure. However, its global risk profile and underlying drivers remain poorly understood. Herein, we integrate a CatBoost-based geospatial classifier achieving cross-validated F1 score of 0.70 and test-set sensitivity of 0.77 with an extended environmental multi-regional input-output (EE-MRIO) model to map global TFA exceedance risk in aquatic environments and trace its supply-chain drivers. We identify prominent TFA

Longer-term changes in drinking behaviour after an alcohol abstinence challenge: a longitudinal study.

AIMS: In the past years, temporary alcohol abstinence challenges (TACs) are rising in popularity. Participation may lead to changes in drinking behaviour in the short- and longer-term. To evaluate the short- and longer-term changes in drinking behaviour after participating in NoThanks (Dutch TAC), this study aimed to examine changes in drinking refusal self-efficacy (DRSE), behavioural automaticity, craving, and alcohol consumption. METHODS: In this longitudinal study, data were used from 2,128 participants in NoThanks 2023. Participants completed a baseline questionnaire, a direct follow-up,…

Effects of salt (NaCl) drinking water on physiological and productivity responses of dairy cattle.

Climate change is increasing the risk of freshwater scarcity in dairy production, highlighting the need to evaluate alternative water sources. This study investigated the physiological and production responses of dairy cows offered drinking water with salinity comparable to that of the brackish Baltic Sea (salinity 0.2 - 0.9%). First, a pilot study was conducted to assess acute responses (48 h) to a near-isotonic solution (0.8% NaCl, i.e., 8 g NaCl/kg) and to inform sampling times for the main study. In the main study, 18 multiparous cows (208 ± 22 d in milk, 35.5 ± 4.7 kg milk yield/d) were a…

Parental alcohol supply in early childhood and adolescent drinking: Evidence from a prospective cohort.

BACKGROUND: Parental alcohol supply in early childhood may increase the risk of alcohol use in late adolescence. This study examined its longitudinal impact and the distinct roles of mothers' and fathers' drinking. METHODS: We studied 1,891 mother-child pairs from the Czech European Longitudinal Study of Pregnancy and Childhood. Mothers reported parental alcohol supply at ages 3, 5, 7, and 11 years, while adolescent alcohol use was reported by mothers, pediatricians, and youth at ages 18 and 19 years. Structural equation modeling assessed the longitudinal link between early alcohol supply (th…

Measurement challenges in binge drinking research: Addressing sex and gender diversity in national surveys.

BACKGROUND: Accurate measurement of binge drinking is essential for understanding population-level health disparities. However, national surveys commonly rely on binary sex-based thresholds, which may introduce classification challenges when applied across diverse sex and gender classifications. METHODS: Data from the 2019-2023 Behavioral Risk Factor Surveillance System were used to examine binge drinking patterns among adults reporting transgender identity. Binge drinking was defined using standard sex-based thresholds (≥5 drinks for men, ≥4 drinks for women on a single occasion). Multivaria…

Occurrence of PFAS in municipal drinking water: a participatory case study in London, UK.

Globally, per- and poly-fluoroalkyl substances (PFAS) contamination has been reported in numerous environmental matrices, and there is a growing body of evidence that links PFAS exposure to adverse health effects. Consuming contaminated drinking water is potentially one of the most common routes of human exposure from these compounds. The spatial and temporal variability of 38 PFAS in 210 household tap water and public water fountains were assessed using a participatory sampling campaign in London, UK. The performance of commercially available water filters to remove PFAS was also assessed. In…

Social media, drinking motives, and alcohol use in young adults: a longitudinal mediation study.

Alcohol use remains a significant problem among young adults (YAs), and growing evidence suggests exposure to alcohol-related content (ARC) on social media may contribute to increased alcohol use. ARC often portrays drinking as rewarding and/or normative, potentially shaping drinking motives, which are key predictors of alcohol use. Therefore, the present study sought to longitudinally examine if drinking motives mediate the associations between (a) exposure to peers' ARC on social media and (b) perceived number of friends posting ARC and YA alcohol use over an 8-week period. A sample of N = 2…

Associations Between Sociosexuality and Intoxicated Sex and Unprotected Sex: Mediating Effects of Hazardous Drinking.

BACKGROUND: Sociosexuality reflects behaviors, attitudes, and desires related to casual sex with multiple, concurrent, novel partners. Research on whether people high in sociosexuality are less likely to engage in safer sex practices (e.g., unprotected sex) is mixed, but there is evidence that those high in sociosexuality consume more alcohol than their peers, and drinking is associated with less likelihood of engaging in safer sex. Here we tested whether sociosexuality is associated with intoxicated and unprotected sex in a sample of college students (Study 1) and adults (Study 2). METHODS: …

Young Adults' Perspectives on an Ecological Momentary Intervention for Drinking to Cope: Qualitative Study.

BACKGROUND: Young adults have high rates of mental health problems, such as mood or anxiety symptoms, and high rates of problematic drinking. Many young adults who undergo psychiatric hospitalization to address depression and anxiety symptoms also engage in risky drinking and tend to drink to cope with negative emotions. However, in many cases, treatment programs focusing on mood and anxiety symptoms often fail to adequately address problematic alcohol use in young adults. OBJECTIVE: This study aimed to address this treatment gap by investigating patient perspectives on a potential ecological…

Alpha-2 Adrenergic Agonists Reduce Heavy Alcohol Drinking and Improve Cognitive Performance in Mice.

Alcohol use disorder (AUD) is one of the top behavioral causes of global disease burden in the United States. Repeated cycles of alcohol intoxication and abstinence induce neuroplastic alterations which induce excessive drinking and cognitive impairments. A system deeply dysregulated by chronic drinking is norepinephrine (NE). At moderate levels, NE has beneficial effects on cognition and behavior, mediated by the α2 adrenergic receptor (AR) subtype. Whether α2 AR activation blunts alcohol consumption in models of heavy drinking has not been determined, and whether α2 AR activation improves co…

Sonoplasma treatment of drinking water reduces yeast culturability both immediately and progressively during holding.

Sonoplasma treatment (SPT) combines hydrodynamic cavitation with electrical discharge, generating reactive oxygen species (ROS) and local mechanical stress in water. In this study, six yeast species (Candida albicans, C. parapsilosis, C. tropicalis, Naganishia albida, Rhodotorula mucilaginosa, and Tausonia pullulans) were used as eukaryotic model organisms to assess SPT efficacy in drinking water and to distinguish immediate from delayed ("treat-and-hold") effects. Standardized suspensions (1.0-1.1 McFarland) were inoculated into 9 L of drinking water and processed at 30 kHz using 1-10 consecu…

RIPK3/MLKL-mediated necroptosis promotes hepatic inflammation in chronic arsenic exposure through drinking water.

Chronic exposure to arsenic-contaminated drinking water is a major environmental risk factor for liver injury, yet its pathogenic mechanisms remain unclear. This study explores the role of RIPK3/MLKL-mediated necroptosis in arsenic-induced hepatotoxicity using a chronic in vivo mouse model. Mice were exposed to sodium arsenite (NaAsO2) at concentrations of 2.8, 8.4, and 25.2 mg/L in drinking water for 8 weeks. Arsenic exposure induced dose-dependent hepatocellular necrosis, elevated liver injury markers (ALT, AST, GST-α, GDH), and increased hepatic expression of p-RIPK3, p-MLKL and HMGB1, alon…
