Sulfation is an important biological process that modulates the function of numerous molecules. It is directly mediated by cytosolic and Golgi sulfotransferases, which use 3'-phosphoadenosine 5'-phosphosulfate to produce sulfated acceptors and 3'-phosphoadenosine 5'-phosphate (PAP). Here, we identify a Golgi-resident PAP 3'-phosphatase (gPAPP) and demonstrate that its activity is potently inhibited by lithium in vitro. The inactivation of gPAPP in mice led to neonatal lethality, lung abnormalities resembling atelectasis, and dwarfism characterized by aberrant cartilage morphology. The phenotyp…

Although IGF-I has been identified as an important growth factor for the skeleton, the role of IGF-I on embryonic bone development remains unknown. Here we show that, in IGF-I-deficient (IGF-I(-/-)) mice, skeletal malformations, including short-limbed dwarfism, were evident at days post coitus (dpc) 14.5 to 18.5, accompanied by delays of mineralization in the spinal column, sternum, and fore paws. Reduced chondrocyte proliferation and increased chondrocyte apoptosis were identified in both the spinal ossification center and the growth plate of long bones. Abnormal chondrocyte differentiation a…

BACKGROUND: There are no previous reports about de novo germ cell tumors without any past history of germ cell tumor. We describe a case of de novo cerebral germinoma in association with Klinefelter's syndrome.
CASE DESCRIPTION: A boy had undergone growth hormone therapy for dwarfism because of hypopituitarism from 10 to 17 years old. The result of karyotyping at the age of 13 was 47,XXY. Magnetic resonance images (MRI) of the brain at the age of 17 years showed no lesions. Two years later, at the age of 19, the patient noticed onset of mild right hemiparesis. MR imaging revealed the existenc…

Fibroblast growth factor (FGF) signaling is involved in skeletal development of the vertebrate. Gain-of-function mutations of FGF receptors (FGFR) cause craniosynostosis, premature fusion of the skull, and dwarfism syndromes. Disruption of Fgfr3 results in prolonged growth of long bones and vertebrae. However, the role that FGFs actually play in skeletal development in the embryo remains unclear. Here we show that Fgf18 is expressed in and required for osteogenesis and chondrogenesis in the mouse embryo. Fgf18 is expressed in both osteogenic mesenchymal cells and differentiating osteoblasts du…

In man, the genetic defects of more than 600 inherited diseases, of which at least 150 skeletal diseases, have been identified as is the chr … mented coronoid process, ununited anconeal process, osteochondrosis dissecans and incongruency) and dwarfism. Where possible a link is made with similar diseases in man or mouse. Then, the molecular biological tools available to analyse the genetic defect will be reviewed and some examples discussed.

Achondroplasia, the most common form of dwarfism in man, is a dominant genetic disorder caused by a point mutation (G380R) in the transmembrane region of fibroblast growth factor receptor 3 (FGFR3). We used gene targeting to introduce the human achondroplasia mutation into the murine FGFR3 gene. Heterozygotes for this point mutation that carried the neo cassette were normal whereas neo+ homozygotes had a phenotype similar to FGFR3-deficient mice, exhibiting bone overgrowth. This was because of interference with mRNA processing in the presence of the neo cassette. Removal of the neo selection m…

Achondroplasia and pseudoachondroplasia are autosomal dominant skeletal dysplasias resulting in short-limbed dwarfism. Histologic and ultrastructural studies of the cartilage in pseudoachondroplasia and in homozygous achondroplasia have suggested a structural abnormality in chondroitin sulfate proteoglycan (CSPG), a major structural protein in the extra-cellular matrix. The gene encoding CSPG core protein (CSPGCP) is thus a logical "candidate gene" for analysis in these conditions. cDNA probes encoding CSPGCP were used to identify restriction fragment length polymorphisms (RFLPs) in DNA from a…

We describe a new type of bone dysplasia, the "acromicric dysplasia," based on the study of six patients. This dysplasia is characterized cl … emoral heads in some patients. No signs of visceral storage were found, which rules out geleophysic dwarfism. The histological, histochemical, and electron microscopical examination of the growth cartilage in two cases showed similar lesions: disorganization of the growth zone with islands of cells, some of them degenerated; abnormal organization of collag …

The recently discovered releasing factors for ACTH and growth hormone, CRF and GRF, have stimulated our understanding of the hypothalamo-pit … ng the cause of anterior pituitary failure and GRF may be of benefit for the treatment of pituitary dwarfism. However, it has to be stressed that the results of the new CRF and GRF tests can only be interpreted together with results of other established pituitary function tests and the neuroradiological evaluation of the hypothalamo-pituitary area.

