Injury-resistant retinal ganglion cells that are rich in cytochrome oxidase.

Previous studies have demonstrated that the vast majority of retinal ganglion cells (RGCs) die one month after optic nerve transection. However, a small percentage do not degenerate. The present study examined one aspect of the chemical nature of these surviving RGCs using cytochrome oxidase (CO) as a neuronal marker in whole-mounted retinae. In the normal retina, 4.3% of the total population of RGCs show high CO activity. One month after optic nerve transection, 37% of the CO-positive RGCs counted in the control retinae survive and, because they stain with CO, must be metabolically active. Pr

Anticalmodulin drugs block the sodium gating current of squid giant axons.

The effects of calmodulin (CaM) antagonists (W-7, W-5, trifluoperazine, chlorpromazine, quinacrine, diazepam, propericyazine and carmidazolium) on the sodium and potassium channels were studied on the intracellularly perfused and voltage-clamped giant axon of the squid. It was found that the drugs are more potent blockers of the sodium current than of the potassium current. The drugs also reduce the sodium gating current. The blockage of the sodium and gating current can be explained by assuming that the drugs interact with the sodium gating subunit in one of its closed states. The site of act

A light microscopic investigation of the afferent connections of the lateral reticular nucleus in the cat.

The topographical organization of the projections to the lateral reticular nucleus (LRN) of the cat was investigated using the horseradish peroxidase (HRP), silver-impregnation and autoradiographic tracing methods. Following injection of HRP into the LRN, labelled cells were found mainly within Rexed's laminae VII and VIII of the spinal cord, the contralateral red nucleus, the ventro-rostral aspect of the contralateral fastigial nucleus and the contralateral anterior sigmoid and coronal gyri of the cerebral cortex. Animals with injections of tritiated amino acids placed within the pericruciate

Partial purification and characterization of (Na+ + K+)-ATPase from garfish olfactory nerve axon plasma membrane.

The (Na+ + K+)-ATPase of garfish olfactory nerve axon plasma membrane was purified about sixfold by treatment of the membrane with sodium dodecyl sulfate followed by sucrose density gradient centrifugation. The estimated molecular weights of the two major polypeptide components of the enzyme preparation on sodium dodecyl sulfate gels were 110,000 and 42,000 daltons, which were different from those of the corresponding peptides of rabbit kidney (Na+ + K+)-ATPase. No carbohydrate was detected in the 42,000-dalton component either by the periodic acid-Schiff reagent or by the more sensitive conca

Reversible effects of olfactory nerve section on behavior and biochemistry in mice.

Coincident with the sectioning of the primary olfactory nerves in mice there is a dramatic loss of both olfactory-mediated behavior and the ability of the primary olfactory neurons to synthesize and transport the neuron specific marker, carnosine. These deficiencies are only temporary, and the ability of food deprived mice to locate buried food pellets returns within 21 days after olfactory nerve section. The ability of the primary olfactory neurons to synthesize and transport carnosine appears to increase between 8 days and 45 days after surgery. These data support the notion of behaviorally

Biodistribution of adeno-associated virus type 2 with mutations in the capsid that contribute to heparan sulfate proteoglycan binding.

We compared the phenotypes of three mutant AAV2 viruses containing mutations in arginine amino acids (R585, R588 and R484) previously shown to be involved in AAV2 heparan sulfate binding. The transduction efficiencies of wild type and mutant viruses were determined in the eye, the brain and peripheral organs following subretinal, striatal and intravenous injection, respectively, in mice and rats. We found that each of the three mutants (the single mutant R585A; the double mutant R585, 588A; and the triple mutant R585, 588, 484A) had a unique phenotype compared to wt and each other. R585A was c

Does intranasal application of zinc sulfate produce anosmia in the rat?

