Altered functional connectivity strength of primary visual cortex in subjects with thyroid-associated ophthalmopathy.

Our objective was to explore the disparities in the intrinsic functional connectivity (FC) patterns of primary visual cortex (V1) between patients with thyroid-associated ophthalmopathy (TAO) and healthy controls (HCs) utilizing resting-state functional MRI. Twenty-one patients with TAO (14 males and 7 females; mean age: 54.17 ± 4.83 years) and 21 well-matched HCs (14 males and 7 females; mean age: 55.17 ± 5.37 years) underwent functional MRI scans in the resting-state. We assessed modifications in the intrinsic FC patterns of the V1 in TAO patients using the FC method. Subsequently, the ident

Diagnosis and Biomarker Screening of Endometrial Cancer Enabled by a Versatile Exosome Metabolic Fingerprint Platform.

Exosomes have emerged as a revolutionary tool for liquid biopsy (LB), as they carry specific cargo from cells. Profiling the metabolites of exosomes is crucial for cancer diagnosis and biomarker discovery. Herein, we propose a versatile platform for exosomal metabolite assay of endometrial cancer (EC). The platform is based on a nanostructured composite material comprising gold nanoparticle-coated magnetic COF with aptamer modification (Fe3O4@COF@Au-Apt). The unique design and novel synthesis strategy of Fe3O4@COF@Au-Apt provide the material with a large specific surface area, enabling the eff

Optimization assisted framework for thyroid detection and classification: A new ensemble technique.

Ultrasound (US) is an inexpensive and non-invasive technique for capturing the image of the thyroid gland and nearby tissue. The classification and detection of thyroid disorders is still in its infant stage. This study aims to present a new thyroid diagnosis approach, which consists of three phases like "(i) feature extraction, (ii) feature dimensionality reduction, and (iii) classification". Initially, the thyroid images as well as its related data are given as input. From the input image, the features such as" Grey Level Co-occurrence Matrix(GLCM), Grey level Run Length Matrix(GLRM), propos

The differential expression of micro-RNAs 21, 200c, 204, 205, and 211 in benign, dysplastic and malignant melanocytic lesions and critical evaluation of their role as diagnostic biomarkers.

Overlapping histological features between benign and malignant lesions and a lack of firm diagnostic criteria for malignancy result in high rates of inter-observer variation in the diagnosis of melanocytic lesions. We aimed to investigate the differential expression of five miRNAs (21, 200c, 204, 205, and 211) in benign naevi (n = 42), dysplastic naevi (n = 41), melanoma in situ (n = 42), and melanoma (n = 42) and evaluate their potential as diagnostic biomarkers of melanocytic lesions. Real-time PCR showed differential miRNA expression profiles between benign naevi; dysplastic naevi and melan

Problem gambling subtypes based on psychological distress, alcohol abuse and impulsivity.

The notion of comorbidities within problem gambling populations has important clinical implications, particularly for appropriate treatment matching. The comorbidities most commonly cited in problem gambling literature include depression, anxiety, alcohol abuse and impulsivity. Previous research shows evidence of patterns in multiple co-occurring comorbidities and that there may be different subtypes of gamblers based on these patterns. To further the current understanding of gambling subtypes, the aim of our study was to identify subtypes of gamblers currently in treatment. Hierarchical Clust

Machine learning-based integration identifies a 10-gene predictive signature and its classification patterns in schizophrenia.

BACKGROUND: Schizophrenia (SCZ) is a highly heritability psychological disorder, however the exact etiology remains unclear, and lack of the reliable and effective biomarkers for diagnosis and treatments in the management of SCZ, thus exploring the novel biomarkers in SCZ may enhance the efficacy of its predictive, preventive, and personalized medicine (PPPM/3PM) approach. METHODS: Based on differentially expressed genes (DEGs) and weighted gene co-expression network (WGCNA) analyses from five brain datasets, we screened SCZ-key genes and then developed a novel machine learning (ML) framework

Predicting risk of early discontinuation of exclusive breastfeeding at a Brazilian referral hospital for high-risk neonates and infants: a decision-tree analysis.

