Instability in the pediatric elbow can be secondary to trauma, developmental disorders, congenital anomalies, inherited disorders, or acquired systemic processes. The pediatric elbow presents unique challenges with regard to open growth plates, propensity for dislocation and spontaneous reduction, and increased time for the development of post-traumatic deformity into adulthood. The purpose of this article is to review current concepts of injuries leading to elbow instability, discuss how to recognize and treat the instability, and address other, nontraumatic causes of elbow instability.

Since 1968, bone marrow transplantation became the first line therapy for selected metabolic and immunological hereditary disorders. Actually, advances in the supportive care in bone marrow transplantation and a better knowledge of the immunology of BMT complications has been associated with a better disease correction and an increase in long term survival. New approaches are under investigation and include: hematopoietic growth factors, enzymatic replacement and gene therapy. However at the present time BMT is still the only curative treatment for selected hereditary disorders.

Growth faltering, previously known as failure to thrive, is a broad term describing children who do not reach their expected weight, length, or body mass index for age. Growth is assessed with standardized World Health Organization charts for children younger than two years and Centers for Disease Control and Prevention charts for children two years and older. Traditional criteria for growth faltering can be imprecise and difficult to track over time; therefore, use of anthropometric z scores are now recommended. These scores can be calculated with a single set of measurements to assess malnut

OBJECTIVES: The aim of the study is to compare complex anthropometric characteristics in non medicated boys with ADHD and normal population.
METHODS: Complex anthropometric examination of non-medicated ADHD boys (n=46, average age 11.03 years), statistical and clinical comparison to the actual population growth norm. In contrast to the most of the previous studies, which analyzed mostly only BMI or basic signs of growth as height and weight, the presented study operates with a complex anthropometrical measurement and comparison with actual population norm.
RESULTS: The results of the study s

Proper cerebrovascular development and neurogliovascular unit assembly are essential for brain growth and function throughout life, ensuring the continuous supply of nutrients and oxygen. This involves crucial events during pre- and postnatal stages through key pathways, including vascular endothelial growth factor (VEGF) and Wnt signaling. These pathways are pivotal for brain vascular growth, expansion, and blood-brain barrier (BBB) maturation. Interestingly, during fetal and neonatal life, cerebrovascular formation coincides with the early peak activity of the hypothalamic-pituitary-gonadal

It has been established that consumption of alcohol during pregnancy has profound developmental and behavioral effects on the fetus and offspring. The present studies were undertaken to examine the consequences of in utero ethanol exposure on the regulation of insulin-like growth factors (IGFs) in relation to observed somatic growth deficits. Ad libitum, pair- and ethanol-fed female rats were maintained on liquid diet from day 2 of pregnancy through parturition. Pups were sacrificed at birth or cross-fostered to nonexperimental mothers and sacrificed at 10, 20 and 40 days of age. Body and brai

The tropomyosin-related tyrosine kinases or neurotrophic tyrosine kinase receptors are a group of tyrosine kinases that play a crucial role in regulating neuronal growth and development. Neurotrophins are a class of protein-secreting cells that serve as the primary ligand for the Trk receptors. The four primary neurotrophins are nerve growth factor (NGF), brain-derived nerve factor (BDNF), neurotrophin-3, and neurotrophin-4/5. Mounting evidence suggests that Trk receptors can be categorized into three types: TrkA, TrkB, and TrkC. These receptors play a crucial role in facilitating neuronal gro

Fibroblast growth factor 23 (FGF23) is a phosphate-regulating (Pi-regulating) hormone produced by bone. Hereditary hypophosphatemic disorders are associated with FGF23 excess, impaired skeletal growth, and osteomalacia. Blocking FGF23 became an effective therapeutic strategy in X-linked hypophosphatemia, but testing remains limited in autosomal recessive hypophosphatemic rickets (ARHR). This study investigates the effects of Pi repletion and bone-specific deletion of Fgf23 on bone and mineral metabolism in the dentin matrix protein 1-knockout (Dmp1KO) mouse model of ARHR. At 12 weeks, Dmp1KO m

PURPOSE: Liver transplantation (LTx) is performed in individuals with urea cycle disorders when medical management (MM) insufficiently prevents the occurrence of hyperammonemic events. However, there is a paucity of systematic analyses on the effects of LTx on health-related outcome parameters compared to individuals with comparable severity who are medically managed.
METHODS: We investigated the effects of LTx and MM on validated health-related outcome parameters, including the metabolic disease course, linear growth, and neurocognitive outcomes. Individuals were stratified into "severe" and

