Tryptic digestion of spectrin in variants of hereditary elliptocytosis.

Spectrin, either in the form of unfractionated low ionic strength extracts of erythrocyte membranes or purified by chromatography on Sepharose (CL)4B, was subjected to tryptic digestion at 0 degrees C. Four patients, each with a different variant of hereditary elliptocytosis, were studied. In one patient, whose erythrocytes showed significant fragmentation on heating on 45 degrees C, such preparations generated a remarkably different pattern of polypeptide fragments on tryptic digestion at low ionic strength. In this patient 32P was released at a slower rate on tryptic digestion of labeled ban

Isolation and characterization of murine lactoferrin.

Lactoferrin, a non-heme, iron-binding glycoprotein, was isolated from mouse milk. The purification procedure involved an initial centrifugation to remove much of the casein and to separate the fat from the whey, followed by ammonium sulfate precipitation and chromatography on carboxymethyl-Sephadex. The lactoferrin preparation obtained was highly purified as judged by polyacrylamide gel electrophoresis under both non-denaturing and denaturing conditions, and the presence of lysine as the only NH2-terminal amino acid. Polyacrylamide gel electrophoresis in the presence of sodium dodecyl sulfate

Effects of low and high protein diets on the induction of microsomal drug-metabolizing enzymes in rat liver.

To determine the effect of low and high levels of dietary proteins plus low doses of phenobarbital (PB) on the induction of drug-metabolizing enzymes, weanling male and female Sprague-Dawley rats were fed, ad libitum, diets containing 3.5 (low protein, LP), 26 (normal protein, NP), and 42% (high protein, HP) casein for 33 days. Five animals from each dietary group were injected with 10 mg of PB/kg body weight, intraperitoneally, for the last 3 days of the experimental period. The 10,000 X g supernatant and microsomes were prepared from perfused livers from each group. Both LP and HP diets caus

An Antibacterial and Toughening Strategy Based on Carbonaceous Nano-Lamellae Applying for Food Preservation.

Integrating walnut green husk-derived carbonaceous nano-lamellae (WCNL) into casein films (CF) addresses the challenge of combining mechanical strength, barrier performance, and antibacterial activity in sustainable food packaging. The hydrothermally synthesized WCNL features defect-rich hierarchical networks that induces strong interfacial hydrogen bonding, creating a nano-confined network, which dramatically improves mechanical strength (563% increase in tensile strength) and reduces water vapor permeability by 22.77% of CF-WCNL, while offering excellent UV shielding and antioxidant activity

Kinetic fingerprinting and food matrix interactions reveal the gastrointestinal behavior of polyphenolics in stir-fried green tea.

Stir-fried green tea polyphenols exhibit strong antioxidant capacity; however, they degrade rapidly during gastrointestinal transit. This study combined antioxidant kinetic profiling, chemometric analysis, simulated digestion, and Caco-2 cell permeability models to identify the principal antioxidant drivers and evaluate the protective effects of the food matrix. Ridge model identified EGCG as the primary determinant of radical-scavenging kinetics, whereas gallic acid contributed more strongly to the thermodynamic antioxidant capacity. Simulated intestinal digestion induced an antioxidant kinet

Computational screening of natural inhibitors against Plasmodium falciparum kinases: Toward novel antimalarial therapies.

An important worldwide problem is the resistance of Plasmodium falciparum to practically all antimalarial medications. Therefore, new treatment approaches are urgently needed. The development of antimalarial medications frequently involves two important therapeutic targets: casein kinase 2 (CK2) and cGMP-dependent protein kinase (PKG). To identify naturally occurring chemicals that could be used as antimalarial medications to combat multidrug-resistant P. falciparum, we used a multi-targeted in silico strategy in this study. The top 20 compounds, including the reference drug RY-1-65, were sele

Protein-rich food matrices from alternative and conventional sources differentially shape gut microbiota, metabolic function, and muscle protein synthesis.

The functional consequences of consuming conventional and alternative protein rich food matrices remain poorly understood. This study characterized diverse protein rich food matrices and investigated their effect on gut microbiota, muscle protein synthesis, and host metabolic function in rats. Arthropods and lentils contained chitin, resistant starch, polyphenols, and antioxidant activity, whereas egg and picanha were enriched in cholesterol and precursors of trimethylamine N-oxide (TMAO). Lentils and crickets consumption resulted in the lowest fat mass gain, glucose area under the curve and s

Sperm-specific fertility factors SMZ-1/2 promote FB-MO biogenesis and MSP filament assembly in C. elegans.

In nematode sperm, motility is powered by polymerization of major sperm protein (MSP) into filaments assembled from a specialized organelle (FB-MO), but how filament formation begins remains unclear. We identify two sperm-specific PDZ proteins, SMZ-1 and SMZ-2 (SMZ-1/2), that initiate MSP loading into pre-formed membranous organelles in Caenorhabditis elegans. SMZ-1/2 were expressed specifically in spermatogenic germ cells, colocalized with developing FB-MOs from diplotene through spermatid formation, and were redundant for fertility. Loss of SMZ-1/2 abolished MSP filaments, left MOs lacking M

Mutant KRAS and CK2 Cooperatively Stimulate SLC16A3 Activity to Drive Intrahepatic Cholangiocarcinoma Progression.

