Neuroimaging and neurogenetics of epilepsy in humans.

In the past decade, several genetic mutations have been associated with different forms of familial focal and generalized epilepsies. Most of these genes encode ion-channel subunits. Based on neurophysiological in vitro and in vivo animal studies, substantial progress has been made in understanding the functional consequences of gene defects associated with epilepsies. However, the knowledge transition from animal studies to patients carrying a mutation, or even suffering from a nonfamilial form of epilepsy, is very limited. This review will illustrate how neuroimaging studies in humans may he

Conditional-associative learning in eating disorders: a comparison with OCD.

The acquisition of conditional associations using neutral and individually threatening verbal stimuli was assessed in 16 females with anorex … icit was not evident in bulimics or in normal controls. These findings support the assumptions from functional neuroimaging investigations in AN and OCD and provide evidence that obsessive and compulsive behavior could have its origin within common neurobiological dysfunctions. The CAL possibly serves as a functional correlate of a neurophysiological dysfunction in obses …

Attenuated left prefrontal activation during a verbal fluency task in patients with depression.

Functional neuroimaging studies on patients with depression have found abnormal activity in the left prefrontal and anterior cingulate cortex compared with healthy controls. Other studies have shown that these regions become active in healthy subjects during verbal fluency tasks, while patients with depression show impaired performance on such tasks. We used functional magnetic resonance imaging to investigate changes in cerebral blood oxygenation associated with a verbal fluency task in depressed patients and healthy volunteers. In contrast to 10 age- and sex-matched healthy control subjects …

How gut microbiota contribute to neuropsychiatric disorders: evidence from neuroimaging studies.

The interaction between the gut microbiota and central nervous system (CNS) diseases has emerged as a major focus in neuroscience and microbiome research. Accumulating evidence shows that gut microbiota influence the pathogenesis of neurodevelopmental, neurodegenerative, autoimmune, and psychiatric conditions via the microbiota-gut-brain axis. However, the underlying mechanisms are complex and not yet fully elucidated. Advances in multimodal magnetic resonance imaging, positron emission tomography, and diffusion tensor imaging, now enable in vivo visualization of associations between gut micro

Functional near-infrared spectroscopy for the assessment and treatment of patients with disorders of consciousness.

BACKGROUND: Advances in neuroimaging have significantly enhanced our understanding of brain function, providing critical insights into the diagnosis and management of disorders of consciousness (DoC). Functional near-infrared spectroscopy (fNIRS), with its real-time, portable, and noninvasive imaging capabilities, has emerged as a promising tool for evaluating functional brain activity and nonrecovery potential in DoC patients. This review explores the current applications of fNIRS in DoC research, identifies its limitations, and proposes future directions to optimize its clinical utility. AI

Clinical-functional brain connectivity signature predicts longitudinal symptom improvement after acupuncture treatment in patients with functional dyspepsia.

Whilst acupuncture has been shown to be an effective treatment for functional dyspepsia (FD), its efficacy varies significantly among patients. Knowing beforehand how each patient responds to acupuncture treatment will facilitate the ability to produce personalized prescriptions, therefore, improving acupuncture efficacy. The objective of this study was to construct the prediction model, based on the clinical-neuroimaging signature, to forecast the individual symptom improvement of FD patients following a 4-week acupuncture treatment and to identify the critical predictive features that could

Transcriptomic dissociation between synaptic plasticity suppression and structure-related maintenance signaling in the Alzheimer's frontal cortex: A molecular analysis.

BACKGROUND: Early Alzheimer's disease (AD) frequently presents with frontal lobe-related behavioral abnormalities, such as executive dysfunction and confabulation, despite minimal frontal cortical atrophy on neuroimaging. The molecular basis of this dissociation between behavioral dysfunction and preserved structure remains poorly understood. METHODS: To address this paradox, we performed a transcriptomic re-analysis of postmortem frontal cortex samples from individuals with AD, vascular dementia (VaD), and cognitively normal controls (GSE122063). Gene set enrichment analysis was applied to i

Intranasal (R)-ketamine modulates depression symptom and neurotransmitters-associated human brain connectivity.

