Structure-activity relationship of adipate-bridged β-sitosterol glycosyl derivatives as novel cholesterol alternatives for liposome stabilization.

Liposomes stabilized by adipate-bridged β-sitosterol glycosyl derivatives (hereafter, β-sitosterol glycosyl adipates) were characterized to elaborate their structure-activity relationship (SAR) as effective cholesterol alternatives. Compared with liposomes stabilized by cholesterol or β-sitosterol, those stabilized by β-sitosterol glycosyl adipates exhibited improved bilayer structural properties. Among all formulations, liposomes stabilized by β-sitosterol sucrose adipate possessed the most compact lipid packing and the smallest particle size (<80 nm). After curcumin (Cur) loading, the optimi

80 years of extracellular vesicles: from discovery to clinical translation.

Extracellular vesicles (EVs) are heterogeneous, lipid bilayer-enclosed vesicles secreted by cells. Research on EVs dates back to the 1940s, and the term "exosomes" - a major subtype of EVs - was coined in 1981 to describe small membrane vesicles shed from cells. However, it is only in the past two decades that research in this area has expanded rapidly. By transferring functional biomolecules, EVs play a pivotal role in intercellular communication and regulate a wide range of cellular functions under both physiological and pathological conditions. Owing to their high biocompatibility, capacity

Bedaquiline inhibits the ATP synthase leak channel and prevents glutamate-induced neuronal death.

FOF1-ATP synthase is one of the most abundant proteins of the mitochondrial inner membrane and the primary enzyme responsible for ATP production in eukaryotic cells. Nevertheless, it was recently reported to play a prominent role in cell death by forming a large-conductance leak channel in the mitochondrial permeability transition pore (mPTP), making it a promising therapeutic target. Bedaquiline (BDQ), a member of the diarylquinoline class of drugs, was shown to selectively inhibit the catalytic activity of Mycobacterium tuberculosis ATP synthase with no effect on the mammalian enzyme. Here,

ATR-Infrared Spectroscopy and Acoustic Sensing in Characterization of Blood and Pancreatic Ductal Adenocarcinoma-Derived Extracellular Vesicles.

Extracellular vesicles (EVs) are phospholipid bilayer-enclosed nanoparticles secreted by most cells into the biofluids. They have an important role in intercellular communication by carrying bioactive components (e.g., lipids, proteins, nucleic acids, and other metabolites) and reflecting the biochemical and metabolic processes of their parent cell. Due to their widespread molecular cargo, they have potential diagnostic and therapeutic roles, so they can be considered as new generation biomarkers. With the drastic increase in the number of publications and the comparability of the obtained res

Effect of the S2' site cleavage on SARS-CoV-2 spike.

SARS-CoV-2 initiates infection of host cells by fusing its envelope lipid bilayer with the cell membrane. To overcome kinetic barriers for membrane fusion, the virus-encoded spike (S) protein refolds from a metastable prefusion state to a lower energy, stable postfusion conformation. The protein is first split into S1 and S2 fragments at a proteolytic site after synthesis, and presumably further cleaved at a second site, known as the S2' site, before membrane fusion can occur. Here, we report a cryo-EM structure of S2 fragment after the S2' cleavage, possibly representing a late fusion interme

Revolutionizing medicine: Harnessing plant-derived vesicles for therapy and drug transport.

The emergence of extracellular vesicles (EVs), which are natural lipid bilayer membrane structures facilitating intercellular substance and information exchange, has sparked innovative approaches in drug development and carrier enhancement. Plant-derived EVs notably offer advantages including low preparation cost, low immunogenicity, flexible drug delivery, high stability, good tissue permeability, and high inherent medicinal value compared to their animal-derived counterparts. Despite these promising attributes, the research on plant-derived EVs remains fragmented and lacks comprehensive synt

Spontaneous Transition between Multiple Conductance States and Rectifying Behaviors in an Artificial Single-Molecule Funnel.