Transport of wheat germ agglutinin-horseradish peroxidase (WGA-HRP) from olfactory sensory neurons to the olfactory bulb as well as odor detection and discrimination were examined in rats in which each nasal epithelium had been irrigated with 0.1-0.5 ml 5% zinc sulfate. After treatment, rats showed few or no deficits in discriminating among odors and in detecting high (1%-0.01%) concentrations of ethyl acetate, but some had deficits in detecting lower concentrations of the odor. In most cases, HRP reaction product filled more than 30% of olfactory bulb glomeruli 2-4 days after treatment with Z

Fast blue and diamidino yellow as retrograde tracers in peripheral nerves: efficacy of combined nerve injection and capsule application to transected nerves in the adult rat.

Capsule application of Diamidino Yellow (DY) to the cut end of the sciatic nerve immediately followed by capsule application of Fast Blue (FB) resulted in approximately 95% double-labelled dorsal root ganglion neurones (DRGn) and motoneurones (Mn). Nerve injection of DY followed either immediately or 2 months later by capsule application of FB resulted in approximately 90% double-labelled DRGn and Mn, indicating that DY and FB label similar populations of DRGn and Mn, and that insignificant DY fading occurred during this period. Inversing the order of application, however, i.e. nerve injection

Formation of the corticopontine projection in a long-term culture system.

The reestablishment in vitro of the corticopontine projection was studied in organotypic co-cultures of cortex and pons of rats 0 (day of birth) - 3 days old. After 2-3 weeks in vitro, application of the lipophilic tracer DiI in the pontine explant retrogradely stained in layer V of the cortical explant pyramidal neurons which were characterized by large somata and spiny dendrites with an apical dendrite that reached upper cortical layers II/III. The projection developed only in co-cultures from rats 0-2 days old when the pontine explant was placed in close vicinity either to the white matter

Congenital insensitivity to pain and anhidrosis with mitochondrial and axonal abnormalities.

Hereditary sensory and autonomic neuropathy type IV, or congenital insensitivity to pain with anhidrosis (CIPA), is a rare clinical disorder with only 32 cases reported in the literature. There has been no consistent pathophysiologic defect of the sensory nerve detected by light microscopic examination, but a frequent finding of decreased small myelinated fibers and a uniform finding of decreased unmyelinated fibers by ultrastructural analysis has been reported. Muscle biopsy in a 2-year-old boy with congenital insensitivity to pain with anhidrosis indicated lipid droplet accumulation and redu

Input-output organization of the sensorimotor striatum in the squirrel monkey.

The basal ganglia receive massive inputs from the neocortex and send outputs that exert both inhibitory and disinhibitory control over parts of the frontal cortex and brainstem. Between these basal ganglia inputs and outputs lies the striatum, which receives most of the cortical afferents and projects to the basal ganglia output nuclei--the globus pallidus and substantia nigra. To analyze this system we conjointly labeled, in squirrel monkeys, sensorimotor cortical inputs to the striatum and striatal outputs to the globus pallidus. Anterograde tracers were injected into the motor (MI) and soma

Distribution of enteric nerve cells projecting to the superior and inferior mesenteric ganglia of the guinea-pig.

Retrograde tracing, using Fast Blue dye, was employed to determine the distribution of enteric nerve cells that project to the superior mesenteric and inferior mesenteric ganglia of the guinea-pig. Retrogradely labelled neurons were found in the myenteric but not submucous ganglia. When the superior mesenteric ganglion was injected, labelled neurons were found in low frequencies (less than 5 nerve cell bodies/cm2) in the duodenum, jejunum, ileum, caecum and proximal colon. The distal colon was analysed in five segments of equal length (1-5; oral to anal). Segment 1 had about 4 labelled nerve c

Topographic organization of corticospinal projections from the frontal lobe: motor areas on the lateral surface of the hemisphere.

