Transdermal neuromodulation of noradrenergic activity suppresses psychophysiological and biochemical stress responses in humans.

We engineered a transdermal neuromodulation approach that targets peripheral (cranial and spinal) nerves and utilizes their afferent pathways as signaling conduits to influence brain function. We investigated the effects of this transdermal electrical neurosignaling (TEN) method on sympathetic physiology under different experimental conditions. The TEN method involved delivering high-frequency pulsed electrical currents to ophthalmic and maxillary divisions of the right trigeminal nerve and cervical spinal nerve afferents. Under resting conditions, TEN significantly suppressed basal sympatheti…

Sensorimotor postural rearrangement after unilateral vestibular deafferentation in patients with acoustic neuroma.

Unilateral vestibular lesion, as acoustic neuroma and its surgical removal, leads to impaired balance control. After initial vertigo and postural instability corresponding to unilateral vestibular deafferentation, improvement in symptoms and global balance functions occurs by a process called vestibular compensation. In this respect, this prospective study aimed to assess the differential contribution of sensory inputs to the regulation of posture during the recovery process after acoustic neuroma removal. Twenty-seven patients with acoustic neuroma underwent vestibular and posturographic test

Pupil cycle induction test: a way of evaluating the pupillary light reflex.

The Pupil Cycle Induction Test (PCIT) described here is a simple and sensitive clinical method for the evaluation of afferent pupillary defect. It assesses the difficulty in setting up regular and sustained light-induced pupil oscillations, in contrast to methods assessing the light reflex by timing the oscillations. PCIT was applied to 186 presumably normal eyes (with normal visual acuity and normal fundi). In ten eyes, sustained oscillations could not be induced; further investigation showed that four of these eyes had altered visual-evoked response. The test was also carried out in 34 patie

Cardiovascular Control During Exercise: The Connectivity of Skeletal Muscle Afferents to the Brain.

The exercise pressor reflex (EPR) is engaged upon the activation of group III/IV skeletal muscle afferents and is one of the principal mediators of cardiovascular responses to exercise. This review explores the hypothesis that afferent signals from EPR communicate via GABAergic contacts within the brain stem to evoke parasympathetic withdrawal and sympathoexcitation to increase cardiac output, peripheral resistance, and blood pressure during exercise.

A new model of strabismic amblyopia: Loss of spatial acuity due to increased temporal dispersion of geniculate X-cell afferents on to cortical neurons.

Although the neural locus of strabismic amblyopia has been shown to lie at the first site of binocular integration, first in cat and then in primate, an adequate mechanism is still lacking. Here we hypothesise that increased temporal dispersion of LGN X-cell afferents driven by the deviating eye onto single cortical neurons may provide a neural mechanism for strabismic amblyopia. This idea was investigated via single cell extracellular recordings of 93 X and 50 Y type LGN neurons from strabismic and normal cats. Both X and Y neurons driven by the non-deviating eye showed shorter latencies than

Acid sensitization of esophageal mucosal afferents: implication for symptom perception in patients across the gastroesophageal reflux disease spectrum.

BACKGROUND: Sensitization of esophageal chemoreceptors, either directly by intermittent acid exposure or indirectly through esophagitis-associated inflammatory mediators, is likely to be the mechanism underlying the perception of heartburn. AIMS: To compare basal esophageal sensitivity with electrical stimulation and acid, and to compare the degree of acid-induced sensitization in controls and in patient groups across the entire spectrum of gastroesophageal reflux disease: erosive oesophagitis (EO), nonerosive reflux disease (NERD), and functional heartburn (FH). METHODS: Esophageal sensory

Role of sensory nerves in the rapid cutaneous vasodilator response to local heating in young and older endurance-trained and untrained men.

The ability to increase skin blood flow (SkBF) rapidly in response to local heating is diminished with advanced age; however, the mechanisms are unclear. The primary aim of this study was to investigate the role of sensory nerves in this age-related change. A secondary aim was to investigate the effect of aerobic fitness on sensory nerve-mediated vasodilatation in young and aged skin. We measured SkBF (using laser Doppler flowmetry) in young and older endurance-trained and untrained men (n= 7 in each group) at baseline and during 35 min of local skin heating to 42°C at two sites on the ventral

Antimuscarinics and overactive bladder: other mechanism of action.

