Immunoproteomic Analysis of the Excretory-Secretory Proteins from Spirometra mansoni Sparganum.

BACKGROUND: Sparganosis is caused by the invasion of Spirometra sparganum into various tissues/organs. Subcutaneous sparganosis can be diagnosed by biopsy, while visceral/cerebral sparganosis is not easy to be diagnosed. The diagnosis depends largely on the detection of specific anti-sparganum antibodies. The specificity of the ELISA could be increased by using S. mansoni sparganum excretory-secretory (ES) antigens, but it also had the cross-reactions with sera of patients with cysticercosis or paragonimiasis. The aim of this study was to identify early specific diagnostic antigens in S. manso

The transcriptional map of the common eliminated region 1 (C3CER1) in 3p21.3.

Occurrence of chromosome 3p deletions in a large number of human tumours suggests the existence of uncharted tumour suppressor gene(s). We previously applied a functional assay, named the Elimination test (Et), for the identification of regions containing tumour growth antagonising genes. This resulted in the definition of chromosome 3 common eliminated region 1 (C3CER1) on 3p21.3, which is regularly eliminated from SCID-derived tumours. Systematic genomic sequencing of 11 PAC clones, combined with comparisons of genomic sequence against EST databases and PCR-based cloning of cDNA sequences al

BRSK2 accumulation mediated by TRIM28 accelerates hepatocellular carcinoma progression via activating the PI3K/AKT/mTOR signaling pathway.

BR serine/threonine kinase 2 (BRSK2) plays critical roles in neuronal polarization, cell cycle progression, and glucose-stimulated insulin secretion. However, its function in HCC remains unknown. Through bioinformatics analysis of public databases, we identified BRSK2 as a PCD-related gene with potential clinical diagnostic value in HCC. Functionally, BRSK2 knockdown significantly suppressed the proliferation, migration, and invasion of HCC cells and promoted apoptosis in vitro, as well as inhibited tumor growth in a xenograft mouse model in vivo. Mechanistically, we found that BRSK2 is associ

Non-Coding RNA Biomarkers in Prostate Cancer: Evidence Mapping and In Silico Characterization.

Non-coding RNAs (ncRNAs) have emerged as promising biomarkers for prostate cancer (PCa), yet evidence remains dispersed across heterogeneous studies and their regulatory context is seldom analyzed in an integrated manner. This study systematically maps ncRNAs reported as diagnostic biomarkers for PCa and characterizes their molecular interactions through in silico analyses. A comprehensive evidence-mapping strategy across major bibliographic databases identified 693 studies, of which 58 met eligibility criteria. Differentially expressed ncRNAs were extracted and classified by RNA type. Subsequ

Mechanisms associated with temozolomide resistance in U87MG cell line: in silico and in vitro approaches.

IDH-wildtype grade diffuse glioma is the most aggressive primary brain tumor in adults, with limited survival despite standard therapy with temozolomide (TMZ). Resistance to TMZ is driven by DNA repair pathways, tumor heterogeneity, and microenvironmental adaptations, indicating the need for novel therapeutic strategies. Using a systems biology approach, a compound-protein and protein-protein interaction networks was constructed with STRING 12.0 and STITCH 5.0 databases, identifying hub-bottleneck proteins associated with TMZ resistance. Topological and gene ontology analyses revealed 5 functi

Cutaneous α-Synuclein and Age Spots in Neurodegeneration: A Systematic Review and Testable Hypothesis.

Recent studies have identified phosphorylated α-synuclein in the skin of individuals with neurodegenerative disease. The levels of this protein in the skin have been correlated with disease severity. This protein has been specifically studied in alpha-synucleinopathies such as Parkinsons' Disease (PD) and Multiple System Atrophy (MSA). Given that skin biopsy studies have often shown high levels of phosphorylated α-synuclein in the neck area, and that other clinical studies have described easily identifiable changes in facial skin features, this systematic review explores whether age spots, whi

Cullin1 overexpression is associated with colorectal cancer progression and poor prognosis.

Colorectal cancer (CRC) is a leading cause of cancer-related mortality worldwide. Cullin1 (CUL1), a key element of the Skp1-Cullin1-Rbx1-F-box protein ubiquitin E3 ligase complex, is implicated in tumorigenesis. Herein, we aimed to investigate the expression pattern and clinical significance of CUL1 in CRC. Publicly available databases and platforms (TNMplot, UALCAN, the Human Protein Atlas, and Kaplan-Meier survival analysis) were used to explore the mRNA and protein expression levels of CUL1 and to evaluate its prognostic significance. TNMplot and UALCAN confirmed CUL1 mRNA overexpression in

Transcriptomic clustering of chronic lymphocytic leukemia: molecular subtypes based on Bruton's tyrosine kinase expression levels.

Historically, CLL prognostication relied on disease burden, reflected in clinical stage. Later, chromosome abnormalities and genomics suggested several CLL subtypes which were aligned with response to therapy. Gene expression profiling data identified pathways associated with CLL progression. We hypothesized that transcriptome and proteome may identify functional omics associated with CLL nosology. As a test cohort, we utilized publicly available treatment-naïve CLL transcriptomics data (n = 130) and did consensus clustering that identified BTK-expression-based clusters. The BTK-High and BTK-L

Two pronged approach for prevention and therapy of COVID-19 (Sars-CoV-2) by a multi-targeted herbal drug, a component of ayurvedic decoction.

INTRODUCTION: SARS-CoV-2 a new virus of the zoonotic coronavirus family causes the disease COVID-19, which has become a global pandemic. One of the ways for prevention of COVID-19 is by disabling its spike protein which results in inhibiting its binding with angiotensin-converting enzyme 2 (ACE-2). The other alternative is to inhibit its replication once inside the body. The aim of this study was to explore the literature to identify whether there were any Ayurvedic remedies which contained ingredients which demonstrated this dual effect. METHODS: In silico studies were carried out to find th

The evolution of the huntingtin-associated protein 40 (HAP40) in conjunction with huntingtin.

BACKGROUND: The huntingtin-associated protein 40 (HAP40) abundantly interacts with huntingtin (HTT), the protein that is altered in Huntington's disease (HD). Therefore, we analysed the evolution of HAP40 and its interaction with HTT. RESULTS: We found that in amniotes HAP40 is encoded by a single-exon gene, whereas in all other organisms it is expressed from multi-exon genes. HAP40 co-occurs with HTT in unikonts, including filastereans such as Capsaspora owczarzaki and the amoebozoan Dictyostelium discoideum, but both proteins are absent from fungi. Outside unikonts, a few species, such as t

The heparin-binding protein interactome in pancreatic diseases.

BACKGROUND: The cellular microenvironment plays an important role in the regulation of homoeostasis and is a source of potential biomarkers and drug targets. In a genome-wide analysis the extracellular proteins that bind to heparin (HBPs) have been shown to form highly modular and interconnected extracellular protein regulatory networks. Using a systems biology approach, we have investigated the role of HBP networks in the normal pancreas and pancreatic digestive diseases. METHODS: Lists of mRNAs encoding for HBPs associated with the normal pancreas (NP), acute pancreatitis (AP), chronic panc
