Phenotyping iPSC-derived neurons from Angelman patients and partial rescue via JNK pathway inactivation.

BACKGROUND: Angelman Syndrome (AS, MIM #105,830) is a rare neurodevelopmental disorder caused by imprinting defects, and it is marked by motor impairment, intellectual disability, speech limitations, and seizures. The condition arises due to the loss of function in neurons of the maternally-inherited UBE3A gene, mapping in the imprinted chromosomal region 15q11.2-q13, through a variety of genetic and epigenetic mechanisms. UBE3A encodes a ubiquitin E3 ligase, which is essential for synaptic plasticity. Most studies of AS have been performed on animal models, while induced pluripotent stem cell

Immunoregulatory role of quercetin in lung cancer.

Long-term nourishment with quercetin, a naturally occurring plant flavonoid abundant in citrus fruits, buckwheat, and onions, has been associated with reduced risks of chronic diseases, including cancer. In this study, we evaluated the therapeutic potential and the anti-cancer properties of quercetin in lung cancer using in vitro, in vivo, and ex vivo models. Quercetin induced tumor cell cycle arrest via the p53-p21 axis and suppressed tumor cell migration in both the A549 and H520 human lung cancer cell lines. Moreover, it reduced oxidative stress and downregulated the protein expression of o

Kinome-Wide RNA Interference Screening Identifies Mitogen-Activated Protein Kinases and Phosphatidylinositol Metabolism as Key Factors for Rabies Virus Infection.

Throughout the rabies virus (RABV) infectious cycle, host-virus interactions define its capacity to replicate, escape the immune response, and spread. As phosphorylation is a key regulatory mechanism involved in most cellular processes, kinases represent a target of choice to identify host factors required for viral replication. A kinase and phosphatase small interfering RNA (siRNA) high-content screening was performed on a fluorescent protein-recombinant field isolate (Tha RABV). We identified 57 high-confidence key host factors important for RABV replication with a readout set at 18 h postin

Differences in early acetaminophen hepatotoxicity between obese ob/ob and db/db mice.

Clinical investigations suggest that hepatotoxicity after acetaminophen (APAP) overdose could be more severe in the context of obesity and nonalcoholic fatty liver disease. The pre-existence of fat accumulation and CYP2E1 induction could be major mechanisms accounting for such hepatic susceptibility. To explore this issue, experiments were performed in obese diabetic ob/ob and db/db mice. Preliminary investigations performed in male and female wild-type, ob/ob, and db/db mice showed a selective increase in hepatic CYP2E1 activity in female db/db mice. However, liver triglycerides in these anim

Development of glutathione S-transferase-P-negative foci accompanying nuclear factor-erythroid 2-related factor 2 expression during early stage of rat hepatocarcinogenesis.

Glutathione S-transferase P (GST-P), a marker for rat hepatic preneoplastic lesions, is suggested to bind to Jun N-terminal kinase (JNK) to repress stress response, and GST-P gene expression is regulated by a transcription factor, nuclear factor-erythroid 2-related factor 2 (Nrf2). In this study, we examined by immunohistochemistry whether JNK2, p38 mitogen-activated protein kinase, and Nrf2 were expressed in GST-P-positive foci induced by the Solt-Farber protocol. At 2 weeks after partial hepatectomy, all GST-P-positive foci were negative for p38, and 86.4 +/- 5.6% and 64.7 +/- 6.3% of GST-P-
