Beam collimation and filtration optimization for a novel orthovoltage radiotherapy system.

BACKGROUND: The inaccessibility of clinical linear accelerators in low- and middle-income countries creates a need for low-cost alternatives. Kilovoltage (kV) x-ray tubes have shown promise as a source that could meet this need. However, performing radiotherapy with a kV x-ray tube has numerous difficulties, including high skin dose, rapid dose fall-off, and low dose rates. These limitations create a need for highly effective beam collimation and filtration. PURPOSE: To improve the treatment potential of a novel kV x-ray system by optimizing an iris collimator and beam filtration using Bayesi

Optimizing Workplace Digital Mental Health Interventions: Systematic Review and Meta-Analysis.

BACKGROUND: Digital mental health interventions (DMHIs) are widely used in workplaces to address common mental health conditions such as depression, anxiety, and stress. Their content and design vary widely, and meta-analyses repeatedly found substantial heterogeneity in their efficacy, but there is no systematic evaluation of the comparative effect of content on efficacy. OBJECTIVE: We aimed to identify whether intervention design, therapeutic approaches, and content features are associated with greater efficacy in randomized controlled trials (RCTs) of DMHIs in workplaces. METHODS: We cond

Noisy-channel language comprehension in aphasia: A Bayesian mixture modeling approach.

Individuals with "agrammatic" receptive aphasia have long been known to rely on semantic plausibility rather than syntactic cues when interpreting sentences. In contrast to early interpretations of this pattern as indicative of a deficit in syntactic knowledge, a recent proposal views agrammatic comprehension as a case of "noisy-channel" language processing with an increased expectation of noise in the input relative to healthy adults. Here, we investigate the nature of the noise model in aphasia and whether it is adapted to the statistics of the environment. We first replicate findings that a

Inter-individual variability in the relationship between propulsion force and walking speed in subacute stroke.

Walking speed is primarily driven by the propulsion force generated during the late stance phase, but the translation of propulsion force to walking speed may differ across individuals. This study aimed to investigate inter-individual variability in the relationship between propulsion force and walking speed in patients with subacute stroke and identify the clinical characteristics associated with this variability. Twenty-five participants with subacute stroke performed walking trials at self-selected and highest possible speeds. We applied hierarchical linear modeling, which allows the estima

Estimation of Genetic Parameters for OPU-IVF-Related Traits in Japanese Black Cattle.

Ovum pick-up (OPU)-in vitro fertilization (IVF), a technique in which cumulus oocyte complexes (COCs) are aspirated from the ovaries of cows and embryos are produced through IVF, is a method that allows repeated oocyte collection and embryo production without the need for slaughtering cattle and is used in breeding of Japanese Black cattle. However, there is no research on OPU-IVF-related traits in Japanese Black cattle from a breeding perspective. Therefore, we estimated genetic parameters of OPU-IVF-related traits in Japanese Black cows farmed in Hiroshima Prefecture. We used four OPU-relate

MPNN-CWExplainer: An enhanced deep learning framework for HIV drug bioactivity prediction with class-weighted loss and explainability.

AIMS: Human Immunodeficiency Virus (HIV) remains a critical global health concern due to its impact on the immune system and its progression to Acquired Immunodeficiency Syndrome (AIDS) if untreated. While antiretroviral therapy has advanced significantly, challenges such as drug resistance, adverse effects, and viral mutation necessitate the development of novel therapeutic strategies. This study aims to improve HIV bioactivity prediction and provide interpretable insights into molecular determinants influencing bioactivity. MATERIALS AND METHODS: We propose MPNN-CWExplainer, a novel graph-b

Multilevel Regression and Poststratification Using Margins of Poststratifiers: Improving Inference for HIV Health Outcomes During the COVID-19 Pandemic.

Multilevel regression and poststratification (MRP) has surged in popularity for population inference using survey samples. The method consists of two stages: First, fitting a regularized model that regresses the outcome on poststratification variables; second, predicting the outcome using the regularized model and aggregating the predictions to make population inferences. Existing MRP methods mostly focus on settings where the joint distribution of the poststratifiers is known in the population. However, in practice, such data information is often not available; instead, we are provided with t

Protocol: a systematic review+ (SR+) to combine associative and mechanistic evidence on the efficacy of face masks in reducing transmission of respiratory diseases.

BACKGROUND: Mechanistic evidence is evidence about how an intervention works. A 2023 Cochrane review, which was restricted to randomised controlled trials (RCTs), concluded that evidence on the efficacy of face masks was weak, conflicting and non-definitive. A 2024 narrative review, which included RCTs plus mechanistic evidence on masks and mask mandates, concluded that evidence of efficacy was strong, consistent and definitive. These strikingly contrasting conclusions reflect differences in how evidence is valued. Orthodox synthesis methods (e.g. those used for Cochrane reviews, informed by G

Invited commentary: motivating better methods-and better data collection-for measuring the prevalence of drug misuse.