Antimuscarinics are considered first-line treatment for patients with overactive bladder (OAB). However, the mechanism by which antimuscarinics improve the symptoms of OAB remains to be elucidated. Animal studies suggest that antimuscarinics may exert an inhibitory effect on afferent nerves without an effect on detrusor contraction. A release of acetylcholine from the urothelium has been demonstrated in isolated human bladder. In addition, muscarinic receptors (MRs) were found in the urothelium and suburothelial myofibroblasts, suggesting a role for MR mechanisms in urothelial sensory function

Neural noise distorts perceived motion: the special case of the freezing illusion and the Pavard and Berthoz effect.

When a slowly moving pattern is presented on a monitor which itself is moved, the pattern appears to freeze on the screen (Mesland and Wertheim in Vis Res 36(20):3325-3328, 1996) even if we move our head with the monitor, as with a head mounted display (Pavard and Berthoz in Perception 6:529-540, 1977). We present a simple model of these phenomena, which states that the perceived relative velocity between two stimuli (the pattern and the moving monitor) is proportional to the difference between the perceived velocities of these stimuli in space, minus a noise factor. The latter reflects the in

Epinephrine potentiates the analgesic and antidepressant effects of polyvinylpyrrolidone and cholecystokinin due to stimulation of afferents in the gastric mucosa.

Treatment with epinephrine, polyvinylpyrrolidone, and cholecystokinin in the minimum effective doses produced maximum analgesic and antidepressant effects and caused bradycardia in rats. Administration of epinephrine in combination with polyvinylpyrrolidone or cholecystokinin in threshold doses (1/10-1/25 of the minimum effective dose) produced maximum analgesic and antidepressant effects, but did not cause bradycardia. The potentiating effect of epinephrine is related to stimulation of afferents in the gastric mucosa.

Cardiovascular responses to vibrostimulation for sperm retrieval in men with spinal cord injury.

BACKGROUND/OBJECTIVE: Cardiovascular abnormalities and arrhythmias are common in individuals with spinal cord injury (SCI) who are undergoing vibrostimulation for sperm retrieval. The study aimed to examine cardiovascular control in men with SCI undergoing this procedure. METHODS: Individuals with chronic cervical (n=8; age: 33.1 +/- 1.9 years) and upper thoracic SCI (n=5; age: 35.2 +/- 2.9 years) volunteered for vibrostimulation, with continuous blood pressure (Finometer) and electrocardiographic monitoring. Patients were characterized further by sympathetic skin responses (SSR) to assess de

Somatosensory evoked magnetic fields to air-puff stimulation on the soft palate.

Impairment of sensory input to the soft palate has been reported in patients with obstructive sleep apnea syndrome. To investigate the reaction in the central nervous system related to soft palate perception, we measured the somatosensory evoked magnetic fields following air-puff stimulation in seven healthy volunteers by using a helmet-shaped 122-channel neuromagnetometer. The air-puffs were produced using compressed nitrogen and directed to the middle of the soft palate with an intraoral device. To evaluate the laterality of responses we used another appliance in which the air-puffs were dir

Activity-dependent slowing of conduction velocity in uninjured L4 C fibers increases after an L5 spinal nerve injury in the rat.

Growing evidence suggests that uninjured afferents may play an important role in neuropathic pain following nerve injury. The excitability of nociceptive neurons in the L4 spinal nerve appears to be enhanced following an injury to the adjacent L5 spinal nerve. In this study, we investigated whether the action-potential conduction properties of unlesioned, unmyelinated fibers are also altered. A teased-fiber technique was used to record from single C fibers from the L4 spinal nerve of the rat in vitro. Repeated electrical stimulation of the tibial nerve was used to investigate activity-dependen

Abnormal vibration induced illusion of movement in essential tremor: evidence for abnormal muscle spindle afferent function.

