The effect of electrophoretic lignocaine blockade in the region of the carotid sinuses on spontaneous and water diuresis.

The effect of electrophoretic lignocaine blockade in the region of the carotid sinuses on changes in the intensity of spontaneous and water diuresis was investigated on anaesthetized dogs. After the blockade a reduction in spontaneous diuresis was observed and a marked and statistically significant restriction of water diuresis was noted as compared with the control experiments. The reduction in diuresis after the blockade was of a brief duration and did not appear to be related to changes in the intensity of the filtration through the kidneys, which was only slightly lower, but to changes in

Bilateral lesions of the medial longitudinal fasciculus in monkeys: effects on the horizontal and vertical components of voluntary and vestibular induced eye movements.

Bilateral transections across the brainstem interrupted the medial longitudinal fasciculus (MLF) in three monkeys trained to make eye movements while subjected to horizontal or vertical angular accelerations. Eye movements measured before and after the lesion revealed deficits in both voluntary and vestibular compensatory eye movements; the deficits differed in the horizontal and vertical directions. Vertical saccades in both directions were normal but eccentric positions of fixation could not be maintained; a drift toward the midline followed by a corrective saccade produced vertical fixation

Fine structure and distribution of axon terminals from the cochlear nucleus on neurons in the medial superior olivary nucleus of the cat.

The morphology and distribution of axon terminals on central column and marginal neurons of the cat medial superior olivary nucleus (MSO) were analyzed by electron microscopy. Individual neurons or groups of cells oriented such that substantial lengths of their dendrites were within a 5-7 mu thich section were selected for detailed study. Thin sections were cut from remounted thick sections. Boutons with spherical vesicles arise directly from myelinated axons; more than one synaptic region of an axon, each separated by a myelinated segment, may contact a given dendrite. Boutons with flattened

Notch Signaling Regulates the Neuroprotective Effects of hUCMSCs in a Mouse Model of Cerebral Ischemia.

Human umbilical cord mesenchymal stem cells (hUCMSCs) have shown promise for treating ischemic stroke (IS) by promoting neural repair. The Notch signaling pathway is known to regulate neural stem cell (NSC) proliferation and differentiation, but its role in hUCMSC-mediated neuroprotection remains unclear. This study aimed to evaluate the therapeutic effects of hUMSCs in a mouse model of middle cerebral artery occlusion (MCAO) and investigate the involvement of the Notch pathway. Fifth generation of hUCMSCs were characterized by flow cytometry. In vitro, hUCMSCs were induced to differentiate wi

Neural mechanisms underlying loss of functional independence after stroke: EEG evidence of impaired motor-to-sensory directed connectivity on the affected side.

BACKGROUND: Post-stroke reorganization of the prefrontal-sensorimotor network is critical for functional recovery, yet single connectivity metrics fail to capture its multidimensional characteristics. This exploratory study investigates network reorganization patterns and their clinical relevance to motor function and daily independence using multimodal electroencephalography (EEG) connectivity analysis, including Coherence (COH), phase lag index (PLI), and Granger causality (GC). METHODS: Resting-state EEG from stroke patients (n = 22) and healthy controls (n = 22) was analyzed. COH, PLI, an

Control of representation updating by higher-order thalamus enables history-based decision-making.

When sensory information is ambiguous, we must interpret current inputs based on a representation constructed from our previous experiences. Although appropriately updating this internal representation to track environmental changes is essential for behavior, the neural basis of this dynamic integration remains unknown. Using a combination of multi-area single-unit recordings and optogenetic techniques in mice under both behavioral and passive conditions, we find that interactions between the posterior parietal cortex (PPC) and its higher-order thalamic counterpart, the pulvinar (PUL), are nec

N-Acetylcysteine is associated with changes in functional connectivity in patients with Parkinson's disease.

INTRODUCTION: This study assessed the changes in functional connectivity from resting functional magnetic resonance imaging (fMRI) in patients with Parkinson's disease (PD) given N-Acetylcysteine (NAC), the prodrug to L-cysteine and a precursor to the natural biological antioxidant glutathione (GSH). The aim of this study was to determine whether NAC is associated with changes in functional connectivity, particularly in the basal ganglia, and improvements in Parkinson's symptoms. METHODS: Forty-four patients with PD were randomized to either weekly intravenous infusions of NAC (50 mg/kg) plus

VTA-forebrain connectivity moderates adaptive behavior.

Neural representation of the environment is pertinent for adaptive behaviors. Such neural maps are processed in the hippocampus and contain information about spatial locations, novelties, context, and cues for context discrimination. In conjunction with other subcortical structures, a key function of the hippocampus is to compare newly detected novelties derived from the environment with previously stored cortical memories. These neural events underscore the role of the hippocampus as a “comparator” and “hub” for sorting novelties to determine priority and weight assignments for long-term cort

A communication subspace relays context-dependent actions from human prefrontal to motor cortex.

Adaptive behavior relies on the ability to translate abstract rules and goals into actions suited to the current context. Neural population activity in the prefrontal cortex (PFC) has been proposed to support such flexible computations through high-dimensional dynamics, whereas activity in the primary motor cortex (M1) is related more directly to movement execution. How contextual representations in PFC are transformed into ensuing action plans within M1 remains unknown. Previous work suggests that low-dimensional coding subspaces might organize interareal communication, but direct evidence fo

Beyond fear circuits: multiscale neurobiological architecture of panic disorder.

