Multivariate Pattern Classification of Primary Insomnia Using Three Types of Functional Connectivity Features.

Objective: To explore whether or not functional connectivity (FC) could be used as a potential biomarker for classification of primary insomnia (PI) at the individual level by using multivariate pattern analysis (MVPA). Methods: Thirty-eight drug-naive patients with PI, and 44 healthy controls (HC) underwent resting-state functional MR imaging. Voxel-wise functional connectivity strength (FCS), large-scale functional connectivity (large-scale FC) and regional homogeneity (ReHo) were calculated for each participant. We used support vector machine (SVM) with the three types of metrics as feature

Cerebrovascular longitudinal atlas: Changes in cerebral arteries in unruptured intracranial aneurysm patients followed with MRA.

BACKGROUND: Patterns of change in cerebrovascular (CV) morphology associated with aging are highly relevant to the incidence and progression of CV disease, particularly stroke. Intracranial aneurysms (IA), a leading cause of hemorrhagic stroke, are linked with factors such as blood flow, arterial stiffness, and inflammation that may also drive other changes in CV morphology. We worked with a cohort of longitudinally-imaged IA patients to construct the first longitudinal atlas of CV morphology and studied its relationship with disease. METHODS: 110 IA patients, ranging from 19 to 84 years old

Development of prognostic model incorporating a ferroptosis/cuproptosis-related signature and mutational landscape analysis in muscle-invasive bladder cancer.

BACKGROUND: Muscle-invasive bladder cancer (MIBC) is a prevalent and aggressive malignancy. Ferroptosis and cuproptosis are recently discovered forms of programmed cell death (PCD) that have attracted much attention. However, their interactions and impacts on MIBC overall survival (OS) and treatment outcomes remain unclear. METHODS: Data from the TCGA-BLCA project (as the training set), cBioPortal database, and GEO datasets (GSE13507 and GSE32894, as the test sets) were utilized to identify hub ferroptosis/cuproptosis-related genes (FRGs and CRGs) and develop a prognostic signature. Phase 3 diagnostic evaluation of a smart tablet serious game to identify autism in 760 children 3-5 years old in Sweden and the United Kingdom.

INTRODUCTION: Recent evidence suggests an underlying movement disruption may be a core component of autism spectrum disorder (ASD) and a new, accessible early biomarker. Mobile smart technologies such as iPads contain inertial movement and touch screen sensors capable of recording subsecond movement patterns during gameplay. A previous pilot study employed machine learning analysis of motor patterns recorded from children 3-5 years old. It identified those with ASD from age-matched and gender-matched controls with 93% accuracy, presenting an attractive assessment method suitable for use in the

Integrative profiling of multi-modal plasma cfRNA signatures enables detection and prognostic risk stratification in breast cancer.

Breast cancer requires non-invasive biomarkers for accurate detection and risk stratification. We comprehensively profiled plasma cell-free RNA (cfRNA) from 41 patients with malignant and 42 with benign breast lesions using SLiPiR-seq. Multiple cfRNA subtypes displayed distinct expression patterns, and machine-learning models were developed with repeated stratified four-fold cross-validation. The integrated cfRNA model achieved a mean AUC of 0.795, while the cf-miRNA model performed best (AUC: 0.814) and was externally validated in an independent cohort (AUC: 0.867). A three-gene tissue expres

Predictive model for proliferative diabetic retinopathy using single-cell transcriptomics.

Proliferative diabetic retinopathy (PDR) is a severe and advanced complication of diabetic retinopathy, representing a leading cause of blindness worldwide. A hallmark of PDR is the formation of fibrovascular membranes (FVMs), which pose a significant threat to visual function due to their propensity to contract and exert traction on the retina. A comprehensive understanding of the underlying molecular mechanisms is essential for addressing this condition effectively. Herein, we first integrated scRNA-seq data from nine FVMs of PDR patients and retinas from healthy donors obtained from the Gen

Accurately assessing congenital heart disease using artificial intelligence.

Congenital heart disease (CHD) remains a significant global health challenge, particularly contributing to newborn mortality, with the highest rates observed in middle- and low-income countries due to limited healthcare resources. Machine learning (ML) presents a promising solution by developing predictive models that more accurately assess the risk of mortality associated with CHD. These ML-based models can help healthcare professionals identify high-risk infants and ensure timely and appropriate care. In addition, ML algorithms excel at detecting and analyzing complex patterns that can be ov

An Alzheimer's disease category progression sub-grouping analysis using manifold learning on ADNI.

Many current statistical and machine learning methods have been used to explore Alzheimer's disease (AD) and its associated patterns that contribute to the disease. However, there has been limited success in understanding the relationship between cognitive tests, biomarker data, and patient AD category progressions. In this work, we perform exploratory data analysis of AD health record data by analyzing various learned lower dimensional manifolds to separate early-stage AD categories further. Specifically, we used Spectral embedding, Multidimensional scaling, Isomap, t-Distributed Stochastic N

MRI-based Radiomics and volumetrics for predicting the onset of Alzheimer's Disease with explainable machine learning.

The detection of Alzheimer's Disease (AD) using structural Magnetic Resonance Imaging (MRI) and Machine Learning (ML) often focuses on late-stage atrophy patterns. End-to-end deep learning models address this by considering MRI signal intensities. However, their explainability components typically focus on attention regions, neglecting underlying patterns. This work overcomes both problems by training and explaining time-to-event models utilizing Radiomics features. SHapley Additive exPlanations (SHAP) and high-level explanations were combined to interpret the effects of MRI texture, shape, an
