Vitamin D binding protein in ventral CA3 of the hippocampus drives stress-induced depression.

The ventral hippocampus plays a crucial role in regulating emotional and motivated behaviors. However, the molecular mechanisms underlying its dysfunction in major depressive disorder (MDD) remain poorly understood. Here, we identified elevated expression of vitamin D binding protein (VDBP) in the hippocampi of chronic unpredictable mild stress (CUMS)-susceptible mice, which correlated with the severity of depression-like behaviors. Notably, VDBP up-regulation was specifically in ventral CA3 excitatory (vCA3CaMKIIα) neurons. CA3CaMKIIα VDBP overexpression led to inhibited neuronal activity, sy

Structural and Functional Consequences of Arg512 Mutations in Methyl-CpG-Binding Domain Protein 4.

Methyl-CpG-binding domain protein 4 (MBD4) is a base excision repair glycosylase that removes the thymine of a T:G mispair. If left unrepaired, these mismatches can cause C>T transitions. Human MBD4 uses an arginine finger, R468, to flip the mispaired thymine into its active site and to stabilize the orphaned guanine. MBD4 also possesses a second highly conserved arginine, R512, in close proximity to R468, whose role remains unknown despite an R512W mutation catalogued in colorectal cancers and an R512Q mutation reported in Myelodysplastic Syndromes. Here, we investigate the structural and fun

RNA-binding protein Rbm38 as a multifaceted post-transcriptional regulator in zebrafish pancreatic development.

RNA-binding motif protein 38 (Rbm38), also known as RNPC1, is a major regulator of post-transcriptional gene expression. It represents a potential candidate gene linked to the susceptibility of type 2 diabetes, and decreased RBM38 expression can enhance the proliferation of pancreatic cancer cells in humans. However, its role in pancreatic development remains elusive. In this study, we explored the function of Rbm38 using zebrafish as a model. Pancreatic expression of Rbm38 is present at larval stages and is controlled by several transcription factors acting on specific rbm38 promoter regions.

HSV-1 origin binding protein UL9 forms intermolecular DNA tethers and intramolecular DNA loops.

Herpesviruses are ubiquitous human pathogens, causing mild to severe symptoms ranging from cold sores to nasopharyngeal carcinoma. Even though replication of the dsDNA genome has been studied for decades, we still lack a complete molecular understanding of its mechanism. It has been previously proposed, but never shown directly, that the HSV-1 origin-binding protein UL9 interacts with two closely spaced sites within the oriS origin sequence, thereby mediating origin looping, which in turn facilitates replication initiation. Here, we use an array of single-molecule approaches to test this long-

Tricyclic Antidepressants as Prospective α-Synucleinopathy Fighters: Evidence of Protein Binding and Antiaggregation Activity.

Intrinsically disordered proteins (IDPs) like α-synuclein are pivotal in neurodegenerative diseases but present formidable challenges for drug discovery due to their conformational heterogeneity and lack of defined binding pockets. Here, we report the finding that clinically approved tricyclic antidepressants imipramine, amitriptyline, and doxepin bind directly to monomeric α-synuclein and inhibit its fibrillation, a key process in Parkinson's pathology. Using a combination of NMR spectroscopy (WaterLOGSY and chemical shift perturbation, CSP) and molecular dynamics simulation methods, we eluci

Heat stress activates the coagulation cascade through Z-DNA-binding protein 1-dependent necroptosis.

Heatstroke, a severe hyperthermic condition, is characterized by a core body temperature >40°C and multiple organ dysfunction syndrome (MODS), with an extremely high mortality rate. Despite advances in identifying heatstroke-induced cell death pathways, the molecular cascades that bridge heat-induced cell death to MODS and mortality are not yet fully characterized. Our findings demonstrate that Z-DNA-binding protein 1 (ZBP1)-triggered disseminated intravascular coagulation critically drives MODS and fatal outcomes in heatstroke. Heat stress activates ZBP1-dependent necroptosis, promoting tissu

XL-MS and De Novo Protein Design Identified a Common Motif for TREM2 Binding.