The United States continues to suffer a drug overdose crisis that has resulted in over 100 000 deaths annually since 2021. Despite decades of attention, estimates of the prevalence of drug use at the spatiotemporal resolutions necessary for resource allocation and intervention evaluation are lacking. Current approaches for measuring the prevalence of drug use, such as population surveys, capture-recapture, and multiplier methods, have significant limitations. In a recent article, Santaella-Tenorio et al (Am J Epidemiol. 2024;193(7):959-967) used a novel joint bayesian spatiotemporal modeling a

A deep learning-based method for predicting the frequency classes of drug side effects based on multi-source similarity fusion.

MOTIVATION: Drug side effects refer to harmful or adverse reactions that occur during drug use, unrelated to the therapeutic purpose. A core issue in drug side effect prediction is determining the frequency of these drug side effects in the population, which can guide patient medication use and drug development. Many computational methods have been developed to predict the frequency of drug side effects as an alternative to clinical trials. However, existing methods typically build regression models on five frequency classes of drug side effects and tend to overfit the training set, leading to

Modeling and optimization of bioproduct formation with purple phototrophic bacteria using machine learning.

Municipal and industrial wastewater, along with organic waste, can be transformed into valuable bioproducts using purple phototrophic bacteria. This study compares the performance of three machine learning models (Random Forest, XGBoost, and CatBoost) in predicting and optimizing the formation of key bioproducts: polyhydroxybutyrate, polyhydroxyvalerate, 5-aminolevulinic acid, coenzyme Q10, carotenoids, bacteriochlorophylls, and biomass. The models were trained on a dataset compiled from previous studies, using input variables such as reaction time, concentration of organic matter, ethanol, bi

High-Throughput Optimization of a High-Pressure Catalytic Reaction.

High-throughput optimization of a hydroformylation reaction using CO2 instead of CO was performed through Bayesian optimization in combination with a high-throughput screening system. CO2 and H2 pressure as well as catalyst composition were efficiently optimized by transferring a surrogate model, constructed through catalyst composition optimization, for the comprehensive optimization of the entire search space. This method successfully increased the aldehyde yield by 1.5 times compared to that reported in the literature with a combination of small amounts of Rh and Ru catalysts combined with

CKD-M2 study: 2-year mortality prediction tool for advanced kidney disease.

BACKGROUND: A few models have been developed in recent years to predict all-cause mortality in patients with chronic kidney disease (CKD). However, many have been developed using inappropriate methods and have not been externally validated. This study aims to improve our previously validated tool for predicting 2-year all-cause mortality in stage 4-5 CKD patients by enlarging the training dataset, thereby enhancing its robustness, which is further supported by a second external validation. METHOD: The Bayesian network-based 2-year all-cause mortality prediction tool was trained on a comprehen

Cognitive performance and lifetime occupational exposures in a regional Australian population: a distributed lag mixtures approach.

Examination of exposures over a lifetime of employment and work-related disease has largely focused on the effects of single agents. In reality, workers are exposed to mixtures of agents. In a community cohort of 1621 older participants (Hunter Community Study) with a mean age of 68.0 years (SD 6.8 yr), cognitive performance was assessed using the Audio Recorded Cognitive Screen, and lifetime occupational histories used to retrospectively estimate yearly exposures to a range of chemicals from the Finnish job-exposure matrix. This study utilised Bayesian Kernel Machine Regression - Distributed

Influence of Exercise Heat Acclimation Protocol Characteristics on Adaptation Kinetics: A Quantitative Review With Bayesian Meta-Regressions.

The integrative influence of heat acclimation (HA) protocol characteristics and approach on adaptation kinetics and exercise capacity/performance in the heat remains unclear. Bayesian multilevel regression models were used to estimate adaptations with the number of exposures, exposure duration, ambient temperature, water vapor pressure, and HA approach (e.g., constant workrate) as predictors. Data from 211 papers were included in meta-analyses with results presented as posterior means and 90% credible intervals. Mean protocol characteristics were as follows: 8 ± 4 exposures, 90 ± 36 min/exposu

Efficacy and Safety of Lithium for Behavioral and Cognitive Symptoms in Alzheimer's Disease Dementia: A Systematic Review With Frequentist and Bayesian Meta-Analysis.

BACKGROUND: Alzheimer's disease (AD) dementia is the leading cause of cognitive decline in late life, yet treatment options remain limited. Lithium, widely used in bipolar disorder, has been suggested to exert neuroprotective effects through inhibition of GSK-3β and modulation of amyloid and tau pathology. We aimed to evaluate the efficacy and safety of lithium in AD dementia. METHODS: This systematic review and meta-analysis was prospectively registered in PROSPERO and conducted following PRISMA guidelines. We searched PubMed, Embase, and Cochrane Library through April 2025 for randomized co