We examined the topographic organization of corticospinal neurons in the primary motor cortex and in the two premotor areas on the lateral surface of the hemisphere [i.e., the dorsal premotor area (PMd) and the ventral premotor area (PMv)]. In two macaques, we labeled corticospinal neurons that project beyond T7 or S2 by placing crystals of HRP into the dorsolateral funiculus at these segmental levels. In another seven macaques, we labeled corticospinal neurons that project to specific segmental levels of the spinal cord by injecting the fluorescent tracers fast blue and diamidino yellow into

Efferent connections of the hypothalamic "grooming area" in the rat.

The efferent connections of the hypothalamic area, where grooming can be elicited by local electrical stimulation or injection of various substances, were studied using iontophoretic injections of Phaseolus vulgaris leucoagglutinin. This hypothalamic "grooming area" consists of parts of the hypothalamic paraventricular nucleus and of the dorsal hypothalamic area. The specificity of these efferents for the hypothalamic "grooming area" was investigated by comparison with efferents of hypothalamic sites adjacent to this area. In addition, the distribution of oxytocinergic fibres was studied, sinc

Cholinergic innervation of the cerebellum of the rat by secondary vestibular afferents.

The cholinergic innervation of the cerebellar cortex of the rat was studied by immunohistochemical localization of choline acetyltransferase, radiochemical measurement of ChAT activity, and double labeling of ChAT-positive neurons with HRP injected into the cerebellum. ChAT immunohistochemistry revealed large mossy fiber rosettes as well as finely beaded terminals with different morphological characterization, laminar distribution within the cerebellar cortex, and regional differences within the cerebellum. Large "grapelike" ChAT-positive mossy fiber rosettes that were distributed primarily in

Peripheral and central aspects of the acoustic and lateral line system of a bottom dwelling catfish, Ancistrus sp.

The topographical relationship between the swim bladder, the inner ear, and the otic lateral line was studied in the bottom dwelling catfish, Ancistrus sp. In addition, afferent and efferent subcomponents of the eighth and lateral line nerves were labelled with horseradish peroxidase (HRP) or with differently fluorescing dextran amines. The swim bladder of Ancistrus consists of two separate, transversely oriented parts of each of which is connected to the sinus impar of the inner ears via two Weberian ossicles and the perilymphatic sac. The osseous capsula of the ear has two foramina other tha

Ectopic excitability of injured nerves in monkey: entrained responses to vibratory stimuli.

1. The responses to mechanical stimulation of myelinated fibers that originate from an acutely cut nerve or a neuroma were studied in the anesthetized monkey. The superficial radial or sural nerve was tightly ligated and cut. Either immediately (acute experiment) or 2-6 wk later (chronic experiment), single-unit recording techniques were used to record the evoked neural activity after vibratory mechanical stimulation (5-100 Hz; 50-800 microns) near the injury site. 2. The 30 myelinated afferents studied in the chronic experiments displayed an entrained response (1 action potential for each sti

[Partial vertical nucleotomy. A neurosurgical intervention in some forms of therapy-resistant facial neuralgia].

The partial vertical nucleotomy is presented as a new neurosurgical method for the treatment of therapy-resistant pain in the spreading area of the N. trigeminus. Strict indications are laid down which are mainly restricted to therapy-resistant pain in the above mentioned spreading area in the presence of diffusely growing malignant tumours, after traumas and infections and predominantly in forms of idiopathic trigeminal neuralgia that has been treated without any results over a period of many years. In the experimental part of the article; material and method as well as results of anterograde

Trigeminal projections to supratentorial pial and dural blood vessels in cats demonstrated by horseradish peroxidase histochemistry.

Anatomical and clinical observations suggest that supratentorial vascular structures contain afferent projections from the trigeminal ganglia. To characterize this innervation, horseradish peroxidase (HRP) and HRP conjugated to wheat germ agglutinin were applied to the pial and dural arteries and sinuses of 33 cats. HRP was restricted to the site of interest by applying it dissolved in a viscous polymer, polyvinyl alcohol (PVA), to achieve slow release and minimize diffusion. The ganglia of cranial nerves V, VII, IX, and X and the superior cervical ganglia (SCGs) were examined bilaterally for
