Activation of lymphocytes by BAT and anti CTLA-4: comparison of binding to T and B cells.

In this study we compare the binding and immune stimulatory properties of BAT and anti CTLA-4 monoclonal antibodies (mAbs). Both antibodies were previously shown to manifest effective immune responses against tumor cells. We have described that BAT antibody, produced against Daudi, a B lymphoblastoid cell line, binds and activates T cells. In this paper we demonstrate that anti CTLA-4, produced against the T-cell activation determinant CTLA-4, binds also to B lymphoblastoid cell lines like Daudi and Raji. Both antibodies do not bind resting B cells. BAT binds resting T lymphocytes as well as a

CTLA4 (CD152) modulates the Th subset response and alters the course of experimental Leishmania major infection.

Since both the nature and the amplitude of an antigen-specific T cell response are dependent on co-stimulatory signals, we have investigated the role of CD28/CD152-mediated T cell co-stimulation in the regulation of experimental cutaneous leishmaniasis. CD28-deficient mice and their wild-type littermates are equally susceptible to Leishmania major infection. Whole anti-CD152 antibody significantly exacerbates the disease while anti-CD152 Fab ameliorates the disease in genetically susceptible BALB/c mice but not in C57BL/6, a resistant strain. The anti-CD152-induced exacerbation of the disease

Antibody-drug conjugates in lung cancer: clinical development, therapeutic advances, and emerging challenges.

Antibody-drug conjugates (ADCs) are designed to deliver cytotoxic agents directly to tumor cells by conjugating an antigen-specific monoclonal antibody with a potent payload. By recognizing tumor-associated antigens and undergoing intracellular processing, ADCs enhance antitumor activity while minimizing damage to normal tissues. This review focuses on the research progress of ADCs targeting non-small cell lung cancer (NSCLC) and small cell lung cancer (SCLC), providing an in-depth analysis of their mechanisms of action, molecular design, and clinical efficacy, with particular emphasis on nove

Duocarmycin-Bearing Antibody-Mimetic Drug Conjugate Combined With an ATR Inhibitor Results in Complete Tumor Regression in a KPL-4 Xenograft Model.

Antibody-drug conjugates (ADCs) have demonstrated superior clinical outcomes in patients with HER2-positive advanced breast cancer compared with previous treatment modalities. However, resistance and treatment-limiting toxicities frequently emerge during repeated administration, underscoring an unmet need for strategies to overcome ADC failure. In this study, we investigated whether inhibition of ataxia telangiectasia and Rad3-related protein (ATR) could enhance the antitumor efficacy of antibody-mimetic drug conjugates (AMDCs), a targeted payload delivery platform. Combination treatment with

Restoration of the immune system with base editing and non-genotoxic conditioning in a Rag2 point-mutant mouse model.

Transplantation of donor hematopoietic stem and progenitor cells (HSPCs) is a well-established curative treatment for various blood and immune diseases, including severe combined immunodeficiency (SCID). However, it comes with significant toxicities, including graft-versus-host disease (GvHD) and tissue damage resulting from the use of genotoxic chemotherapy-containing conditioning regimens. Autologous transplantation using gene-modified HSPCs eliminates GvHD but currently still relies on genotoxic conditioning. Further, gene modification of HSPCs has commonly utilized integrating viruses, whi

A Novel FGFR3-Targeting Antibody-Drug Conjugate Induces Tumor Cell Apoptosis through the cGAS-STING Pathway in Bladder Cancer.

Antibody-drug conjugates (ADCs) emerge as a potent cancer therapeutic strategy by enabling precise antigen recognition and efficient intracellular delivery of cytotoxic payloads. In this study, 7-ethyl-9-fluorocamptothecin (A2), a camptothecin derivative, which demonstrates potent tumor-suppressive effects across cellular models, patient-derived organoids (PDOs), and cell line-derived xenograft/patient-derived xenograft (CDX/PDX) models is identified. Through pull-down/mass spectrometry analysis, MAD2L1 is identified as the direct target of A2. A2 specifically binds to the Lys73 site of MAD2L1

TRBC2-targeting antibody-drug conjugates for the treatment of T cell cancers.

Antibody-drug conjugates (ADCs) have been remarkably successful in treating solid and hematological malignancies. Generation of ADCs for T cell cancers is challenging because the ADCs must selectively target cancerous T cells while sparing some normal T cells necessary for immune function. T cells express one of two TRBC alleles: TRBC1 or TRBC2. Normal T cells are composed of about 40% TRBC1-expressing and 60% TRBC2-expressing cells. In contrast, T cell malignancies are characterized by the clonal expression of either TRBC1 or TRBC2. Selective targeting of TRBC1 or TRBC2 enables the killing of
