Long-Term Blockade of Cocaine Self-Administration and Locomotor Activation in Rats by an Adenoviral Vector-Delivered Cocaine Hydrolase.

A promising approach in treating cocaine abuse is to metabolize cocaine in the blood using a mutated butyrylcholinesterase (BChE) that functions as a cocaine hydrolase (CocH). In rats, a helper-dependent adenoviral (hdAD) vector-mediated delivery of CocH abolished ongoing cocaine use and cocaine-primed reinstatement of drug-seeking for several months. This enzyme also metabolizes ghrelin, an effect that may be beneficial in maintaining healthy weights. The effect of a single hdAD-CocH vector injection was examined in rats on measures of anxiety, body weight, cocaine self-administration, and co

Effect of early life housing manipulation on baseline and drug-induced behavioural responses on neurochemistry in the male rat.

Employing environmental enrichment (EE) provides continual sources of dynamic interaction for animals. Though an established discipline in behavioural science, the consequences of EE on behavioural pharmacological tests have not been extensively examined. The purpose of this study was to examine the consequences of EE (or isolation housing) on a range of behavioural pharmacological tests and brain monoamine and brain-derived neurotrophic factor (BDNF) expression in the rat. Male rats were randomly assigned to IC (isolation), SC (standard group-housed) or EE conditions. IC and SC animals were h

Maternal separation with early weaning: a novel mouse model of early life neglect.

BACKGROUND: Childhood adversity is associated with increased risk for mood, anxiety, impulse control, and substance disorders. Although genetic and environmental factors contribute to the development of such disorders, the neurobiological mechanisms involved are poorly understood. A reliable mouse model of early life adversity leading to lasting behavioral changes would facilitate progress in elucidating the molecular mechanisms underlying these adverse effects. Maternal separation is a commonly used model of early life neglect, but has led to inconsistent results in the mouse. RESULTS: In an

Situational factors, conditions and individual variables which can determine ultrasonic vocalizations in male adult Wistar rats.

The fact that rats emit different types of ultrasonic vocalizations in a variety of contexts has received increasing experimental attention, since such calls might serve as indices of the animal's subjective state, and/or as social signals in various types of interactions with other rats. Here, we present two experiments in adult male Wistar rats where we tested several different situations and conditions with respect to the occurrence of high-frequency (50-kHz) and low-frequency (22-kHz) calls. These experiments showed that rats emitted high-frequency calls when tested singly in a housing cag

Early life stress caused glucose homeostasis impairment and anxiety-like behavior along with oxidative stress in adolescent male rat offspring: Protective effect of saffron extract.

Early life stress (ELS) significantly contributes to metabolic and psychological disorders by triggering oxidative stress and disrupting the hypothalamic-pituitary-adrenal (HPA) axis. This research examined ELS effects on redox status, along with anxiety-like behavior, in adolescent male Wistar rats, also assessing saffron (Crocus sativus L.) extract's potential benefits. Rats were exposed to foot shock stress from postnatal days 15-19, with treatment groups receiving saline or saffron extract (10 mg/kg) via intraperitoneal injection. Control groups were maintained without stress exposure. Ele

SV2A Attenuates Interictal anxiety in Pharmacoresistant Epilepsy via the SYT3-GluA2 Axis.

OBJECTIVE: Synaptic vesicle glycoprotein 2A (SV2A) is an established target of antiseizure therapy; however, its role in excitatory synaptic function and downstream postsynaptic machinery remains poorly defined. This study examined whether SV2A limits excitotoxic signaling by recruiting synaptotagmin-3 (SYT3), thereby modulating the α-amino-3-hydroxy-5-methyl-4-isoxazolepropionic acid receptor subunit GluA2. METHODS: A multilevel strategy was used to define the contribution of SV2A to anxiety in a pharmacoresistant epilepsy (PRE) model. The phenobarbital/phenytoin-resistant PRE phenotype was

Maternal anti-collapsin response mediator protein 1 antibody inducing autism-like behaviors in offspring.

Maternal autoantibodies targeting neural proteins represent a major environmental contributor to autism spectrum disorder (ASD). Although anti-collapsin response mediator protein 1 (CRMP1) antibodies (Abs) have been detected in mothers of ASD children, the pathogenic role of anti-CRMP1 Ab in ASD remains unclear. Here, we evaluated the ability of maternal anti-CRMP1 Ab to induce pathology in a mouse model. Using a cell-based assay, we found that maternal anti-CRMP1 Ab persisted throughout gestation and was detected in offspring serum on postnatal day 16. Exposed offspring exhibited delayed deve

Beyond marbles buried: A multidimensional analysis of the marble burying test in the C57BL/6J mouse strain.

