Opportunistic pathogens are a major threat to people, especially those with impaired immune systems. Two of the most important microbes in this category are the fungus Candida albicans and Gram-positive bacteria of the genus Enterococcus, which share overlapping niches in the oral cavity, gastrointestinal and urogenital tracts. The clinical importance of oral C. albicans biofilm and its interaction with the host under immunosuppressive conditions remains largely understudied. Here, we used a mouse model of oropharyngeal candidiasis (OPC) with cortisone acetate injection on alternate days and a

Candida albicans (C. albicans) is a dimorphic commensal human fungal pathogen that can cause severe oropharyngeal candidiasis (oral thrush) in susceptible hosts. During invasive infection, C. albicans hyphae invade oral epithelial cells (OECs) and secrete candidalysin, a pore-forming cytolytic peptide that is required for C. albicans pathogenesis at mucosal surfaces. Candidalysin is produced in the hyphal invasion pocket and triggers cell damage responses in OECs. Candidalysin also activates multiple MAPK-based signaling events that collectively drive the production of downstream inflammatory

BACKGROUND: Mucormycosis is an invasive fungal disease severely complicating treatment of patients with hematologic diseases. Effective therapy is represented by the combination of surgery and amphotericin B administration and early initiation of the therapy is necessary for favorable outcome. The first clinical symptoms are usually non-specific and this can lead to late therapy onset. The objective of this retrospective work was to determine the frequency, risk factors and outcome of invasive mucormycosis in pediatric hematology patients.
MATERIAL AND METHODS: The study cohort comprised 399

PURPOSE: Endophthalmitis caused by endogenous Candida and Aspergillus species has emerged as a visually threatening complication in patients with immune deficiency of various causes. Twenty-five patients who underwent enucleation, 13 with endogenous Aspergillus endophthalmitis and 12 with endogenous Candida intraocular infections, were evaluated. Both clinical features and intraocular spread of the fungi were studied to determine which clinical and/or histopathologic features could help distinguish aspergillosis from Candida infections.
METHODS: Clinical information was sought from each case

Objectives: Oral potentially malignant disorders may present as white, red, or mixed red-white lesions and require accurate early recognition. This study evaluated whether texture-analysis features extracted from clinical digital images could distinguish oral precancerous lesions from other oral mucosal lesions and normal mucosa using gray-level co-occurrence matrix (GLCM), gray-level run-length matrix (GLRLM), and wavelet analysis. Materials and Methods: Sixty-four clinical digital images were selected according to predefined inclusion and exclusion criteria. The dataset included leukoplakia,

Dental infections continue to pose a major health concern worldwide, driven by persistent microbial biofilms, dysregulated host responses, and suboptimal efficacy of conventional antimicrobial therapies. The emergence of nanotechnology has facilitated the development of therapeutic approaches that offer spatiotemporal, minimally invasive, and resistance-preventing alternatives for treating complex dental infections. This review critically assesses novel nanomaterial-based therapies designed to directly eradicate pathogenic microorganisms involved in periodontal disease, dental caries, oral can

Invasive fungal infections (IFIs) represent a serious global health threat due to rising incidences and drug resistance. Traditional antifungal therapies face limitations, including toxicity, rapid clearance, and resistance, demanding nanotechnology-driven solutions. While lipid-based formulations, such as AmBisome, have achieved clinical success, scalability is restricted by high costs and complex manufacturing. This study advances the solvent-free approach by developing "ThermoSomes", bilayered lipid vesicles encapsulating the FDA-approved antifungal Posaconazole (PCZ) for IFI treatment. The

SUMMARYCandida species are major yeast pathogens that cause both mucosal candidiasis and life-threatening invasive infections. Most Candida species, including Candida albicans, have long been thought to be "imperfect" due to the lack of a complete sexual reproduction cycle. Since the discovery of the mating type-like locus in C. albicans in 1999, the regulation of (para)sexual reproduction has been intensively investigated in this organism as well as in several phylogenetically closely related species. The (para)sexual cycle is not only critical for the generation of genetic and phenotypic div

Candidiasis belongs to common fungal infections and is usually mild and self-limiting. However, in patients with immunodeficiencies, it can transform into invasive infections with high mortality. Long-term antifungal treatment can lead to the emergence of resistance. The problem is further complicated by the development of fungal biofilm resistant to conventional antimicrobials. Due to a limited choice of available antifungals, the development of novel active agents, such as antimicrobial peptides (AMPs), is highly desirable. Human cathelicidin LL-37 is an intensively studied AMP with a confir

Contemporary data on the epidemiology and outcomes of invasive candidiasis (IC) in solid organ transplant recipients (SOTr) are limited. This retrospective multicenter cohort study, nested within the Swiss Transplant Cohort Study, describes the epidemiology and outcomes of IC in SOTr from 2008 to 2020. Among 4755 patients identified, 205 (4.3%) had 262 episodes of IC. One-year cumulative incidence of IC was 2.8% (95% confidence interval [CI], 2.4, 3.3) and decreased during the study period (P = .046). Candidemia was less frequently encountered (0.67%, 95% CI, 0.47, 0.94) than intra-abdominal (

With increasing concern about the negative health impact of fungal disease, there is a need to survey what is and is not known about the epidemiology of these infections in Tunisia. We have estimated the incidence and prevalence of the most serious fungal diseases in Tunisia for the first time. Using published literature from Tunisia, or if absent other countries, we have estimated the burden of life-threatening fungal infections and those causing significant morbidity, using deterministic modeling, based on populations at greatest risk. An estimated 250,494 (2.12% of the Tunisian population)

Candida auris, an emerging non-albicans multidrug-resistant yeast, has become a significant cause of invasive candidiasis in healthcare settings. So far, data on the metabolites of C. auris in different clades are minimal, and no studies have focused on clade V metabolites. Therefore, Gas chromatography-mass spectrometry (GC-MS) was used for the metabolomic profiling of clade I C. auris compared with fluconazole-resistant and susceptible C. auris in clade V strains. GC-MS chromatography revealed 28, 22, and 30 compounds in methanolic extracts of the fluconazole-susceptible and fluconazole-resi

Invasive fungal infections (IFIs) are frequent and outcome-relevant complications in the early postoperative period after orthotopic liver transplantation (OLT). Recent guidelines recommend targeted antimycotic prophylaxis (TAP) for high-risk liver transplant recipients (HR-LTRs). However, the choice of antimycotic agent is still a subject of discussion. Echinocandins are increasingly being used due to their advantageous safety profile and the increasing number of non-albicans Candida infections. However, the evidence justifying their use remains rather sparse. Recently published data on break

Antimicrobial resistance is a global health crisis to which pathogenic fungi make a substantial contribution. The human fungal pathogen C. auris is of particular concern due to its rapid spread across the world and its evolution of multidrug resistance. Fluconazole failure in C. auris has been recently attributed to antifungal "tolerance". Tolerance is a phenomenon whereby a slow-growing subpopulation of tolerant cells, which are genetically identical to susceptible cells, emerges during drug treatment. We use microbroth dilution and disk diffusion assays, together with image analysis, to inve

Candida auris has become a major concern in critical care medicine due to the increasing number of immunocompromised patients and candidiasis is the most frequent cause of fungal infections. C. auris and other fungal pathogens are responsible for at least 13 million infections and 1.5 million deaths globally per year. In immunocompromised patients, infections can quickly become severe, causing wound infections, otitis and candidemia, resulting in high morbidity and mortality. The clinical presentation of C. auris is often non-specific and similar to other types of systemic infections; in addit