Apolipoprotein E (APOE) and Triggering Receptor Expressed on Myeloid cells 2 (TREM2) are the two strongest genetic risk factors of late-onset Alzheimer's disease. ApoE binds to the low-density lipoprotein receptor (LDLR) to facilitate the uptake of ApoE-lipoprotein particles. TREM2 is a cell surface receptor expressed on microglia in the brain. The activation of TREM2 is essential for microglia to carry out protective functions against AD pathology. Several studies have shown that TREM2 signaling is activated through direct interaction between TREM2 and ApoE. In addition to its important role

High-Throughput FRET Affinity Screening Technique (HTFAST) For Cell-Free Expressed Binding Protein Characterization.

The rapid engineering of high-affinity binding proteins, such as nanobodies and single-domain antibodies (sdAbs), is increasingly driven by cell-free, machine-learning-guided optimization. However, high-throughput, quantitative characterization of binding affinity remains a major bottleneck, particularly for proteins expressed in cell-free systems without purification. Here, we present High-Throughput FRET Affinity Screening Technique (HTFAST) for rapid affinity characterization of binders expressed directly in crude E. coli cell-free protein synthesis reactions. HTFAST leverages Förster reson

JPT2/HN1L functions as an NAADP-binding protein in a cell-type-specific manner.

Nicotinic acid adenine dinucleotide phosphate (NAADP) is a second messenger evoking Ca²⁺ release from intracellular Ca²⁺ stores by targeting several Ca2+ channels, including two-pore channels (TPC1/2), transient receptor potential mucolipin-1 (TRPML1), or ryanodine receptor type 1 (RYR1). For activation of Ca2+ channels, NAADP requires binding proteins, such as JPT2/HN1L and LSM12. So far, their function has been analyzed in several cell lines and only a very limited number of primary cells; however, their physiological relevance in cell types known to utilize NAADP signaling remains unclear.

Heme-binding protein CYB5D1 couples intraflagellar redox to calcium signaling for coordinated flagellar beating.

Coordinated ciliary/flagellar beating requires precise spatiotemporal regulation of molecular motors such as dyneins, yet the molecular mechanisms governing ciliary synchrony remain poorly understood. Here, we demonstrate that a heme-binding axonemal protein CYB5D1 functions as a redox-sensitive switch that controls flagellar beating coordination by regulating Ca2+ dynamics. Both the D58G point mutation, which abolishes heme-binding activity, and the complete loss of CYB5D1 lead to a reduction in the flagellar redox potential. More importantly, the hyperreductive intraflagellar redox shift in

In vitro antiviral effects of an mTORC1 inhibitor targeting the rapamycin-binding protein complex.

The persistent threat of mutable viral pathogens highlights the need for broad-spectrum antiviral drugs that target stable host factors to provide a high genetic barrier to resistance. The mammalian target of rapamycin complex 1 (mTORC1) serves as a central metabolic bottleneck in cellular metabolism. It is commonly co-opted by both DNA and RNA viruses to enhance the translation of their structural proteins. Here, we evaluated WRX606, a novel mTORC1 inhibitor, as a broad-spectrum antiviral candidate against representative RNA viruses. WRX606 demonstrated low cytotoxicity across multiple host c

Fatty acid binding protein 4 (FABP4): a key player in neuroinflammation and neuropathic pain.

Neuropathic pain (NP) is caused by lesions or diseases of the somatosensory system. Emerging evidence implicates adipokines in NP pathogenesis, yet the role of fatty acid-binding protein 4 (FABP4) remains unclear. Using a mouse model of sciatic nerve crush injury, we found that wild-type (WT) mice developed robust NP behaviors, concomitant with significant FABP4 upregulation and extensive macrophage infiltration in the injured nerve. In contrast, FABP4-knockout (FABP4-KO) mice exhibited markedly attenuated pain hypersensitivity. Single-cell RNA sequencing and subsequent histological analyses r

RNA-binding protein OTUD1 maintains skeletal homeostasis by determining mesenchymal stem cell lineage commitment.

Mesenchymal stem cells (MSCs) maintain bone homeostasis through osteogenic differentiation. During aging, MSCs undergo a fate shift toward adipogenesis rather than osteogenesis, but the post-transcriptional mechanisms remain unclear. Here, we identify ovarian tumor domain-containing protein 1 (OTUD1) as an RNA-binding protein that controls MSC fate by stabilizing osteogenic transcripts. OTUD1 directly binds and stabilizes BMP2 mRNA, thereby supporting osteogenic differentiation. In vivo, OTUD1 deficiency does not affect early skeletal development but progressively impairs bone homeostasis duri

Fatty acid-binding protein 2 in inflammatory bowel disease: mechanistic insights and translational perspectives.