It has long been an aspirational goal to create artificial channel structures that replicate the feat achieved by ion channel proteins. Biological ion channels occasionally demonstrate multiple conductance states (known as subconductance), remaining a challenging property to achieve in artificial channel molecules. We report a funnel-shaped single-molecule channel constructed by an electron-deficient macrocycle and two electron-deficient aromatic imide arms. Planar lipid bilayer measurements reveal distinct current recordings, including a closed state, two conducting states, and spontaneous tr

Recent advances of exosomal circRNAs in cancer and their potential clinical applications.

Circular RNA (circRNA) is a type of non-coding RNA that forms a covalently closed, uninterrupted loop. The expression of circRNA differs among cell types and tissues, and various circRNAs are aberrantly expressed in a variety of diseases, including cancer. Aberrantly expressed circRNAs contribute to disease progression by acting as microRNA sponges, functional protein sponges, or novel templates for protein translation. Recent studies have shown that circRNAs are enriched in exosomes. Exosomes are spherical bilayer vesicles released by cells into extracellular spaces that mediate intercellular

The Role of Extracellular Vesicles as Allies of HIV, HCV and SARS Viruses.

Extracellular vesicles (EVs) are lipid bilayer-enclosed entities containing proteins and nucleic acids that mediate intercellular communication, in both physiological and pathological conditions. EVs resemble enveloped viruses in both structural and functional aspects. In full analogy with viral biogenesis, some of these vesicles are generated inside cells and, once released into the extracellular milieu, are called "exosomes". Others bud from the plasma membrane and are generally referred to as "microvesicles". In this review, we will discuss the state of the art of the current studies on the

How non-enveloped viruses hijack host machineries to cause infection.

Viruses must navigate the complex endomembranous network of the host cell to cause infection. In the case of a non-enveloped virus that lacks a surrounding lipid bilayer, endocytic uptake from the plasma membrane is not sufficient to cause infection. Instead, the virus must travel within organelle membranes to reach a specific cellular destination that supports exposure or arrival of the virus to the cytosol. This is achieved by viral penetration across a host endomembrane, ultimately enabling entry of the virus into the nucleus to initiate infection. In this review, we discuss the entry mecha

A Ceramide-Regulated Element in the Late Endosomal Protein LAPTM4B Controls Amino Acid Transporter Interaction.

Membrane proteins are functionally regulated by the composition of the surrounding lipid bilayer. The late endosomal compartment is a central site for the generation of ceramide, a bioactive sphingolipid, which regulates responses to cell stress. The molecular interactions between ceramide and late endosomal transmembrane proteins are unknown. Here, we uncover in atomistic detail the ceramide interaction of Lysosome Associated Protein Transmembrane 4B (LAPTM4B), implicated in ceramide-dependent cell death and autophagy, and its functional relevance in lysosomal nutrient signaling. The ceramide

Regulation of actin assembly by PI(4,5)P2 and other inositol phospholipids: An update on possible mechanisms.

Actin cytoskeleton dynamics depend on a tight regulation of actin filament formation from an intracellular pool of monomers, followed by their linkage to each other or to cell membranes, followed by their depolymerization into a fresh pool of actin monomers. The ubiquitous requirement for continuous actin remodeling that is necessary for many cellular functions is orchestrated in large part by actin binding proteins whose affinity for actin is altered by inositol phospholipids, most prominently PI(4,5)P2 (phosphatidylinositol 4,5-bisphosphate). The kinetics of PI(4,5)P2 synthesis and hydrolysi

Existing and novel biomarkers for precision medicine in systemic sclerosis.

The discovery and validation of biomarkers resulting from technological advances in the analysis of genomic, transcriptomic, lipidomic and metabolomic pathways involved in the pathogenesis of complex human diseases have led to the development of personalized and rationally designed approaches for the clinical management of such disorders. Although some of these approaches have been applied to systemic sclerosis (SSc), an unmet need remains for validated, non-invasive biomarkers to aid in the diagnosis of SSc, as well as in the assessment of disease progression and response to therapeutic inter

Notch and Presenilin: regulated intramembrane proteolysis links development and degeneration.

Intensive studies of three proteins--Presenilin, Notch, and the amyloid precursor protein (APP)--have led to the recognition of a direct intersection between early development and late-life neurodegeneration. Notch signaling mediates many different intercellular communication events that are essential for determining the fates of neural and nonneural cells during development and in the adult. The Notch receptor acts in a core pathway as a membrane-bound transcription factor that is released to the nucleus by a two-step cleavage mechanism called regulated intramembrane proteolysis (RIP). The se

Evidence for a six-transmembrane domain structure of presenilin 1.

