Catechol pyrazolinones as trypanocidals: fragment-based design, synthesis, and pharmacological evaluation of nanomolar inhibitors of trypanosomal phosphodiesterase B1.

Trypanosomal phosphodiesterases B1 and B2 (TbrPDEB1 and TbrPDEB2) play an important role in the life cycle of Trypanosoma brucei, the causative parasite of human African trypanosomiasis (HAT), also known as African sleeping sickness. We used homology modeling and docking studies to guide fragment growing into the parasite-specific P-pocket in the enzyme binding site. The resulting catechol pyrazolinones act as potent TbrPDEB1 inhibitors with IC₅₀ values down to 49 nM. The compounds also block parasite proliferation (e.g., VUF13525 (20b): T. brucei rhodesiense IC₅₀ = 60 nM, T. brucei brucei IC₅

Conditioned taste aversion and drugs of abuse: history and interpretation.

Conditioned taste aversion (CTA) learning describes a phenomenon wherein an animal learns to avoid consumption of a particular taste or food following its pairing with an aversive stimulus. Although initially demonstrated with radiation and classical emetics, CTAs have also been shown with drugs of abuse. The ability of rewarding drugs to support CTA learning was described as paradoxical by many investigators, and a number of attempts have been made to resolve this paradox. The present review offers a historical perspective on the CTA literature with a particular focus on CTAs induced by self-

[Influence of positive and negative symptoms on suicidal behaviour in schizophrenia. Review of current literature].

Suicide is the main cause of death among persons with schizophrenia. The risk of suicide among this group is continually high and it is estimated at 10%. The aim of the study was a review of current literature concerning positive and negative symptoms on suicidal behaviour in schizophrenia. Some studies showed, that the active phase of the disease and worsening of the sickness increase the risk of suicide among those diagnosed with schizophrenia. It is contradictory to the works, which prove that suicide of a schizophrenic person is not a result of experiencing psychotic symptoms. According to

Mechanisms of action of acetazolamide in the prophylaxis and treatment of acute mountain sickness.

Acetazolamide, a potent carbonic anhydrase (CA) inhibitor, is the most commonly used and best-studied agent for the amelioration of acute mountain sickness (AMS). The actual mechanisms by which acetazolamide reduces symptoms of AMS, however, remain unclear. Traditionally, acetazolamide's efficacy has been attributed to inhibition of CA in the kidneys, resulting in bicarbonaturia and metabolic acidosis. The result is offsetting hyperventilation-induced respiratory alkalosis and allowance of chemoreceptors to respond more fully to hypoxic stimuli at altitude. Studies performed on both animals an

Clinical utility of the erythrocyte sedimentation rate.

The erythrocyte sedimentation rate (ESR) determination is a commonly performed laboratory test with a time-honored role. However, the usefulness of this test has decreased as new methods of evaluating disease have been developed. The test remains helpful in the specific diagnosis of a few conditions, including temporal arteritis, polymyalgia rheumatica and, possibly, rheumatoid arthritis. It is useful in monitoring these conditions and may predict relapse in patients with Hodgkin's disease. Use of the ESR as a screening test to identify patients who have serious disease is not supported by the

A model of otolith stimulation.

A new model of otolithic stimulation by linear acceleration is presented and compared to previous models, based upon anatomical evidence and on the ability of normal subjects to sense the direction of a linear acceleration vector acting in the coronal plane (roll-tilt perception). There are two basic methods of generating roll-tilt stimuli: 1) tilt-chairs either inside or outside a centrifuge and 2) fixed-chair centrifuges. The present model is based on consideration of the probable otoconial displacement produced by these two different methods of stimulation and the model incorporates a major

From breast cancer patient to teammate: The role of exercise group identity in enhancing well-being.

PURPOSE: Numerous studies have found evidence of the link between physical exercise and improved well-being in breast cancer patients. However, there is little research on physical exercise in a group setting that examines the (re)configuration of social identities and their relationship with well-being among these patients. The study aimed to analyze the configuration of cancer patient identity and the change in its salience when a new social identification as a member of a physical exercise group is incorporated in self-concept, and how this new identity could strengthen the perception of we

"I probably shouldn't go in today": Inequitable access to paid sick leave and its impacts on health behaviors during the emergence of COVID-19 in the Seattle area.

This study examines inequities in access to paid sick leave (PSL) by race/ethnicity, income, and sex and the role of PSL access on leave-taking and care-seeking behaviors among Seattle-area workers in the months leading up to and during the emergence of COVID-19 in the region. Survey responses were collected online and in-person from individuals experiencing acute respiratory illness symptoms between November 2019 and March 2020 as part of a community-based respiratory viral surveillance study. Chi-square tests and log-binomial models were used to assess the association between PSL access and

Vitamin D levels and monospot tests in military personnel with acute pharyngitis: a retrospective chart review.

Some recent studies have proposed an important role for vitamin D in reducing the risk of infection by assisting in the suppression of viruses and by controlling the inflammatory response. A low vitamin D state may have a detrimental effect on the immune system's ability to produce activated CD8+ T cells, and it may increase the inflammatory reaction to Epstein Barr virus. The aim of this chart review was to see if serum 25 OH vitamin D3 levels in service members with acute pharyngitis were lower in those who had positive rather than negative monospot tests. A retrospective chart review was co

Correlates of occupational injuries for various jobs in railway workers: a case-control study.