Fatty acid-binding protein 2 (FABP2) is an intestine-specific cytoplasmic lipid chaperone with a molecular weight of approximately 14-15 kDa that plays a pivotal role in the uptake, intracellular trafficking, and metabolism of long-chain fatty acids. Emerging evidence indicates that FABP2 is not only a sensitive biomarker of intestinal mucosal injury but also may contribute to the pathogenesis and progression of IBD, potentially through the modulation of PPARγ signaling pathway, although the underlying mechanisms require further investigation. This review systematically summarizes the structur

The RNA-Binding Protein ELAVL4/HuD Promotes Stress-Associated Tumor Survival in Neuroendocrine Cancers.

Neuro-endocrine-related cancers overexpress ELAVL4 (HuD), a neuron-specific RBP implicated in post-transcriptional regulation. HuD plays a role in tumor cell survival; its knockdown in cancer cells and tumors leads to reduced growth and viability. In this review, we addressed HuD's role in several molecular pathways that contribute to the development of cancer. For example, cancer cells that have high levels of HuD are able to better withstand stressful tumor microenvironments because HuD stabilizes certain mRNAs, which promotes tumor development, modulates oxidative and metabolic stress, enha

A lipid-binding protein in black-legged tick saliva selectively recognizes Borrelia burgdorferi lipids.

A proteomic analysis of Ixodes scapularis nymph saliva identified 252 proteins, including six tubular lipid-binding proteins (TULIPs). Comparing nymphs fed on mice that were uninfected or infected with Borrelia burgdorferi, twelve salivary proteins showed significant differences in the amounts detected, including XP_040079658.2, which we refer to as TULIP2. Considering the known immunity-related functions of some TULIPs, we expressed and purified TULIP2 from Escherichia coli and analyzed its interaction with B. burgdorferi lipids. The purification of TULIP2 from E. coli presented many obstacle

RNA-Binding Protein NF90 Mediates Polycomb-Independent Transactivation by EZH2 to Promote Cancer Growth.

Increasing evidence suggests critical roles of the polycomb-independent transactivation function of EZH2 in promoting some cancers, such as prostate cancer (PCa), yet the underlying mechanism remains poorly understood. Here, we identify the RNA-binding protein NF90 as a key mediator of this activity. NF90 interacts with EZH2, but not with other core components of the polycomb repressive complex 2 (PRC2), through its RNA-binding modules. Conversely, EZH2 engages NF90 via its intrinsically disordered RNA-binding domain in an RNA-dependent manner. NF90 and EZH2 mutually recruit each other to the

RNA binding of GAPDH controls transcript stability and protein translation in acute myeloid leukemia.

Dysregulation of RNA binding proteins (RBPs) is a hallmark in cancerous cells. In acute myeloid leukaemia (AML) RBPs are key regulators of tumour proliferation. While classical RBPs have defined RNA binding domains, RNA recognition and function in AML by non-canonical RBPs (ncRBPs) remain unclear. Given the inherent complexity of targeting AML broadly, our goal was to uncover potential ncRBP candidates critical for AML survival using a CRISPR/Cas-based screening. We identified the glycolytic enzyme glyceraldehyde-3-phosphate dehydrogenase (GAPDH) as a pro-proliferative factor in AML cells. Bas

The RNA-Binding Protein IGF2BP1 Marks Germ Cells but Is Dispensable for Mouse Fertility.

Insulin-like growth factor 2 mRNA binding protein 1 (IGF2BP1) is a key reader of N6-methyladenosine modifications that regulate target mRNA stability in eukaryotic cells; however, its role in germ cells has never been explored. Here, we analyzed the spatiotemporal expression of IGF2BP1 and revealed that it was present not only in oocytes of the mouse ovary but also in ZBTB16-positive undifferentiated spermatogonia in the mouse testis. Coimmunoprecipitation and fluorescence staining revealed that IGF2BP1 interacted with TRIM71, a regulator of spermatogonia differentiation, but that its expressi
