Genetic evidence that advanced COVID-19 accelerates longitudinal brain atrophy: A Mendelian randomization study.

Coronavirus disease 2019 (COVID-19) was reported to persist long-term in the brain and leave several long-term neurologic sequelae. However, the causal relationship between COVID-19 and brain aging is still unknown. The genome-wide association study (GWAS) data on COVID-19 phenotypes (susceptibility, hospitalization, and severity), involving a total of 5,779,391 participants, were collected from the COVID-19 Host Genetics Initiative. In addition, GWAS data on longitudinal changes in 15 brain structures, assessed via magnetic resonance imaging across the lifespan, were sourced from the ENIGMA C

Glucose-Related Genetic Risk Score and Its Association With Cerebrovascular Abnormality, White-Matter Hyperintensity Volume, and Brain Microstructure Markers.

AIM: Cerebral white-matter lesions serve as biomarkers for stroke, cardiovascular disease, and mortality. However, no genetic association studies on white-matter hyperintensities (WMH) have focused specifically on the Han Chinese population. Despite previous studies on candidate genes, none have explored association with glucose regulation. This study aimed to investigate the association between a genetic risk score (GRS) derived from glucose-related loci and cerebrovascular abnormality, cerebral WMH, and brain microstructure markers. METHODS: This study is a community-based family research c

Identification of the Critical Life-Stage of Obesity Contributing to Brain Functional Networks.

AIMS: Observational studies suggest that obesity impacts brain functional connectivity, but critical developmental periods for these effects remain unclear. Herein, we aimed to investigate the causal relationships between life-course body weight and brain functional connectivity. METHODS: Mendelian randomization (MR) was applied to infer the causality between life-course body weight (birth weight [n = 80,745], childhood body mass index [BMI; n = 39,620], and adulthood BMI [n = 322,154]) and 191 resting-state functional magnetic resonance imaging traits (n = 34,691) using genome-wide associati

Childhood adiposity underlies numerous adult brain traits commonly attributed to midlife obesity.

Obese adults are often reported to have smaller brain volumes than their non-obese peers. Whether this represents evidence of accelerations in obesity-driven atrophy or is instead a legacy of developmental differences established earlier in the lifespan remains unclear. This study investigated whether early-life differences in adiposity explain differences in numerous adult brain traits commonly attributed to mid-life obesity. We used a two-sample life course Mendelian randomization study in 37 501 adults recruited to UK Biobank (UKB) imaging centres from 2014, with secondary analyses in 6996

Mapping Genetic Associations With Functional Brain Area Alterations in Schizophrenia and Implications for Cortical Development.

BACKGROUND: While prior studies have identified regional reduction in gray matter (GM) volume in schizophrenia, it remains unclear whether these alterations are concentrated in specific brain functional areas and how they relate to genetic factors. This study aimed to identify Brodmann's areas (BAs) with affected GM volume in individuals with schizophrenia, explore associated genetic variants through a genome-wide association study (GWAS), and investigate the potential roles of these genes during cortical development. METHODS: The study recruited 194 individuals with schizophrenia and 330 hea

Causal relationship analysis of MRI measurements of major human internal organs and liver disease.

BACKGROUND: This study aimed to explore the causal association between imaging measurement indicators of major internal organs and liver lesions using a two-sample Mendelian randomization (MR) method. METHODS: Data from the UK Biobank and GWAS Catalog platform were used to select single nucleotide polymorphisms (SNPs) associated with MRI or derived measurement results of various organ indicators as genetic instrumental variables. Data from the FinnGen project's R9 version were used to select liver lesion outcomes, such as nonalcoholic fatty liver disease (NAFLD), nonalcoholic cirrhosis, and p

Neuronal ceroid lipofuscinosis (NCL) is caused by the entire deletion of CLN8 in the Alpenländische Dachsbracke dog.

Neuronal ceroid lipofuscinoses (NCLs) are inherited lysosomal storage diseases that have been described in a variety of dog breeds, where they are caused by different mutations in different genes. However, the causative gene defect in the breed Alpenländische Dachsbracke remained unknown so far. Here we present two confirmed cases of NCL in Alpenländische Dachsbracke dogs from different litters of the same sire with a different dam harboring the same underlying novel mutation in the CLN8 gene. Case 1, a 2-year-old male Alpenländische Dachsbracke was presented with neurological signs including

Hemiconvulsion-hemiplegia-epilepsy syndrome with 1q44 microdeletion: causal or chance association.