Panic disorder is a prevalent and disabling condition marked by recurrent panic attacks and high treatment resistance. While previous research has focused on dysfunction within canonical fear circuits, the neurobiological basis of panic disorder may involve broader alterations across multiple brain systems. Here, we conducted a meta-analysis of functional neuroimaging experiments to identify consistent patterns of altered brain activity in panic disorder. We found increased activity in a prefrontal-hippocampus-brainstem axis, which was not confined to traditional fear-related regions. This pat

Mesoscale connectivity of the human hippocampus and fimbria revealed by ex vivo diffusion MRI.

The human hippocampus is essential to cognition and emotional processing. Its function is defined by its connectivity. Although some pathways have been well-established, our knowledge about anterior-posterior connectivity and the distribution of fibers from major fiber bundles remains limited. Mesoscale (250 μm isotropic acquisition, upsampled to 125 μm) resolution MR images of the human temporal lobe afforded a detailed visualization of fiber tracts, including those that related anterior-posterior substructures defined as subregions (head, body, tail) and subfields (cornu ammonis 1-3, dentate

Assessment of resting state structural-functional relationships in perisylvian region during the early weeks after birth.

This study investigates the structural-functional (S-F) relationships of the perisylvian region during the first weeks after birth in the resting state. Previous joint S-F studies of perisylvian development were mainly conducted on individual structural and functional metrics. By employing a weighted combination of metrics, joint S-F studies can enhance the understanding of perisylvian development in neonates, thereby providing valuable features for the prediction of neurodevelopmental disorders. To this end, we employed both structural and functional metrics derived from 16 perisylvian sub-re

Distinct representations of economic variables across regions and projections of the frontal cortex.

Economic decision-making requires evaluating information about available options, such as their expected value and economic risk. Previous studies have shown that frontal cortical neurons encode these variables, but how this encoding is structured across different frontal regions and projection pathways remains unclear. We developed a decision-making task for head-fixed mice in which we varied the expected value and risk associated with reward-predicting stimuli. Using large-scale electrophysiology, two-photon imaging, and projection-specific optotagging, we identified distinct spatial gradien

Establishing dorsal-ventral patterning in human neural tube organoids with synthetic organizers.

Precise dorsal-ventral (D-V) patterning of the neural tube (NT) is essential for the development and function of the central nervous system. However, existing models for studying NT D-V patterning and related human diseases remain inadequate. Here, we present organizers derived from pluripotent stem cell aggregate fusion ("ORDER"), a method that establishes opposing BMP and SHH gradients within neural ectodermal cell aggregates. Using this approach, we generated NT organoids with ordered D-V patterning from both zebrafish and human pluripotent stem cells (hPSCs). Single-cell transcriptomic ana

Hippocampal neural circuit mechanisms in Alzheimer's disease revealed by viral-genetic circuit mapping.

Alzheimer's disease (AD) is a progressive neurodegenerative disorder with growing major health impacts in countries with aging populations. Existing therapeutic approaches that have been based on neurochemical and neuropathological findings are largely ineffective. This lack of progress suggests we require a new framework for future AD therapies. The examination of neural circuit mechanisms in AD mouse models is an emerging focus for identifying new AD treatment strategies. We now know there are neural circuit-level maladaptive alterations in AD brains, some of which appear very early in the d

Two different mirror neuron pathways for social and non-social actions? A meta-analysis of fMRI studies.

Mirror neurons (MNs) represent a class of neurons that are activated when performing or observing the same action. Given their role in social cognition and previous research in patients with psychiatric disorders, we proposed that the human MN system (MNS) might display different pathways for social and non-social actions. To examine this hypothesis, we conducted a comprehensive meta-analysis of 174 published human functional magnetic resonance imaging studies. Our findings confirmed the proposed hypothesis. Our results demonstrated that the non-social MN pathway exhibited a more classical pat

A dynamic computational model of the parallel circuit on the basal ganglia-cortex associated with Parkinson's disease dementia.

The cognitive impairment will gradually appear over time in Parkinson's patients, which is closely related to the basal ganglia-cortex network. This network contains two parallel circuits mediated by putamen and caudate nucleus, respectively. Based on the biophysical mean-field model, we construct a dynamic computational model of the parallel circuit in the basal ganglia-cortex network associated with Parkinson's disease dementia. The simulated results show that the decrease of power ratio in the prefrontal cortex is mainly caused by dopamine depletion in the caudate nucleus and is less relate

α-Synuclein aggregation decreases cortico-amygdala connectivity and impairs social behavior in mice.

Abnormal accumulation of insoluble α-synuclein (α-Syn) inclusions in neurons, neurites, and glial cells is the defining neuropathology of synucleinopathies, including Parkinson's disease (PD), dementia with Lewy bodies (DLB), and multiple system atrophy. Accumulation of α-Syn inclusions in the amygdala has been well-documented in post-mortem studies of PD and DLB brains, as well as preclinical animal models of these conditions. Though α-Syn pathology is closely associated with neurodegeneration, there is a poor correlation between neuronal loss in the amygdala and the clinical features of PD a

Neuronal network controlling REM sleep.

Rapid eye movement sleep is a state characterized by concomitant occurrence of rapid eye movements, electroencephalographic activation and muscle atonia. In this review, we provide up to date knowledge on the neuronal network controlling its onset and maintenance. It is now accepted that muscle atonia during rapid eye movement sleep is due to activation of glutamatergic neurons localized in the pontine sublaterodorsal tegmental nucleus. These neurons directly project and excite glycinergic/γ-aminobutyric acid-ergic pre-motoneurons localized in the ventromedial medulla. The sublaterodorsal tegm