The Relationship between ouabain-sensitive ATPase (Na-K ATPase) activity in erythrocytes and the thyroid status was studied in 36 patients w … n primary hypothyroidism, the factor appeared to be the endogenous T4 level, while in patients with dwarfism, the secretory capacity of TSH or TSH-releasing hormone (TRH) was contributory. Thus Na-K ATPase activity in red cells remains within the normal range after L-T4 replacement in the presence of a severe degree of primary hypothyroidism or in associat …

Background: The exocyst complex, a conserved hetero-octameric complex including the EXO70 subunit, is a pivotal regulator of various cellular and developmental processes in plants. Among the diverse EXO70 isoforms, the specific function of EXO70E2-a primary organizer of exocyst-positive organelles (EXPOs)-remains to be fully elucidated. Methods: Here, we investigated the functional impact of constitutive EXO70E2 overexpression in Arabidopsis thaliana. Results: We observe that EXO70E2 overexpression leads to severe etiolation and dwarfism, accompanied by dose-dependent inhibition of primary roo…

Foxtail millet (Setaria italica) has lagged in yield compared with major cereals, largely due to suboptimal architecture. Here, we report a telomere-to-telomere (T2T) genome assembly of the dwarf, multi-tillering cultivar Dungu and identify SiSD1 (Semi Dwarf 1) via map-based cloning. This gene encodes GA20ox2, the rice SD1 ortholog involved in gibberellin biosynthesis. The loss-of-function sisd1 reduces bioactive gibberellins and remodels nitrogen metabolism, conferring semi-dwarfism, increased tillering, and higher yield. Critically, the semi-dominantly heterozygous SiSD1/sisd1 in F1 hybrids …

Dwarfing cultivation is a transformative strategy in modern fruit tree management, yet the molecular underpinnings of interstock-induced dwarfism remain elusive in persimmon (Diospyros kaki Thunb.). Here, we unveil DkMYB117, a novel R2R3-MYB transcription factor, as a key molecular switch orchestrating scion dwarfing via gibberellin (GA) catabolism. Using the dwarfing interstock "Nantongxiaofangshi," we demonstrate that DkMYB117 expression is specifically suppressed in grafted scions, leading to reduced active GA levels and stunted growth. Mechanistically, DkMYB117 directly binds to the MBSII …

Background: In the literature, the occurrence of menarche, the index of female sexual maturation, is related to chronological age (CA), but in conditions of abnormal growth it was proposed that determination of bone age (BA) was a better index of pubertal maturation than CA. Aim: To compare CA with BA at menarche in girls with Laron Syndrome (LS), a model of genetic IGF-I deficiency, which causes severe dwarfism and delayed puberty. Subjects: Data were retrieved retrospectively from our medical records. Nine untreated and four IGF-I-treated girls with LS were included in the study. Eleven GH-t…

Nucleotide-binding leucine-rich repeat (NLR) proteins assemble into genetically linked pairs to mediate effector-triggered immunity (ETI) in plants. Here, we characterize the paired NLRs NRCX and NARY (NRCX adjacent resistance gene Y) in Nicotiana benthamiana. CRISPR/Cas9 knockout of NRCX caused severe dwarfism and constitutively activated immunity, marked by PR1 upregulation and enhanced resistance to Phytophthora capsici. Co-silencing or double knockout of the adjacent NLR NARY partially rescued the nrcx phenotype, revealing NARY as a compensatory regulator that modulates growth and immunity…

In crop plants, there is often a trade-off between growth and defense; modulating the balance is crucial for effective crop improvement. In rice (Oryza sativa), the DENSE AND ERECT PANICLE 2 (DEP2) mutant shows altered phenotypes such as a more vertical leaf angle, erect panicles, small and round grains, slightly lower thousand-grain weight, and dwarfism. Here, we determined that loss of DEP2 function enhances resistance to the rice blast fungus Magnaporthe oryzae. We established that DEP2 interacts with the E3 ubiquitin ligase OsRING80, which ubiquitinates DEP2 and promotes its degradation vi…

Fibroblast growth factor receptor 3 (FGFR3) is an important regulator of bone development and healing. Gain-of-function mutations in FGFR3 cause achondroplasia, the most common skeletal dysplasia characterized by short-limbed dwarfism. Our previous study showed that FGFR3 signaling accelerated bone repair through endochondral ossification during distraction osteogenesis. This study aimed to investigate the role of FGFR3 in intramembranous bone repair using a calvarial fracture model in gain-of-function transgenic Fgfr3 mice. Using micro-computed tomography, histomorphometric analysis, and real…