Mutations in genes encoding presenilin 1 and presenilin 2 account for the majority of cases of early-onset familial Alzheimer's disease. The presenilins have been localized to the endoplasmic reticulum and Golgi, but which of the multiple hydrophobic segments of the polypeptide chain span the lipid bilayer is unclear. To address this question, we have constructed a series of chimeric molecules in which a topologically neutral reporter protein (a C-terminal fragment of prolactin) containing three artificial glycosylation sites is fused to presenilin 1 following each of the 10 potential transmem

Translating umbilical cord blood extracellular vesicles into a novel therapy for postpartum alopecia: a mechanistic and preclinical review.

Postpartum alopecia (PPA) is a common physiological hair loss in women, characterized by complex pathogenesis involving abrupt postpartum hormonal withdrawal, disrupted hair follicle cycling, local microenvironmental inflammation, and oxidative stress. Umbilical cord blood-derived extracellular vesicles (UCB-EVs)-nanosized, lipid-bilayer-enclosed particles enriched with microRNAs, growth factors, and proteins-have shown considerable promise in regenerative medicine owing to their low immunogenicity, high biocompatibility, and tissue-repair capacity. Bacterial extracellular vesicles as emerging platforms to combat multidrug-resistant bacterial infections: Mechanisms, therapeutic applications, and future prospects.

Multidrug-resistant bacteria (MDRB) have become a global health crisis that challenges the effectiveness of conventional antibiotics. Bacterial extracellular vesicles (BEVs) are nanoscale bilayered membrane vesicles secreted by both Gram-positive and Gram-negative bacteria. They can encapsulate proteins, lipids, and nucleic acids, and transfer these molecules between bacteria and host cells without direct contact. Owing to their natural ability to transport bioactive molecules, BEVs have recently gained attention as potential anti-infective platforms. They can deliver antimicrobial agents dire

Quinone-transporting filaments expand bioenergetic capacity in Gram-positive Bacillota.

Cellular respiration depends on transferring electrons to hydrophobic quinones in membrane bilayers, meaning bioenergetic capacity is constrained by available membrane surface area. While Gram-negative bacteria expand this capacity through internal membrane invaginations and eukaryotes use membrane-bound organelles, whether Gram-positive bacteria have alternative capacity-generating mechanisms is unknown. Here we show that Bacillus subtilis forms a quinone-transporting pseudomembrane composed of filaments of the NADH dehydrogenase Ndh and the quinone-transporting protein Ncp. Cryo-EM, lipidomi

Controlled Rigidity Nanolipogel-Mediated Topical Delivery of Fucosterol for Treating Androgenic Alopecia through Follicle Targeting, Promoting Angiogenesis and Inhibiting Inflammation.

Androgenetic alopecia (AGA) is the most common type of hair loss. Its successful treatment depends on effective transdermal drug delivery strategies. In this study, we introduce a novel method utilizing a controlled rigidity nanolipogel (NLG) for the local delivery of fucosterol in the treatment of AGA. The NLG is formed by an identical lipid bilayer encapsulating an alginate core, with rigidity regulated by the degree of sodium alginate (SA) cross-linking. Young's moduli obtained by AFM were 2.91 ± 0.41, 61.5 ± 1.6, and 84.9 ± 1.1 MPa for the soft NLP, moderately rigid NLG-2.5, and most rigid

Roles of extracellular vesicles in the pathogenesis of chronic myeloid leukemia.

Extracellular vesicles (EVs) are lipid bilayer-enclosed particles released by cells into the extracellular space. They are produced by the majority of cell types under both physiological and pathological conditions. EVs mediate intercellular communication by delivering bioactive molecules - such as RNAs, adhesion molecules, signaling mediators, and antigen-presenting molecules - to recipient cells, thereby modulating their phenotype and function. A growing body of evidence highlights the involvement of EVs in the pathophysiology of leukemia, including their roles in remodeling the bone marrow
