Possible role for glutathione-S-transferase in the oligozoospermia elicited by acute zearalenone administration in Swiss albino mice.

Zearalenone (ZEA) is a non-steroidal estrogenic mycotoxin produced by several species of Fusarium, commonly found in the soil in temperate and warm countries and is a frequent contaminant of cereal crops worldwide. Accordingly, it has been implicated in several mycotoxicosis in farm animals and in humans, but the underlying mechanisms remain largely unknown. Therefore, the current study was aimed to investigate the effect of an acute dose of ZEA (40 mg/kg, p.o.) on reproductive and hematological parameters, as well as on markers of oxidative stress in liver, kidney and testes in mice. Adult Sw

Antifungal metabolites (monorden, monocillins I, II, III) from Colletotrichum graminicola, a systemic vascular pathogen of maize.

Colletotrichum graminicola is a systemic vascular pathogen that causes anthracnose stalk rot and leaf blight of maize. In the course of an effort to explore the potential presence and roles of C. graminicola metabolites in maize, ethyl acetate extracts of solid substrate fermentations of several C. graminicola isolates from Michigan and Illinois were found to be active against Aspergillus flavus and Fusarium verticillioides, both mycotoxin-producing seed-infecting fungal pathogens. Chemical investigations of the extract of one such isolate (NRRL 47511) led to the isolation of known metabolites

Ancient horizontal gene transfer from bacteria enhances biosynthetic capabilities of fungi.

BACKGROUND: Polyketides are natural products with a wide range of biological functions and pharmaceutical applications. Discovery and utilization of polyketides can be facilitated by understanding the evolutionary processes that gave rise to the biosynthetic machinery and the natural product potential of extant organisms. Gene duplication and subfunctionalization, as well as horizontal gene transfer are proposed mechanisms in the evolution of biosynthetic gene clusters. To explain the amount of homology in some polyketide synthases in unrelated organisms such as bacteria and fungi, interkingdo

Induction of calcium influx from extracellular fluid by beauvericin in human leukemia cells.

Beauvericin, a cyclic hexadepsipeptide, is a mycotoxin that can induce cell death in human lymphoblastic leukemia CCRF-CEM cells. Our previous data have shown that beauvericin induces cell death in CCRF-CEM cells in a dose- and time-dependent manner, and that this beauvericin-induced cell death can be prevented by administration of intracellular calcium chelator-BAPTA. Therefore, the intracellular Ca2+ concentration ([Ca2+]i) may play an important role in beauvericin-induced cell death in CCRF-CEM cells. In this study, the effect of beauvericin on [Ca2+]i and the possible mechanism responsible

Ochratoxin A induces oxidative DNA damage in liver and kidney after oral dosing to rats.

The nephrotoxic/carcinogenic mycotoxin ochratoxin A (OTA) occurs as a contaminant in food and feed and may be linked to human endemic Balkan nephropathy. The mechanism of OTA-derived carcinogenicity is still under debate, since reactive metabolites of OTA and DNA adducts have not been unambiguously identified. Oxidative DNA damage, however, has been observed in vitro after incubation of mammalian cells with OTA. In this study, we investigated whether OTA induces oxidative DNA damage in vivo as well. Male F344 rats were dosed with 0, 0.03, 0.1, 0.3 mg/kg bw per day OTA for 4 wk (gavage, 7 days/

Investigation of the effects of patulin on thyroid and testis, and hormone levels in growing male rats.

Patulin is a mycotoxin produced by several species of Penicillium, Aspergillus and Byssachlamys. Patulin can be produced on different food products including fruits, grains, cheese, cured meats, but in natural situations patulin is usually found in apple and apple products. In the present study, the time-dependent effects of patulin on the T3, T4, thyroid stimulating hormone, testosterone, luteinizing hormone and growth hormone levels of growing male rats were investigated. Patulin, at a dose of 0.1 mg/kg bw/day, was administered by gavage to growing male rats aged 5-6 weeks for a period of 60

Identification and functional assessment of the novel murine organic anion transporter Oat5 (Slc22a19) expressed in kidney.

An uncharacterized murine cDNA clone was identified and, through sequence, phylogenetic, and functional analysis, determined to encode the newest member of the organic anion transporter family, organic anion transporter 5 (Oat5; Slc22a19). The Oat5 cDNA clone contained an insert 1,964 bp in length with a predicted open reading frame (from bp 84 to bp 1,739) coding for a peptide 551 amino acids long. Slc22a19 was localized to mouse chromosome 19 near the genes encoding Oat1 (Slc22a6) and Oat3 (Slc22a8). Northern blot analysis revealed Oat5 is highly expressed in the kidney of adult mice and rat

Reduction of patulin during apple juice clarification.

Patulin is a mycotoxin produced by a number of molds involved in fruit spoilage. This compound is carcinogenic and teratogenic. Various methods are currently used to reduce the levels of patulin in apple juice, namely, charcoal treatment, chemical preservation (sulfur dioxide), gamma irradiation, fermentation, and trimming of fungus-infected apples. Many of these processes are expensive and time-consuming. Therefore, there is a need to find a convenient and economical process to control patulin levels. This study was undertaken to evaluate the effectiveness of several clarification processes f

Endosome motility controls light-responsive reproductive development and secondary metabolite production in Aspergillus.

