TyG index is associated with colorectal adenomatous polyps and genetically linked to colon polyps: a cross-sectional and Mendelian randomization study.

BACKGROUND: The triglyceride-glucose (TyG) index, a simple and validated surrogate marker of insulin resistance, may be clinically relevant to colorectal polyp development because insulin resistance can promote colorectal neoplasia through hyperinsulinemia, chronic inflammation, oxidative stress, and dysregulated insulin-like growth factor signaling. However, the relationship between TyG index and colorectal polyps remains incompletely understood. This study aimed to investigate the association between TyG index and colorectal polyp risk using both observational and genetic approaches. METHOD

Genetically determined causality between immune cells and anxiety disorders: A Mendelian randomization study.

Prior investigations have endeavored to unveil the connection between immune cells and anxiety disorders. However, the majority are limited to a single or few indicators and have not comprehensively evaluated the causal relationship between immune cells and anxiety disorders. In this study, we performed a two-sample Mendelian randomization (MR) analysis to assess the relationship between 731 immune phenotypes and anxiety disorders, offering genetic evidence to substantiate the correlation between immune phenotypes and the risk of anxiety disorders. Using publicly available data from genome-wid

Causal determinants of gout in 614,000 adults: A two-sample Mendelian randomization study of dietary, lifestyle, and metabolic traits.

Gout affects over 55 million people worldwide, with prevalence projected to rise by 70% by 2050. Although observational studies have implicated several dietary, lifestyle, and metabolic risk factors, causal relationships remain uncertain because of confounding and reverse causation. Univariable and multivariable two-sample Mendelian randomization (MR) analyses were performed using genome-wide association data from 2 European cohorts: UK Biobank (6543 self-reported gout cases and 456,390 controls) and FinnGen (3576 cases and 147,221 controls). Genetic instruments for 12 exposures, including dri

Causal effects of ferroptosis-related traits on ovarian dysfunction: insights from integrating genome-wide Mendelian randomization, DNA methylation, gene expression, and proteome.

BACKGROUND: Ferroptosis has been implicated in the development of ovarian dysfunction; however, the underlying causal genetic mechanisms remain largely unclear. In this study, a multi-omics Mendelian randomization (MR) approach was applied to systematically investigate the causal effects of ferroptosis-related molecular traits on ovarian dysfunction. METHODS: Summary-level data for blood-based methylation quantitative trait loci (mQTLs), expression QTLs (eQTLs), and protein QTLs (pQTLs) were obtained from large-scale consortia. Genetic associations with ovarian dysfunction were sourced from th

Multi-omics analysis of perfluorooctanoic acid and glioblastoma: insights from Mendelian randomization, network toxicology, and molecular docking.

Glioblastoma (GBM) is the most lethal primary brain tumor with limited treatment options. Perfluorooctanoic acid (PFOA), a ubiquitous "forever chemical" classified as a group 1 carcinogen by the IARC, can cross the blood-brain barrier and accumulate in neural tissue. This study aimed to determine the causal relationship between genetically predicted higher circulating PFOA levels and GBM risk, elucidate underlying molecular mechanisms, and identify potential therapeutic targets using an integrated multi-omics framework. Two-sample Mendelian randomization (MR) analysis was conducted using 23 si

Genetic Evidence Suggesting Potential Associations Between Sleep-Related Phenotypes and Psychiatric Disorders: A Two-Sample Mendelian Randomization Study.

OBJECTIVE: Sleep-related phenotypes, including insomnia symptoms, sleep apnea, and habitual sleep duration, are increasingly recognized as potentially modifiable factors associated with psychiatric disorders. However, their potential causal roles remain uncertain because observational studies are susceptible to confounding and reverse causation. STUDY DESIGN: Two-sample Mendelian randomization (MR) was performed using self-reported insomnia symptoms, sleep apnea and habitual sleep duration, as exposures against 10 psychiatric outcomes: ADHD, PTSD, MDD, PPD, BIP, ASD, OCS, SCZ, AN, and PD. Copper-linked metabolic stress as a potential contributor to gastric epithelial transformation and metastasis.

