Renin-independent aldosteronism and chronic kidney disease in diabetes: Observational and Mendelian randomization analyses.

BACKGROUND: Renin-independent aldosteronism (RIA) describes the spectrum of autonomous aldosterone secretion from mild to overt. We aimed to explore whether RIA is causally associated with chronic kidney disease (CKD) in patients with diabetes. METHODS: We cross-sectionally included 1027, 402 and 39,709 patients with any type of diabetes from cohorts of EIMDS, CONPASS and UK Biobank, respectively. In EIMDS, we defined RIA and renin-dependent aldosteronism based on plasma aldosterone and renin concentrations. We performed captopril challenge test to confirm renin-dependent or independent aldos

Gut microbiota and hypertensive disorders in pregnancy: evidence from the Mendelian randomization study.

BACKGROUND: Recent studies have shown that gut microbiota (GM) is related to hypertensive disorders in pregnancy (HDP). However, the causal relationship needs to be treated with caution due to confounding factors and reverse causation. METHODS: We obtained genetic variants from genome-wide association studies including GM (N = 18,340) in MiBioGen Consortium as well as HDP (7,686 cases/115,893 controls) and specific subtypes in FinnGen Consortium. Then, Inverse variance weighted, maximum likelihood, weighted median, MR-Egger, and MR.RAPS methods were applied to examine the causal association.

Implicating Causal Brain Magnetic Resonance Imaging in Glaucoma Using Mendelian Randomization.

BACKGROUND: Glaucoma is hypothesized to originate in the brain but manifests as an eye disease as it possesses the common features of neurodegeneration diseases. But there is no evidence to demonstrate the primary brain changes in glaucoma patients. In the present study, we have used Mendelian randomization (MR) to understand the causal effect of brain alterations on glaucoma. METHODS: Our MR study was carried out using summary statistics from genome-wide associations for 110 diffusion tensor imaging (DTI) measurements of white matter (WM) tracts (17,706 individuals), 101 brain region-of-inte

Comorbidity of Novel CRHR2 Gene Variants in Type 2 Diabetes and Depression.

The corticotropin-releasing hormone receptor 2 (CRHR2) gene encodes CRHR2, contributing to the hypothalamic-pituitary-adrenal stress response and to hyperglycemia and insulin resistance. CRHR2-/- mice are hypersensitive to stress, and the CRHR2 locus has been linked to type 2 diabetes and depression. While CRHR2 variants confer risk for mood disorders, MDD, and type 2 diabetes, they have not been investigated in familial T2D and MDD. In 212 Italian families with type 2 diabetes and depression, we tested 17 CRHR2 single nucleotide polymorphisms (SNPs), using two-point parametric-linkage and lin

Investigating the causality of metabolites involved in one-carbon metabolism with the risk and age at onset of Parkinson's disease: A two-sample mendelian randomization study.

With the aging population and increasing life expectancy, Parkinson's disease (PD), a neurological disorder rapidly increasing in morbidity and mortality, is causing a huge burden on society and the economy. Several studies have suggested that one-carbon metabolites, including homocysteine, vitamin B6, vitamin B12 and folate acid, are associated with PD risk. However, the results remain inconsistent and controversial. Thus, we performed a two-sample Mendelian randomization (MR) study to detect the causality between one-carbon metabolites and PD susceptibility as well as age at PD onset. We col

The role of inflammation and genetics in periodontal disease.

Periodontitis is a complex disease: (a) various causative factors play a role simultaneously and interact with each other; and (b) the disease is episodic in nature, and bursts of disease activity can be recognized, ie, the disease develops and cycles in a nonlinear fashion. We recognize that various causative factors determine the immune blueprint and, consequently, the immune fitness of a subject. Normally, the host lives in a state of homeostasis or symbiosis with the oral microbiome; however, disturbances in homeostatic balance can occur, because of an aberrant host response (inherited and

Genetically Predicted Causal Relationship Between Polycystic Ovary Syndrome and Preeclampsia.

BACKGROUND: Clarifying the causal relationship between polycystic ovary syndrome (PCOS) and its characteristic markers with preeclampsia (PE) is of utmost importance in preventing the development of PE. Observational studies have investigated the relationship, but the conclusions have been inconsistent. METHODS: Utilizing publicly accessible genomewide association study (GWAS) data, we performed a two-sample Mendelian randomization analysis to investigate causal relationships between PCOS, its characteristic biomarkers, and PE. The inverse variance weighting (IVW) method served as the primary

Strength of selection potentiates distinct adaptive responses in an evolution experiment with outcrossing yeast.

Selection intensity is expected to influence the magnitude and genetic architecture of adaptive responses, yet it is rarely evaluated as a standalone variable in experimental evolution studies. Here, we evolved outcrossing populations of Saccharomyces cerevisiae for ∼200 generations across a spectrum of environmental stress from zero to moderate to high ethanol exposure, to examine how genomic responses vary with stress intensity. Across treatments, adaptation proceeded through many subtle allele and haplotype frequency shifts rather than large changes at single loci, consistent with a highly

Exploring the causal relationship and pathogenesis of intracranial aneurysms with osteoporosis based on Mendelian randomization and bioinformatics study.

INTRODUCTION: Intracranial aneurysms (IA) and osteoporosis (OP) are both common conditions among the elderly population. Observational studies suggest a potential association between OP and IA. However, the causal relationships and biological mechanisms underlying these associations remain unclear. MATERIALS AND METHODS: We performed bidirectional two-sample Mendelian randomization (TSMR) analysis using summary statistics from European genome-wide association studies. The inverse variance weighted (IVW) method served as the primary approach for causal inference. Furthermore, sensitivity analy

Migraine is not a risk factor for glaucoma: Evidence from a bidirectional Mendelian randomization study.

Numerous compelling observational studies have indicated that migraine is a risk factor for the development of glaucoma. Nevertheless, some studies have observed entirely opposite results. The objective of our research is to evaluate the potential relationship between migraine and glaucoma by employing a bidirectional Mendelian Randomization (MR) approach. This method enables us to rigorously assess the causal links between these 2 conditions, thereby addressing the discrepancies in existing literature. Independent genetic variants associated with glaucoma and migraine at the genome-wide signi