Few studies have simultaneously addressed the role of occupational factors, individual characteristics and living conditions in occupational injuries, and to the best of our knowledge none on railway workers. This survey assessed the roles of these factors in various types of injuries and for various jobs in French railway workers. This case-control study was conducted on 1,305 male workers with an occupational injury during a one-year period and 1,305 male controls. A standardized questionnaire was administrated by an occupational physician. Data were analyzed by the logistic regression metho

The epigenome of Trypanosoma brucei: a regulatory interface to an unconventional transcriptional machine.

The epigenome represents a major regulatory interface to the eukaryotic genome. Nucleosome positions, histone variants, histone modifications and chromatin associated proteins all play a role in the epigenetic regulation of DNA function. Trypanosomes, an ancient branch of the eukaryotic evolutionary lineage, exhibit some highly unusual transcriptional features, including the arrangement of functionally unrelated genes in large, polymerase II transcribed polycistronic transcription units, often exceeding hundreds of kilobases in size. It is generally believed that transcription initiation plays

Boundary Control and Obtaining Care: Which Workplace Policies Are Associated with Obtaining Preventative Care During Work Hours?

CONTEXT: Because preventative care is often only available during standard business hours, workplace policies and work arrangements may play a central role in enabling people to obtain needed care. Yet it remains unclear which specific policies and practices are most effective in facilitating preventative care use, especially during the workday. This study draws on survey data from workers in Michigan and Arizona, where Michigan and Arizona workers have statutory access to paid sick leave, to examine how policies related to schedule control, work location, paid time off, and weekly work hours

I see sick people: Beliefs about sensory detection of infectious disease are largely consistent across cultures.

Identifying cues to contagious disease is critical for effectively tracking and defending against interpersonal infection threats. People hold lay beliefs about the types of sensory information most relevant for identifying whether others are sick with transmissible illnesses. Are these beliefs universal, or do they vary along cultural and ecological dimensions? Participants in 58 countries (N = 19,217) judged how effective, and how likely they were to use, cues involving each of the five major sensory modalities in an imagined social interaction during a flu outbreak. Belief patterns were str

Biophysical and microbiological aspects of the interaction between Yersinia pestis PsaA and bacteriophage L-413C.

There has been a great interest in developing the phage-containing remedy against plague caused by antimicrobial resistant strains of Yersinia pestis, which have been increasingly isolated in recent years from sick humans and animals. Studies thus are under way to develop a "phage cocktail", which is expected to be effective against a wide range of pathogenic strains. Our paper sheds light on the role of Y. pestis antigen PsaA in reception of the phage L-413C, which might be a possible component of such a "cocktail". Using optical trapping (OT) and atomic force microscopy (AFM), we showed that

Intersex Pretenders.

False claims of having an intersex condition have been observed in print, video, Internet media, and in live presentations. Claims of being intersexed in publicly accessible media were examined and evidence that they were false was considered sufficiently conclusive in 37 cases. Falsity was most often detected due to medical implausibility and/or inconsistency, but sometimes also using information from third-party or published sources. The majority, 26/37, of cases were natal males; 11/37 were natal females. Almost all (34/37) were transgendered, living, or aspiring to live, in their non-natal

Case Report: SCN5A mutations in three young patients with sick sinus syndrome.

BACKGROUND: Sick Sinus Syndrome (SSS) is generally regarded as a degenerative disease with aging; however, genetic mutations have been confirmed to be associated with SSS. Among them, mutations in SCN5A are common in patients with SSS. We report three young SSS patients with SCN5A mutations at different sites that have not been previously reported in Asian patients. CASE PRESENTATION: The three patients were all young females who presented with symptoms of severe bradycardia and paroxysmal atrial flutter, for which two patients received ablation therapy. However, after ablation, Holter monito

A qualitative investigation of the phenomenology of muscle dysmorphia: Part 2.

Muscle dysmorphia (MD) is a mental disorder characterised by preoccupation that one lacks muscularity and/or leanness, leading to substantial functional impairment. This article is the second of a two-part qualitative study examining the phenomenology of MD among 29 adults with diagnosed MD. We used reflexive thematic analysis to generate five themes which capture the role of exercise in the lives of people with MD. In summary, participants were highly motivated by a constant desire to progress their strength and appearance, and bad or missed workouts which threatened to derail their progress

The treatment lottery of chronic back pain? A case series at a multidisciplinary pain centre.

OBJECTIVES: Despite the number of people affected by chronic back pain, and the many available treatment options, even the best modalities provide limited pain reduction on a group level, often without simultaneous improvements in functioning or health-related quality of life. The objective was to provide an overview of the treatment of chronic back pain in clinical practice at a multidisciplinary pain centre, and to study patient and pain characteristics in different treatment groups. METHODS: 104 chronic back pain patients (primary ICD-10-SE-diagnosis M53.0-M54.9 excluding M54.1 and M54.3),

Osteoarthritis: Novel Molecular Mechanisms Increase Our Understanding of the Disease Pathology.

Although osteoarthritis (OA) is the most common musculoskeletal condition that causes significant health and social problems worldwide, its exact etiology is still unclear. With an aging and increasingly obese population, OA is becoming even more prevalent than in previous decades. Up to 35% of the world's population over 60 years of age suffers from symptomatic (painful, disabling) OA. The disease poses a tremendous economic burden on the health-care system and society for diagnosis, treatment, sick leave, rehabilitation, and early retirement. Most patients also experience sleep disturbances,
