In this study, we evaluate the effect of an intraportal injection of absolute ethanol in the canine liver and discuss how to introduce it in clinical application. The intraportal injection of ten milliliters of ethanol using a balloon occlusion catheter created an evident and well-demarcated necrotic lesion in the right central lobe. Histological examination showed a typical coagulation necrosis with fibrous bands, which became thicker day by day. The weight of the necroses ranged from 5 to 55 grams with an average of 26.0 +/- 5.5 grams (mean +/- SE), which was approximately 20 to 40 per cent

Trypanosoma brucei brucei experimentally infected sheep with central nervous system involvement were treated with melarsoprol (Arsobal, Specia, France). The purpose was to determine minimum efficiency concentrations of this drug. Eight sheep divided in four groups of two animals were treated intravenously for 3 days with the following respective dosages: 1.8, 0.9, 0.45 and 0.225 mg/kg/d. After a year of observation, efficient dosages were 1.8 and 0.9 mg/kg/d. The experimentation suggests the possibility to reduce usually given dosages in man.

The absorption of aspirin used in the form of lysine acetylsalicylate was studied in the rabbit. Each animal received the drug by three routes: intravenous, gastric and duodenal. Plasma concentrations of acetylsalicylic acid (ASA) and salicylic acid (SA) were compared. The pharmacokinetics of intravenously injected potassium cyanide have been studied in Beagle bitches. In the period up to about 80 min after dosing, blood levels fell in a manner consistent with first-order elimination kinetics. Thereafter blood cyanide concentrations fell at a slower rate, indicating that a second phase of slower elimination had been entered.

Metronidazole is being extensively used in neonates for the prevention and treatment of anaerobic infections. However, detailed pharmacokinetic studies are not available. Thirty neonates receiving intravenous metronidazole 7.5 mg/kg at eight-hour intervals were studied with serial blood levels. The levels increased up to the third day of treatment and then reached a plateau. Neonates less than seven days of age showed higher levels than older infants. High doses of gammaglobulin intravenously have been found useful in controlling thrombocytopenia of a neonate born to a mother who developed acute idiopathic thrombocytopenic purpura during pregnancy. Administration of 0.4 gm/kg/day of intact gammaglobulin intravenously for five days was followed by dramatic rise of platelet count without any side-effect. After 1 month the platelet count was still normal. Passively-acquired immune thrombocytopenia of the newborn consequently is another indication for intravenous gammaglobulin therapy.

Two preparations of diazepam, Valium Roche mixed Micelles and Diazemuls, were compared as intravenous premedicants in 119 patients undergoing minor gynaecological surgery. There were no differences between the formulations with regard to anxiolytic effects and systemic tolerance. There were no symptoms typical of thrombophlebitis in the Valium Roche Mixed-Micelles group and only one case in the Diazemuls group. The effects of acute administration of ethanol were studied in a total of 18 normal male subjects. Sixteen men were given oral ethanol (2.5 ml/kg vodka or gin) and four received intravenous infusions of ethanol (1 ml/kg), at two times of day, 0900 h and 1800 h. Neither intravenous nor oral ethanol caused a specific rise in plasma cortisol, in contrast to previously reported work. There was no evidence for circadian variation in response. Since ethanol does not release plasma cortisol in normal individuals, the pathogenesis of 'alcohol-induced pseudo-Cushing's syndrome' should be re-considered.