Racemic (R,S)-ketamine exerts rapid antidepressant effects, and growing evidence suggests its R-isomer may offer sustained efficacy with fewer side effects. However, the neurobiological mechanisms underlying (R)-ketamine's action in the human brain are largely unknown. To address this, we acquired resting-state fMRI data from 32 healthy volunteers 24 h before and after intranasal administration of (R)-ketamine (n = 24) or placebo (n = 8). We primarily assessed changes in long-range functional synchrony using degree centrality (DC) and elucidated the sources of these changes with functiona

Sleep deprivation disrupts postural balance and sensorimotor integration: A combined psychophysiological-behavioral analysis.

BACKGROUND: Sleep is crucial for optimal sensorimotor integration, a fundamental process enabling coordinated motor responses to sensory inputs. However, the neurophysiological mechanisms through which acute sleep deprivation impairs this integration remain incompletely understood. This study investigates the impact of acute sleep deprivation on postural balance and elucidates the underlying multilayered mechanisms using a combination of behavioral, psychophysiological, and neuroimaging indicators. METHODS: Twenty-five healthy young participants underwent 36 h of total sleep deprivation. Befo

Default mode network-basal ganglia network connectivity predicts the transition to postherpetic neuralgia.

BACKGROUND: Neuroimaging studies have revealed aberrant network functional connectivity in postherpetic neuralgia (PHN) patients. However, there is a lack of knowledge regarding the relationship between the brain network connectivity during the acute period and disease prognosis. OBJECTIVE: The purpose of this study was to detect characteristic network connectivity in the process of herpes zoster (HZ) pain chronification and to identify whether abnormal network connectivity in the acute period can predict the outcome of patients with HZ. METHODS: In this cross-sectional study, 31 patients wi

Advances in fetal and neonatal neuroimaging and everyday exposures.

The complex, tightly regulated process of prenatal brain development may be adversely affected by "everyday exposures" such as stress and environmental pollutants. Researchers are only just beginning to understand the neural sequelae of such exposures, with advances in fetal and neonatal neuroimaging elucidating structural, microstructural, and functional correlates in the developing brain. This narrative review discusses the wide-ranging literature investigating the influence of parental stress on fetal and neonatal brain development as well as emerging literature assessing the impact of expo

Clinical MRI morphological analysis of functional seizures compared to seizure-naïve and psychiatric controls.

PURPOSE: Functional seizures (FS), also known as psychogenic nonepileptic seizures (PNES), are physical manifestations of acute or chronic psychological distress. Functional and structural neuroimaging have identified objective signs of this disorder. We evaluated whether magnetic resonance imaging (MRI) morphometry differed between patients with FS and clinically relevant comparison populations. METHODS: Quality-screened clinical-grade MRIs were acquired from 666 patients from 2006 to 2020. Morphometric features were quantified with FreeSurfer v6. Mixed-effects linear regression compared the

Subtle in-scanner motion biases automated measurement of brain anatomy from in vivo MRI.

While the potential for small amounts of motion in functional magnetic resonance imaging (fMRI) scans to bias the results of functional neuroimaging studies is well appreciated, the impact of in-scanner motion on morphological analysis of structural MRI is relatively under-studied. Even among "good quality" structural scans, there may be systematic effects of motion on measures of brain morphometry. In the present study, the subjects' tendency to move during fMRI scans, acquired in the same scanning sessions as their structural scans, yielded a reliable, continuous estimate of in-scanner motio…

Human Ecstasy use is associated with increased cortical excitability: an fMRI study.

The serotonergic neurotoxin, 3,4-methylenedioxymethamphetamine (MDMA/Ecstasy), is a highly popular recreational drug. Human recreational MDMA users have neurocognitive and neuropsychiatric impairments, and human neuroimaging data are consistent with animal reports of serotonin neurotoxicity. However, functional neuroimaging studies have not found consistent effects of MDMA on brain neurophysiology in human users. Several lines of evidence suggest that studying MDMA effects in visual system might reveal the general cortical and subcortical neurophysiological consequences of MDMA use. We used 3 …

Network-based alterations in task-induced functional connectivity in bipolar disorder: An functional near-infrared spectroscopy study.

BACKGROUND: Functional near-infrared spectroscopy (fNIRS) is increasingly used to examine cognitive abilities and support the diagnosis of bipolar disorder (BD). Despite its growing application, research on functional connectivity in BD remains limited, and task-induced functional connectivity remains under-investigated. This study aims to differentiate patients with BD from healthy controls (HCs) based on fNIRS-derived functional connectivity. METHODS: We analyzed fNIRS data from 50 patients with BD and 50 HCs during resting state and two cognitive tasks, including the verbal fluency test (V