Intrahepatic cholangiocarcinoma (iCCA) is a lethal malignancy affecting the liver and biliary system. Enhanced understanding of the pathogenic mechanisms underlying iCCA tumorigenesis and the discovery of appropriate therapeutic targets are imperative to improve patient outcomes. In this study, we investigated the functions and regulations of solute carrier family 16 member 3 (SLC16A3), which has been reported to be a biomarker of poor prognosis in iCCA. High SLC16A3 expression was enriched in KRAS viral oncogene homolog-mutated iCCA tumors, and mutant KRAS elevated SLC16A3 expression via the

Natural Colorants for a Bio-Based Economy-Recovering a Lost Knowledge for Novel Applications of Chrozophora tinctoria Extracts as Paints Through a Multi-Analytical Approach.

Natural colorants, with their sustainable origins, offer a promising alternative for various applications. Advanced studies have unveiled the remarkable properties, resilience, and durability of these ancient dyes, which our ancestors developed through sustainable material processing. This serves as a testament to the potential of sustainable solutions in our field. As part of our research, we prepared three medieval temperas using gum arabic, parchment glue, and casein glue. These tempera were explicitly designed to protect the purples obtained from Chrozophora tinctoria extracts. A comprehen

Proteomic and phosphoproteomic analysis of rabies pathogenesis in the clinical canine brain and identification of a kinase inhibitor as a potential repurposed antiviral agent.

Rabies is a fatal zoonosis caused by the rabies virus (RABV) that has afflicted humans for thousands of years. RABV infection leads to neurological symptoms and death; however, its pathogenesis in the brain is unclear, which complicates patient care. Given that no treatment exists for symptomatic cases, there is an urgent need for effective antiviral drugs. In this study, we aimed to investigate the pathogenic mechanism of RABV in the brain and screen for potential anti-RABV drugs. Protein samples were extracted from the brains of RABV-positive and RABV-negative dogs, and proteomic and phospho

Comparative Investigation of pH-Dependent Availability of Pancreatic Enzyme Preparations In Vitro.

This study aimed to compare different pancreatic enzyme preparations (PEPs) available in Germany regarding particle geometry and size, and to evaluate enzyme activity under physiologically relevant conditions in vitro. Pancreatic endocrine insufficiency is characterized by deficiency of pancreatic enzymes resulting in maldigestion. It is orally treated by pancreatic enzyme replacement therapy. The formulations differ in their physical properties and enzyme release behavior, potentially resulting in inconsistent dosages and poor interchangeability of products. A total of 25 products were analyz

Discovery of CRBN-dependent WEE1 Molecular Glue Degraders from a Multicomponent Combinatorial Library.

Small molecules promoting protein-protein interactions produce a range of therapeutic outcomes. Molecular glue degraders exemplify this concept due to their compact drug-like structures and ability to engage targets without reliance on existing cognate ligands. While Cereblon molecular glue degraders containing glutarimide scaffolds have been approved for treatment of multiple myeloma and acute myeloid leukemia, the design of new therapeutically relevant monovalent degraders remains challenging. We report here an approach to glutarimide-containing molecular glue synthesis using multicomponent

Characterization of the Antibacterial Activity of Quinone-Based Compounds Originating from the Alnumycin Biosynthetic Gene Cluster of a Streptomyces Isolate.

Bacteria of the genus Streptomyces produce various specialized metabolites. Single biosynthetic gene clusters (BGCs) can give rise to different products that can vary in terms of their biological activities. For example, for alnumycin and the shunt product K115, antimicrobial activity was described, while no antimicrobial activity was detected for the shunt product 1,6-dihydro 8-propylanthraquinone. To investigate the antibacterial activity of 1,6-dihydro 8-propylanthraquinone, we produced alnumycin and 1,6-dihydro 8-propylanthraquinone from a Streptomyces isolate containing the alnumycin BGC.

Splicing Interruption by Intron Variants in CSNK2B Causes Poirier-Bienvenu Neurodevelopmental Syndrome: A Focus on Genotype-Phenotype Correlations.

CSNK2B has recently been identified as the causative gene for Poirier-Bienvenu neurodevelopmental syndrome (POBINDS). POBINDS is a rare neurodevelopmental disorder characterized by early-onset epilepsy, developmental delay, hypotonia, and dysmorphism. Limited by the scarcity of patients, the genotype-phenotype correlations in POBINDS are still unclear. In the present study, we describe the clinical and genetic characteristics of eight individuals with POBINDS, most of whom suffered developmental delay, generalized epilepsy, and hypotonia. Minigene experiments confirmed that two intron variants

Mechanics of a Biomimetic Moisture Sensitive Actuator Based on Compression Wood.

Various mechanisms of plant organ movements have been reported, including the close association of two layers with expressed differences in hygroscopic properties. Following this principle, actuator beams composed of thin veneers out of normal and compression wood cut from Scots pine (Pinus sylvestris L.) were prepared by using two types of adhesives. The mismatch of the swelling properties of the two layers in tight combination resulted in an expressed bending deflection in response to set humidity changes. The resulting curvatures were measured and analyzed by the Timoshenko bi-metal-model,
