Small-caliber vascular grafts (< or =5 mm) constructed from synthetic materials for coronary bypass or peripheral vascular repair below the knee have poor patency rates, while autologous vessels may not be available for harvesting. The present study aimed to create a completely autologous small-caliber vascular graft by utilizing a bioabsorbable, macroporous poly(L/D)lactide 96/4 [P(L/D)LA 96/4] mesh as a support scaffold system combined with an autologous fibrin cell carrier material. A novel molding device was used to integrate a P(L/D)LA 96/4 mesh in the wall of a fibrin-based vascular graf

Current treatments of osteoarthritis (OA) focus on pain and loss of joint function. When these interventions fail, the destroyed joint is replaced by implants of metal, plastic and ceramics. In the future, we need to detect cartilage loss before it is too severe, prevent further loss and stimulate regrowth of lost cartilage. Research in tissue engineering can help us understand the complex requirements for regeneration of joint cartilage. Results from animal experiments and small, uncontrolled, open series of human cartilage repair suggest that functional repair can be accomplished in some joi…

In urethra reconstruction, the creation of a new urethra from a free oral mucosa graft is an established surgical technique. The oral mucosa is removed at the same time that the urethra reconstruction procedure is performed. Depending on the size of graft required, the intraoral wound is closed primarily or left to heal secondarily. The latter method limits this technique by leading to scars or strictures, which have a negative impact on the condition of the intraoral soft tissue. Therefore, in this study, a pilot study involving 12 patients, tissue-engineered mucosa was tested for covering in

Immortalized human epidermal keratinocytes, including HaCaT, N/TERT-1, and N/TERT-2G keratinocytes, are increasingly being used as alternatives to primary human keratinocytes to produce epidermal and full-thickness skin equivalents. The use of immortalized cell lines requires a critical evaluation of their genotypes and functional characteristics to establish their suitability to model the epidermis in vitro. Here, we assess the accumulating evidence-based experimental data on HaCaT, N/TERT-1, and N/TERT-2G keratinocytes, as well as additional immortalized keratinocyte cell lines, to aid the s

Microgravity provides a unique model for accelerated skeletal muscle loss and potentially muscle ageing. During spaceflight, astronauts experience pronounced muscle atrophy, similar to age-related decline on Earth but over a much shorter timescale. Despite daily aerobic and resistance exercise on the International Space Station (ISS), countermeasures remain suboptimal, reflecting similar challenges seen in ageing populations. The MicroAge Mission used microgravity on the ISS to assess whether the molecular mechanisms behind reduced adaptive responses to contractile activity during ageing resem

Human salivary stem/progenitor cell (hS/PC)-loaded hyaluronic acid (HA)-based hydrogels, termed 3D-salivary tissue constructs (3D-ST), hold great promise for restoring salivary gland function post-radiation injury. Here, we developed a next-generation 3D-ST using heparin-modified HA and bioactive peptide-modified hydrogels. This new formulation enables controlled pre-loading and localized presentation of heparin-binding growth factors prior to surgical implantation, providing opportunities to enhance in vivo hS/PC bioactivity. To model clinically relevant radiation injury, we established an at

The ability to a-priori identify antigenic components responsible for driving recipient graft-specific adaptive immune destruction of tissue-engineered biomaterials is critical for their safe clinical translation and long-term function. However, despite our in-depth understanding of cellular responses toward antigenic stimuli, the question of whether individual antigens are equally stimulatory to the immune system remains largely unanswered. This lack of knowledge limits our capacity to rationally design immunologically acceptable biomaterials. Previously published decellularization/antigen re

Fully reduced high-mobility group box 1 (HMGB1) binds CXCL12 and signals via CXCR4 when released into the extracellular space. It acts as a chemokine and transitions stem cells from quiescent G0 to a primed GAlert state. Cells in GAlert rapidly enter G1 in response to activating factors released by tissue injury to promote tissue repair. However, oxidative conversion of FR-HMGB1 into the disulfide form activates proinflammatory pathways via TLR-2, TLR-4 and RAGE. Peptide mapping and nuclear magnetic resonance (NMR) spectroscopy identified a conserved CXCL12-binding motif within each Box and ad

Tissue-engineered tendon constructs that replicate the structural and functional properties of natural tendons are crucial in regenerative medicine to improve treatment outcomes after tendon injury. This study aimed to engineer 3-dimensional biomimetic tendon macro-tissues through bioassembly of cell spheroids. Rat tendon fibroblasts seeded at different cell numbers (1 × 104, 5 × 104, 1 × 105, 2 × 105, 3 × 105) were analysed for spheroid formation and development for 28 d. The spheroid diameters decreased over time with a reduction in cell density while synthesising their own collagen fibres.

Repairing infectious tissues, particularly bone and wound infections, poses significant challenges to clinicians due to the lack of effective treatment modalities. Sonodynamic therapy, which utilizes sonosensitizers to generate reactive oxygen species (ROS) under ultrasound (US) energy for antibacterial purposes, still lacks suitable sonosensitizers with good biocompatibility that can simultaneously promote the repair of various infected tissues. In this study, we constructed a novel sonosensitizer composed of hemoglobin derived from nature and zeolitic imidazolate framework (ZIF)-8, which exh

Background/Objectives: Periodontal diseases are highly prevalent worldwide, causing progressive destruction of the alveolar bone and eventual tooth loss when not treated. Despite advances in conventional periodontal therapies, complete tissue regeneration remains limited. This review aims to evaluate the efficacy, safety, and clinical relevance of stem cell-based tissue engineering approaches for regeneration of periodontal bone lesions. Methods: Following PRISMA guidelines, a systematic search was conducted across multiple databases, resulting in the inclusion of 17 studies in humans that met…

Diabetic cardiomyopathy (DbCM) is increasingly prevalent, but intervention targets remain unclear due to the lack of appropriate models and the complexity of risk factors. Here, this work establishes an in vitro assessment system for DbCM function using cardiomyocytes derived from human pluripotent stem cells and engineered heart tissue. This work finds high-fat status in complex diabetes risk factors majorly contributes most to cardiomyocyte death and contractile dysfunction. Notably, PA induced early electrophysiological abnormalities, and lately is associated with cardiac fibrosis, mitochon
