PURPOSE: Recurrent refractory testicular germ cell tumors (rrTGCT) represent very rare disease with limited therapeutic options. Antibody-drug conjugate Sacituzumab govitecan (ADC-SG) targeting TROP2 protein (TACSTD2 gene) demonstrated anti-tumor activity in solid tumors. Our study aimed to investigate efficacy of ADC-SG in rrTGCT.
EXPERIMENTAL DESIGN: TROP2 expression in tumor samples (n = 71) and cell line-derived xenografts (CDXs) by immunohistochemistry (IHC); and TACSTD 2 expression by qRT-PCR was determined. Cytotoxicity was evaluated in adherent and spheroid conditions. We generated CD

Newer, more effective and non-cytotoxic therapies are an unmet need for patients with diffuse large B-cell lymphoma (DLBCL) and other B-cell malignancies. Recently approved agents include polatuzumab with bendamustine and rituximab, selinexor, and tafasitamab plus lenalidomide. Three CAR-T cell products are currently approved by the FDA, with others in clinical trials. Additional agents in development include bispecific antibodies and antibody drug conjugates. Combinations of targeted therapies should lead to further improvement in the outcome of patients with B-cell malignancies.

Sjögren's syndrome (SjS) is a frequent systemic autoimmune disease responsible for a major decrease in patients' quality of life, potentially leading to life-threatening conditions while facing an unmet therapeutic need. Hence, we assessed the immunogenicity, efficacy, and tolerance of IFN-Kinoid (IFN-K), an anti-IFNα vaccination strategy, in a well-known mouse model of systemic autoimmunity with SjS-like features: MRL/MpJ-Faslpr/lpr (MRL/lpr) mice. Two cohorts (with ISA51 or SWE01 as adjuvants) of 26 female MRL/lpr were divided in parallel groups, "controls" (not treated, PBS and Keyhole Limp

PURPOSE: New therapies have changed the outlook for patients with multiple myeloma, but novel agents are needed for patients who are refractory or relapsed on currently approved drug classes. Novel targets other than CD38 and BCMA are needed for new immunotherapy development, as resistance to daratumumab and emerging anti-BCMA approaches appears inevitable. One potential target of interest in myeloma is ICAM1. Naked anti-ICAM1 antibodies were active in preclinical models of myeloma and safe in patients, but showed limited clinical efficacy. Here, we sought to achieve improved targeting of mult

Purpose Genetic alterations causing overexpression of programmed death-1 ligands are near universal in classic Hodgkin lymphoma (cHL). Nivolumab, a programmed death-1 checkpoint inhibitor, demonstrated efficacy in relapsed/refractory cHL after autologous hematopoietic cell transplantation (auto-HCT) in initial analyses of one of three cohorts from the CheckMate 205 study of nivolumab for cHL. Here, we assess safety and efficacy after extended follow-up of all three cohorts. Methods This multicenter, single-arm, phase II study enrolled patients with relapsed/refractory cHL after auto-HCT treatm

CD30 is a 120-kDa type I transmembrane glycoprotein belonging to the tumor necrosis factor receptor superfamily. Overexpression of CD30 has been reported in Hodgkin's lymphoma (HL) and anaplastic large-cell lymphoma (ALCL). CD30-targeted treatment with antibody-drug conjugates (ADCs) can lead to promising clinical benefit. Lidamycin (LDM), consisting of an apoprotein LDP and an active enediyne chromophore AE, is a member of the enediyne antibiotic family and one of the most potent antitumor agents. AE and LDP can be dissociated and reconstituted under certain conditions in vitro. LDM is an ide

Despite considerable efforts to develop efficient carriers, the major target organ of short-interfering RNAs (siRNAs) remains limited to the liver. Expanding the application outside the liver is required to increase the value of siRNAs. Here we report on a novel platform targeted to muscular organs by conjugation of siRNAs with anti-CD71 Fab' fragment. This conjugate showed durable gene-silencing in the heart and skeletal muscle for one month after intravenous administration in normal mice. In particular, 1μg siRNA conjugate showed significant gene-silencing in the gastrocnemius when injected

INTRODUCTION: Anti-HER1 monoclonal antibody (mAb), panitumumab (Vectibix) is a fully human mAb approved by the FDA for the treatment of epidermal growth factor receptor (EGFR, HER1)-expressing colorectal cancers. By combining the targeted specificity of panitumumab with the quantitative in vivo imaging capabilities of PET, we evaluated the potential of (89)Zr-DFO-panitumumab PET/CT imaging and performed non-invasive, in vivo imaging of HER1 expression and estimated human dosimetry.
METHODS: Panitumumab was radiolabeled with (89)Zr using a derivative of desferrioxamine (DFO-Bz-NCS) and with (1