OBJECTIVES: Vibration induced illusion of movement (VIIM) is abnormal in patients with idiopathic focal dystonia, an abnormality which corrects with fatigue of the vibrated muscle. Since dystonia and essential tremor sometimes coexist in families, we investigated the perception of VIIM and the effect of fatigue on VIIM in patients with essential tremor. METHODS: VIIM in 18 patients with essential tremor was compared with VIIM in 18 healthy control participants before and after volitional fatigue of the vibrated muscles. RESULTS: Vibration of the immobilised biceps produced a subnormal VIIM i

Fos induction in the amygdala by vestibular information during hypergravity stimulation.

Altered gravity environments including both hypo- and hypergravity can elicit motion sickness. Vestibular information is known to be essential for motion sickness, but its other neural substrates are poorly understood. We previously showed that bilateral lesions of the amygdala suppressed hypergravity-induced motion sickness in rats, using pica behavior as an emetic index. We show in the present study that during hypergravity stimulation, vestibular information activated the central nucleus of the amygdala (CeA), as determined by the induction of Fos expression, in comparison between normal an

Immunoneutralization of nerve growth factor in lumbosacral spinal cord reduces bladder hyperreflexia in spinal cord injured rats.

PURPOSE: We investigated the effects of intrathecal application of nerve growth factor (NGF) antibodies (Ab) on bladder hyperreflexia in chr … der afferent neurons, and also suppressed bladder hyperreflexia. Thus, suppression of NGF levels in afferent pathways could be useful for treating bladder hyperreflexia associated with spinal cord injury.

Changes in neuropeptide expression in the trigeminal ganglion following inferior alveolar nerve section in the ferret.

Changes in neuropeptide expression in afferent nerve fibres may play a role in the persistent sensory abnormalities that can be experienced following trigeminal nerve injuries. We have therefore studied changes in the expression of the neuropeptides substance P, calcitonin gene-related peptide, enkephalin, galanin, neuropeptide Y and vasoactive intestinal polypeptide in the trigeminal ganglion following peripheral nerve injury. In anaesthetised adult female ferrets, the left inferior alveolar nerve was sectioned and recovery allowed for three days, three weeks or 12 weeks prior to perfusion-fi

Identification of intestinofugal neurons projecting to the coeliac and superior mesenteric ganglia in the rat.

Intestinofugal neurons are parts of the afferent limbs of inhibitory intestino-intestinal reflexes. These neurons have been mapped in guinea-pigs, where they have a gradient of increasing frequency of occurrence from oral to anal, but not in other species. In the present work in the rat, a species that is more amenable to physiological study than the guinea-pig, we have used retrograde tracing to map the distribution of the cell bodies of intestinofugal neurons projecting to the coeliac-superior mesenteric ganglion complex. Labelled nerve cells were found in the myenteric, but not the submucos

CGRP activates renal pelvic substance P receptors by retarding substance P metabolism.

Substance P and calcitonin gene-related peptide (CGRP) are colocalized in renal pelvic sensory nerves. Increasing renal pelvic pressure results in an increase in afferent renal nerve activity that is blocked by a substance P receptor antagonist but not by a CGRP receptor antagonist. CGRP potentiates the effects of substance P by preventing the metabolism of substance P. Therefore, we examined whether CGRP enhanced the afferent renal nerve activity responses to substance P and increased renal pelvic pressure, a stimulus known to increase substance P release. Combined administration of substance

Neural response of mechanoreceptors to acute inflammation in the rotator cuff of the shoulder joint in rabbits.

We examined with electrophysiological techniques the effects of experimentally induced inflammation on the mechanosensitive afferent units in the rotator cuff of the shoulder joint of 21 rabbits. We identified 21 mechanosensitive units belonging to group III. 12 units had mechanical thresholds of > 7.0 g and 9 units had thresholds of < 7.0 g. After injection of inflammatory agents, kaolin and carrageenan, into the joint space, ongoing afferent discharge rates increased in all units. The average discharge rate increased significantly from 7 imp/s to 15 imp/s after injection. 5 units had a decre