Filamentous fungi, such as Aspergillus species, use microtubule transport to move early endosomes. Other cargos, such as peroxisomes and mRNAs, "hitchhike" on early endosomes to move throughout the long hyphae of these organisms. In Aspergillus nidulans, peroxisomes hitchhike on early endosomes using the endosomal protein PxdA and the peroxisomal protein AcbdA. The HookA adaptor protein links endosomes to microtubule motors. Here, we set out to explore the physiological functions of peroxisome hitchhiking and endosome motility. A. nidulans has a complex life cycle that includes asexual and sex

Decrypting potential mechanisms linking ochratoxin A to hepatocellular carcinoma: an integrated approach combining toxicology, machine learning, molecular docking, and molecular dynamics simulation.

BACKGROUND: Ochratoxin A (OTA), a common food-borne mycotoxin, is a potential human carcinogen, yet the specific molecular mechanisms linking it to hepatocellular carcinoma (HCC) remain unclear. METHODS: We integrated network toxicology to predict OTA targets and intersected them with HCC transcriptomic data to identify key candidate genes. Functional enrichment analysis was then conducted. Multiple machine learning algorithms were applied to screen and validate core genes. Furthermore, molecular docking and molecular dynamics (MD) simulations were employed to evaluate the binding stability b

Beauvericin Induces Mitochondrial Apoptosis and Attenuates EMT-Associated Phenotypes and Angiogenic Signaling in Colorectal Cancer Cells In Vitro.

Colorectal cancer (CRC) remains a leading cause of cancer-related mortality, largely due to metastasis and therapeutic resistance. Beauvericin, a cyclohexadepsipeptide mycotoxin produced by Beauveria and Cordyceps species, has demonstrated anticancer activity in multiple malignancies; however, its mechanistic effects in CRC have not been fully defined. In this study, we investigated the cellular and molecular effects of beauvericin in HCT116 and SW480 CRC cells using in vitro models. Beauvericin reduced cell viability and clonogenic growth, induced G0/G1 cell-cycle arrest, and activated mitoch

Covalent Michael-type recognition-based dual-mode sensing platform using thiol-functionalized nitrogen-doped carbon dots and Fe-MOF nanozyme for ultrasensitive patulin detection.

Patulin is a toxic mycotoxin commonly found in apple and fruit products, requiring sensitive and reliable detection methods. Herein, a novel dual-mode sensing platform integrating turn-off fluorescence and colorimetric detection was developed for rapid patulin determination in apple juice. The strategy exploits patulin's unique electrophilic reactivity toward thiols across two independent signal channels. Thiol-functionalized nitrogen-doped carbon dots (SH-N-CDs) serve as both recognition element and signal transducer, where direct covalent Michael-type addition of patulin onto the CD surface

An in-situ photoderivative fluorescence enhancement method without chromatographic separation for Aflatoxin B1 (AFB1) detection.

Aflatoxin B1 (AFB1), a potent carcinogenic mycotoxin, poses critical health risks to both animals and humans. Thus, rapid and highly sensitive detection of AFB1 is crucial for ensuring food safety. However, current AFB1 detection methods face a trade-off between laboratory-level sensitivity and on-site immediacy. Instead of relying on absolute fluorescence intensity, this study exploits the UV-triggered addition at the terminal furan double bond of AFB1, which yields a large fluorescence enhancement (ΔF), thereby proposing a novel quantification strategy centered on this photo-induced signal i

Heterologous Expression of Cryphonectria Hypovirus 1 in Fusarium verticillioides Reveals Stable Replication and Reduced Fumonisin Production.

Mycoviruses represent promising tools for the biological control of phytopathogenic fungi. However, their application is often constrained by host specificity and low natural prevalence in certain pathogens. Fusarium verticillioides, a major maize pathogen and fumonisin B1 (FB1) producer, harbors very few known mycoviruses, a circumstance that limits the development of virus-based control strategies. In this study, we explored a virocontrol approach for F. verticillioides based on the use of a heterologous mycovirus, Cryphonectria hypovirus 1 (CHV1). For the artificial transfection of F. verti

Deoxynivalenol exposure promotes apoptosis and disrupts GnRH-dependent LH production in pituitary gonadotrope-like cells.

Deoxynivalenol (DON) is a highly prevalent mycotoxin contaminating cereals and cereal-based products and is increasingly recognized for its adverse effects on reproductive function. DON is known to activate mitogen-activated protein kinases (MAPKs), which are also critical mediators of gonadotropin-releasing hormone (GnRH)-dependent synthesis and secretion of luteinizing hormone (LH) and follicle-stimulating hormone (FSH) by pituitary gonadotrope cells. Despite this mechanistic overlap, the direct endocrine-disrupting effects of DON on pituitary gonadotropin production remain poorly understood

How temperature reshapes interactions in mixed cultures of mycotoxigenic fungi.

Climate change is shifting the distribution of mycotoxigenic fungi, which may alter patterns of co-occurrence in staple crops. Yet, quantitative, reproducible in vitro approaches to characterise multispecies growth and interaction outcomes under environmentally relevant conditions remain limited. Here, we developed a small-scale workflow to quantify temperature-dependent interaction outcomes among three major cereal-associated fungi, Aspergillus flavus, Fusarium verticillioides, and F. graminearum. High-frequency optical density time series were collected in 96-well liquid cultures across 10 °