OBJECTIVE: Over the past decade, the availability of biological agents for the treatment of juvenile idiopathic arthritis (JIA) has increased substantially. Because direct head-to-head trials comparing these agents are lacking, we indirectly compared their efficacy.
METHODS: In a systematic review, all available efficacy data from randomised controlled trials performed in JIA with inclusion of biological agents were retrieved. Indirect between-drug comparisons (based on Bucher's method) were conducted only if trials were comparable with regard to design and patients' characteristics related t

Goodpasture's disease, or anti-glomerular basement membrane (anti-GBM) disease, is a systemic autoimmune disorder defined by anti-GBM antibody-mediated damage (mainly immunoglobulin G-1) resulting in progressive crescentic glomerulonephritis and, frequently, diffuse pulmonary alveolar hemorrhage. It may be regarded as a "conformeropathy" where the quaternary structure of the α345NC1 hexamer that constitutes GBM undergoes a conformational change, exposing pathogenic epitopes on the α3 and α5 chains, eliciting a pathogenic autoantibody anti-GBM response. Goodpasture's disease accounts for 20% of

OBJECTIVE: To analyze the association of Wegener's granulomatosis (WG) with 2 single nucleotide polymorphisms (SNP), a +49 A/G polymorphism in coding sequence (CDS) 1 and a C/T base exchange in the promoter region at position -318.
METHODS: Restriction enzyme digestion of PCR amplified genomic DNA was used to analyze the CTLA-4 SNP in 32 patients with WG and 100-122 ethnically matched healthy controls.
RESULTS: Patients were more often heterozygous for C/T in the promoter region (31% of the patients vs 14% of controls; p < 0.05). Homozygosity for C was less frequent in patients (69% of patie

BACKGROUND: A genetic susceptibility to coeliac disease is well established, involving HLA and non-HLA components. CTLA4 is an important regulator of T-cell function and some studies have suggested that sequence variation in the gene might be a determinant of disease susceptibility, although the evidence is conflicting.
METHODS: Sixty-two children with biopsy-proven coeliac disease attending a single centre in Sweden were studied. All were genotyped for presence of the HLA-DQA1*0501, B 1*0201 alleles. Those who carried the HLA-DQ heterodimer (58/62) were genotyped for the +49 (A/G) exon I pol

This study introduces a novel drug-disease modeling framework designed to assess the benefit-risk balance of antibody-drug conjugates (ADC) in oncology. The framework integrates dose levels, pharmacokinetics, tumor growth dynamics, progression-free survival (PFS), and dose-adjusted adverse events. We demonstrated this through its application to tusamitamab ravtansine (Tusa), an ADC targeting Carcinoembryonic Antigen-Related Cell Adhesion Molecule 5 in non-squamous non-small cell lung cancer (nsq NSCLC). We developed our model using phase I trial safety data from 254 patients (doses: 5-190 mg/m

As an anti-HER2 antibody-drug conjugate, SHR-A1811 has exhibited significant antitumor activity in patients with HER2-low advanced breast cancer. This trial aims to explore the efficacy and safety profile of SHR-A1811 as a neoadjuvant therapy for hormone receptor-positive, HER2-low (HR + /HER2-low) breast cancer. We conducted a single-arm, phase 2 study enrolling women aged 18 to 70 years with untreated HR + /HER2-low invasive breast cancer (cT2-3, cN0-3) and a Ki-67 proliferation index exceeding 14%. Eligible patients had an Eastern Cooperative Oncology Group performance status of 0 or 1, and

Pancreatic cancer remains a lethal malignancy with limited treatment options. Trophoblast cell surface antigen 2 (TROP2), which is frequently overexpressed in pancreatic tumors and closely associated with poor prognosis, represents a promising therapeutic target. hIMB1636-LDM is a novel antibody-drug conjugate (ADC) developed by our laboratory, comprising the anti-TROP2 antibody hIMB1636 covalently linked to cytotoxic payload lidamycin (LDM) via an uncleavable linker. In this study, we systematically evaluated the antitumor efficacy and underlying mechanism of action of hIMB1636-LDM in pancrea

Bone metastasis is a major cause of morbidity in patients with advanced cancers and remains largely incurable. Trophoblast cell surface antigen 2 (TROP2), which is highly expressed in various malignancies, represents a promising therapeutic target. Sacituzumab govitecan (SG), an antibody-drug conjugate (ADC) targeting TROP2 and carrying SN-38, has demonstrated clinical efficacy in treating several types of solid tumors. However, its effects on bone metastases remain unclear. In this study, we investigated the inhibitory effects of SG on bone metastases using preclinical models of breast and pr

Acute monocytic leukemia (AML-M5) is a rapidly progressing hematological malignancy characterized by limited therapeutic options and poor survival outcomes. Conventional therapies, such as intensive chemotherapy offer limited efficacy due to their lack of tumor specificity, significant systemic toxicities, and high risk of disease relapse after initial treatment, underscoring the need for more targeted therapeutic approaches. CD64 (FcγRI) is highly expressed on monocyte-derived myeloid leukemia cells and has been identified as a promising therapeutic target for the treatment of AML-M5. We have