BACKGROUND: Determinants at several levels may affect breastfeeding practices. Besides the known historical, socio-economic, cultural, and individual factors, other components also pose major challenges to breastfeeding. Predicting existing patterns and identifying modifiable components are important for achieving optimal results as early as possible, especially in the most vulnerable population. The goal of this study was building a tree-based analysis to determine the variables that can predict the pattern of breastfeeding at hospital discharge and at 3 and 6 months of age in a referral cent

A salivary protein panel for detecting malignant transformation of oral leukoplakia.

Oral leukoplakia (OLK) is the most common oral potentially malignant disorder and carries a lifetime risk of transformation to oral squamous cell carcinoma (OSCC) of 0.1% to 9.8%. The aim of the current study is to explore salivary proteomics as a non-invasive approach for detecting the malignant transformation from OLK to OLK-associated OSCC (OLK-OSCC). We performed mass spectrometry-based proteomic profiling of saliva from 131 patients (48 OLK and 83 OLK-OSCC patients) and integrated the resulting data with machine learning. Salivary protein diversity was significantly increased in OLK-OSCC

Targeting HsDHODH: Shape and machine-learning guided discovery and structural validation of SARS-CoV-2 antivirals.

The frequent emergence of novel RNA viruses and the rapid development of viral resistance highlight the urgent need for new antiviral therapies. In this study, we employed an integrated computational pipeline combining shape-based models, machine-learning (ML)-based models, and hotspot analysis to identify novel human dihydroorotate dehydrogenase (HsDHODH) inhibitors. A virtual screening (VS) campaign of the H3D chemical library identified H3D-002856 as the primary hit (IC50 = 2.92 ± 0.07 μM), which subsequently guided a similarity-based hit expansion (Tanimoto coefficient≥0.7) of 17 structura

Spatially Resolved Free Fatty Acid Profiling Reveals Region- and Age-Dependent Remodeling of Alzheimer's Disease Plaques.

Free fatty acids (FFAs) are bioactive mediators of inflammation, energy metabolism, and membrane remodeling, yet their spatial organization within the Alzheimer's disease (AD) brain and at individual amyloid-β (Aβ) plaques has remained inaccessible. We developed a novel, chemically tailored MALDI workflow that enables simultaneous, spatially resolved detection of nearly 30 FFAs alongside over 100 complex lipid species within the same tissue section. Applying this approach to a transgenic AD mouse model across brain regions and disease stages, and combining it with single-plaque microenvironmen

Different oscillatory mechanisms of dementia-related diseases with cognitive impairment in closed-eye state.

The escalating global trend of aging has intensified the focus on health concerns prevalent among the elderly. Notably, Dementia related diseases, including Alzheimer's disease (AD) and frontotemporal dementia (FTD), significantly impair the quality of life for both affected seniors and their caregivers. However, the underlying neural mechanisms of these diseases remain incompletely understood, especially in terms of neural oscillations. In this study, we leveraged an open dataset containing 36 CE, 23 FTD, and 29 healthy controls (HC) to investigate these mechanisms. We accurately and clearly

Discovering the neuronal dynamics in major depressive disorder using Hidden Markov Model.

INTRODUCTION: Major Depressive Disorder (MDD) is a leading cause of worldwide disability, and standard clinical treatments have limitations due to the absence of neurological evidence. Electroencephalography (EEG) monitoring is an effective method for recording neural activities and can provide electroneurophysiological evidence of MDD. METHODS: In this work, we proposed a probabilistic graphical model for neural dynamics decoding on MDD patients and healthy controls (HC), utilizing the Hidden Markov Model with Multivariate Autoregressive observation (HMM-MAR). We testified the model on the M