Restricted and repetitive behaviors (RRBs), defined as responses with high frequency and for unusually long duration, are distinctive features of several psychiatric conditions, including autism spectrum disorder and obsessive-compulsive disorder. A common paradigm to assess RRBs in murine models is the marble burying test (MBT), whose main outcome is the total number of marbles buried under bedding substrate. Despite its widespread application for evaluating RRBs, the behavioral significance of marble burying remains highly debated in the literature. This study aimed to: i) systematically rec

Tetrabromobisphenol A exerts thyroid disrupting effects but has little overt impact on postnatal brain development and neurobehaviors in mice.

Tetrabromobisphenol A (TBBPA) is a widely used brominated flame retardant. There is evidence showing that TBBPA can exert thyroid disrupting effects in mammals, but different results were also reported, along with inconsistent reports regarding its neurotoxicity. Here, we investigated thyroid disrupting effects and neurotoxicity of TBBPA (5, 50, 500 µg/(kg·day)) to male mice following maternal and direct exposure through drinking water, with the anti-thyroid drug propylthiouracil (PTU) as the positive control. On postnatal day (PND) 15, we expectedly observed severe thyroid compensatory hyperp

Screening for Neuroprotective and Rapid Antidepressant-like Effects of 20 Essential Oils.

Depression is a serious psychiatric disorder with high prevalence, and the delayed onset of antidepressant effects remains a limitation in the treatment of depression. This study aimed to screen essential oils that have the potential for rapid-acting antidepressant development. PC12 and BV2 cells were used to identify essential oils with neuroprotective effects at doses of 0.1 and 1 µg/mL. The resulting candidates were treated intranasally (25 mg/kg) to ICR mice, followed by a tail suspension test (TST) and an elevated plus maze (EPM) after 30 min. In each effective essential oil, five main co

Stress resilience-associated behaviors following predator scent stress are accompanied by upregulated nucleus accumbens mGlu5 transcription in female Sprague Dawley rats.

Despite the higher prevalence of post-traumatic stress disorder (PTSD) in women, the majority of preclinical research has been conducted utilizing male subjects. We have found that male rats exposed to the predator scent 2,4,5-trimethyl-3-thiazoline (TMT) show heterogenous long-term anxiety-like behavior and conditioned fear to the TMT environment. Stress-Resilient males exhibit increased mGlu5 mRNA expression in the basolateral amygdala (BLA) and prefrontal cortex (PFC). Here we sought to determine whether the same behavioral and genetic responses would be observed in female rats exposed to T

Effects of chronic fluoxetine treatment on anxiety- and depressive-like behaviors in adolescent rodents - systematic review and meta-analysis.

BACKGROUND: Drugs prescribed for psychiatric disorders in adolescence should be studied very extensively since they can affect developing and thus highly plastic brain differently than they affect the adult brain. Therefore, we aimed to summarize animal studies reporting the behavioral consequences of chronic exposure to the most widely prescribed antidepressant drug among adolescents i.e., fluoxetine. METHODS: Electronic databases (Medline via Pubmed, Web of Science Core Collection, ScienceDirect) were systematically searched until April 12, 2022, for published, peer-reviewed, controlled tri

Interaction of lithium and sleep deprivation on memory performance and anxiety-like behavior in male Wistar rats.

Sleep plays a crucial role in the modulation of physiological and cognitive functions. Many studies have reported the impairment effect of sleep deprivation (SD) on cognitive functions such as learning and memory. On the other hand, lithium as one of the oldest drugs used for the treatment of psychiatric disorders, affects cognitive functions and mood state. In this study, we aimed to assess the effect of lithium, SD (for 24 h), and the interaction effect of SD and lithium, on memory function and anxiety-like behavior. The water box, the shuttle box, elevated plus maze, and the three-chamber p

Sex differences in behavioral traits related with high sensitivity to the reinforcing effects of cocaine.

Cocaine is the most prevalent illegal stimulant drug in Europe among the adult population. Its abuse is characterized by a faster substance abuse disorder (SUD) development than other drugs, with high vulnerability to relapse. However, there does not exist an effective treatment for cocaine dependence. Sex differences have been reported in psychological disorders including SUD. For this reason, it is essential to identify risk factors that predict susceptibility or resilience to cocaine addiction for the development of effective prevention strategies considering sex differences. In the present