Gastric cancer remains a major health burden, and the molecular events that accompany the transition from normal gastric epithelium to malignancy (and the factors that facilitate dissemination) are incompletely defined. In particular, the cellular phenotypes that precede overt transformation and the role of copper-linked metabolic stress in this process are not well understood. We integrated 2 public single-cell RNA-seq datasets of gastric tissues (gene expression omnibus) using canonical correlation analysis-based batch correction and performed quality control and pre/post-integration assessm

Associations between daytime napping, sleep duration, and depression and 15 cardiovascular diseases: a Mendelian randomization study.

BACKGROUND: Numerous studies have documented the effects of daytime napping, sleep duration, and depression on cardiovascular diseases (CVDs). However, the evidence has been gleaned from observational studies that might be riddled with confounding variables and the possibility of reverse causation bias. Therefore, the present study employed a Mendelian randomization (MR) methodology to meticulously explore the relationships between daytime napping, sleep duration, and depression, and the risk profiles of CVDs. METHODS: Genome-wide significant genetic variants associated with daytime napping,

Vitamin D and COVID-19 susceptibility and severity in the COVID-19 Host Genetics Initiative: A Mendelian randomization study.

BACKGROUND: Increased vitamin D levels, as reflected by 25-hydroxy vitamin D (25OHD) measurements, have been proposed to protect against COVID-19 based on in vitro, observational, and ecological studies. However, vitamin D levels are associated with many confounding variables, and thus associations described to date may not be causal. Vitamin D Mendelian randomization (MR) studies have provided results that are concordant with large-scale vitamin D randomized trials. Here, we used 2-sample MR to assess evidence supporting a causal effect of circulating 25OHD levels on COVID-19 susceptibility a

Genetic prediction of immune cells, inflammatory proteins, and metabolite-mediated association between gut microbiota and COPD: a Mendelian randomization study.

Observational studies have suggested a potential association between the gut microbiota and chronic obstructive pulmonary disease (COPD); however, the causal relationship between them, as well as the mediating roles of metabolites, inflammatory proteins, and immune cells, remain unclear. This study aims to elucidate the causal relationship between gut microbiota and COPD using genetic approaches and to explore the potential mediating roles of metabolites, inflammatory proteins, and immune cells. To investigate the causal association between gut microbiota and COPD, we first employed univariabl

Selenoprotein S and the Causal Risk of Hypertension in Pregnancy: A Mendelian Randomization Study.

Background: Pregnancy-induced hypertension (PIH) affects approximately 10% of pregnancies worldwide, representing a leading cause of maternal and perinatal morbidity and mortality. The relationship between plasma selenium levels and PIH remains controversial, with observational studies limited by confounding factors. Selenoprotein S (SELENOS) has emerged as a potential biomarker for PIH risk. As one of the carrier proteins for dietary selenium, SELENOS plays a crucial role in oxidative stress and inflammatory pathways. However, the causal relationship between the plasma levels of the SELENOS a

Association between bone mineral density and scoliosis: a two-sample mendelian randomization study in european populations.

BACKGROUND: Previous studies have shown that bone mineral density (BMD) has a certain impact on scoliosis. However, up to now, there is no clear evidence that there is a causal association between the two. The aim of this study is to investigate whether there is a causal association between BMD at different body positions and scoliosis by two-sample Mendelian randomization (MR). METHODS: Genetic variants (SNPS) strongly associated with BMD (total body BMD (TB-BMD), lumbar spine BMD (LS-BMD), femoral neck BMD (FN-BMD), heel BMD (HE-BMD), and forearm BMD (FA-BMD)) were extracted from GEFOS and

FHL5 Controls Vascular Disease-Associated Gene Programs in Smooth Muscle Cells.

BACKGROUND: Genome-wide association studies have identified hundreds of loci associated with common vascular diseases, such as coronary artery disease, myocardial infarction, and hypertension. However, the lack of mechanistic insights for many GWAS loci limits their translation into the clinic. Among these loci with unknown functions is UFL1-four-and-a-half LIM (LIN-11, Isl-1, MEC-3) domain 5 (FHL5; chr6q16.1), which reached genome-wide significance in a recent coronary artery disease/ myocardial infarction GWAS meta-analysis.