Hemiconvulsion-hemiplegia-epilepsy (HHE) syndrome is a rare syndrome characterized by childhood onset partial motor convulsions, hemiplegia, and epilepsy in sequence. Exact pathogenesis is not clear. Here we are describing a 3-year-old girl with HHE syndrome with cytogenetic microarray (CMA) showing deletion of 1.8 Mb in 1q44 region. Along with HHE syndrome, the patient also had global developmental delay, subtle facial dysmorphism, and preaxial polydactyly. Clinical phenotype of 1q44 microdeletion syndrome is quite variable. Main clinical features are microcephaly, seizures, and abnormality o

A wide spectrum of clinical and brain MRI findings in patients with SLC19A3 mutations.

BACKGROUND: SLC19A3 (solute carrier family 19, member 3) is a thiamin transporter with 12 transmembrane domains. Homozygous or compound heterozygous mutations in SLC19A3 cause two distinct clinical phenotypes, biotin-responsive basal ganglia disease and Wernicke's-like encephalopathy. Biotin and/or thiamin are effective therapies for both diseases. METHODS: We conducted on the detailed clinical, brain MRI and molecular genetic analysis of four Japanese patients in a Japanese pedigree who presented with epileptic spasms in early infancy, severe psychomotor retardation, and characteristic brain

Dynamic phenotyping of radioiodine-refractory differentiated thyroid cancer: thyroglobulin trajectories, dual-modal imaging, and genotype-defined treatment resistance.

BACKGROUND: Radioiodine-refractory differentiated thyroid cancer (RAIR-DTC) is a clinically aggressive disease state with limited treatment options. While individual biomarkers, imaging modalities, and genomic drivers have each been studied in isolation, the dynamic relationships among longitudinal thyroglobulin kinetics, evolving dual-modal imaging phenotypes, and underlying genotypes, together with their consequences for targeted therapy response, remain insufficiently characterized. METHODS: We retrospectively analyzed 424 patients with intermediate- to high-risk differentiated thyroid can

A Payment Incentive to Improve Confirmatory Testing in Men With Prostate Cancer.

IMPORTANCE: Among men with favorable-risk (ie, low-risk or favorable intermediate-risk) prostate cancer, confirmatory testing substantially improves the detection of aggressive cancers that may merit treatment instead of conservative management. Despite guideline recommendations, confirmatory testing is inconsistently used, and more than half of men do not receive it. Value-based interventions and payment incentives may improve care quality by motivating adherence to guideline-concordant care. OBJECTIVE: To examine the use of confirmatory testing among men with low-risk prostate cancer, after

Identification of a Novel Nonsense Mutation in PLA2G6 and Prenatal Diagnosis in a Chinese Family With Infantile Neuroaxonal Dystrophy.

BACKGROUND AND PURPOSE: Infantile neuroaxonal dystrophy (INAD) is a subtype of PLA2G6-Associated Neurodegeneration (PLAN) with an age of early onset and severe clinical phenotypes of neurodegeneration. Individuals affected with INAD are characterized by rapid progressive psychomotor deterioration, neuroregression, and hypotonia followed by generalized spasticity, optic atrophy, and dementia. In this case, we aimed to identify the underlying causative genetic factors of a Chinese family with two siblings who presented with walking difficulty and inability to speak. We provided a prenatal diagno

Missense mutation in DYNC1H1 gene caused psychomotor developmental delay and muscle weakness: A case report.

BACKGROUND: The DYNC1H1 gene encodes a part of the dynamic protein, and the protein mutations may further affect the growth and development of neurons, resulting in degeneration of anterior horn cells of the spinal cord, and a variety of clinical phenotypes finally resulting in axonal Charcot-Marie-Tooth disease type 20 (CMT20), mental retardation 13 (MRD13) and spinal muscular atrophy with lower extremity predominant 1 (SMA-LED). The incidence of the disease is low, and it is difficult to diagnose, especially in children. Here, we report a case of DYNC1H1 gene mutation and review the related

Dysregulated Glycoprotein B-Mediated Cell-Cell Fusion Disrupts Varicella-Zoster Virus and Host Gene Transcription during Infection.

UNLABELLED: The highly conserved herpesvirus glycoprotein complex gB/gH-gL mediates membrane fusion during virion entry and cell-cell fusion. Varicella-zoster virus (VZV) characteristically forms multinucleated cells, or syncytia, during the infection of human tissues, but little is known about this process. The cytoplasmic domain of VZV gB (gBcyt) has been implicated in cell-cell fusion regulation because a gB[Y881F] substitution causes hyperfusion. gBcyt regulation is necessary for VZV pathogenesis, as the hyperfusogenic mutant gB[Y881F] is severely attenuated in human skin xenografts. In th