In recent years, defined progress has been made in understanding the mechanisms of hemodynamic disturbances occurring in liver cirrhosis, which are based on portal hypertension. In addition to pathophysiological disorders related to endothelial dysfunction, it was revealed: There is the restructuring of the vasculature, which includes vascular remodeling and angiogenesis. In spite of the fact that these changes are the compensatory-adaptive response to the deteriorating conditions of blood circulation, taken together, they contribute to the development and progression of portal hypertension ca

Endocrine changes supervening after parturition and menopause participate in the control of sebum production and hair growth modulation. The ensuing conditions include some peculiar aspects of hair loss (effluvium), alopecia, and facial hirsutism. The hair cycling is of major clinical relevance because most hair growth disorders result from disturbances in this chronobiological feature. Of note, any correlation between a biologic abnormality and hair cycling disturbance does not prove a relationship of causality. The proportion of postmenopausal women is rising in the overall population. There…

In developing brains, neural progenitors exhibit cell cycle-dependent nuclear movement within the ventricular zone [interkinetic nuclear migration (INM)] and actively proliferate to produce daughter progenitors and/or neurons, whereas newly generated neurons exit from the cell cycle and begin pial surface-directed migration and maturation. Dysregulation of the balance between the proliferation and the cell cycle exit in neural progenitors is one of the major causes of microcephaly (small brain). Recent studies indicate that cell cycle machinery influences not only the proliferation but also IN

PURPOSE OF REVIEW: Human growth ensues from a complex interplay of physiological factors, in the wider setting of varying genetic traits and environmental influences. Intensive research in these divergent areas, and particularly in the field of genetics, continues to clarify the molecular basis of disorders which result in overgrowth, and it is therefore timely to provide a review of these findings.
RECENT FINDINGS: This article provides an overview of the factors which regulate growth, followed by a discussion of the more commonly encountered overgrowth syndromes and their genetic basis as i

Hearing loss, auricular anomalies and middle ear infections are common findings in many genetic disorders, but the mechanisms have remained unknown. We studied ear and hearing problems in Turner's syndrome (TS) in relation to the degree of X chromosome loss (i.e. degree of mosaicism) and growth. One hundred and nineteen girls and women with TS were studied regarding audiometry, fluorescent in situ hybridisation, serum concentration of insulin-like growth factor-1 (IGF-1) and body height. It was found that sensorineural hearing loss and occurrence of auricular anomalies were significantly incre

Late puberty is defined as the lack of pubertal development at two standard deviations above the mean age for the general population of the geographical area. In practical terms, this is a chronological age of 14 years for males (testicular volume <4 ml) and 13 years for girls (lack of thelarche). The goal of the assessment is to determine whether the delay or lack of development is due to a lag in normal pubertal maturation or represents an abnormality that must be investigated. Etiologies of pubertal delay and pubertal failure include: a) Constitutional delay of puberty (healthy patients wit

BACKGROUND: Early and effective treatment for children with developmental language disorder (DLD) is important. Although a growing body of research shows the effects of interventions at the group level, clinicians observe large individual differences in language growth, and differences in outcomes across language domains. A systematic understanding of how child characteristics contribute to changes in language skills is still lacking.
AIMS: To assess changes in the language domains: expressive morphosyntax; receptive and expressive vocabulary; and comprehension, in children in special needs e

The skin acts as a reflection of systemic endocrine activity, revealing the complex interaction among hormonal, metabolic and immune mechanisms. This narrative review synthesizes current evidence on the molecular and clinical connections between endocrine disorders and dermatologic findings. Endocrine diseases such as diabetes mellitus (DM), thyroid disorders (TD), polycystic ovary syndrome (PCOS), Cushing syndrome (CS) and acromegaly show characteristic skin features that often appear before systemic diagnosis. Dysregulation of insulin-like growth factor-1 (IGF-1) and PI3K/Akt/mTOR pathways,

Pregnancy-related disorders, including preterm birth, preeclampsia, recurrent spontaneous abortion, and fetal growth restriction, represent major contributors to global maternal and infant morbidity and mortality. Their pathophysiology involves complex mechanisms such as immune dysregulation, inflammatory responses, oxidative stress, and metabolic disturbances. Interferon-stimulated gene 15 (ISG15), a ubiquitin-like protein, regulates diverse biological processes-including antiviral immunity, cellular metabolism, oxidative stress, and DNA damage response-through ISGylation, which covalently co

Exosomes are nanosized extracellular vesicles increasingly recognized as regulators of cutaneous biology with direct clinical relevance. In dermatology, they influence keratinocyte and fibroblast proliferation, pigmentation pathways, and immune modulation in disorders such as atopic dermatitis and cutaneous lupus. Their strongest evidence lies in wound healing, where exosomes accelerate closure by dampening proinflammatory cytokines (tumor necrosis factor-α, interleukin-6, matrix metalloproteinase-9), enhancing angiogenesis through vascular endothelial growth factor signaling, and improving co
