Efficacy and safety of ketamine for post-traumatic stress disorder: A systematic review with narrative synthesis and consideration of military and veteran populations.

Post-traumatic stress disorder (PTSD) is a debilitating disorder with limited pharmaceutical treatment options. Ketamine, a noncompetitive N-methyl-d-aspartate (NMDA) receptor antagonist with rapid-onset effects, has emerged as a potential treatment, particularly for patients who do not respond to first-line therapies. This systematic review synthesizes the current evidence on the effectiveness and safety of ketamine for PTSD, with a focus on military and veteran populations. Eligible studies included adults with PTSD, who evaluated ketamine or esketamine, and reported efficacy and safety outc

The relationship between perceived social support and post-traumatic stress disorder in Jordanian women experiencing intimate partner violence.

BACKGROUND: Previous studies agreed that post-traumatic stress disorder (PTSD) is a pervasive consequence affecting women who were exposed to intimate partner violence. Low levels of perceived social support could be associated with PTSD. This study aimed to investigate the association between social support provided to women exposed to intimate partner violence and PTSD. METHODS: A cross-sectional quantitative research design was used to recruit women exposed to intimate partner violence residing in Amman, Irbid, and Zarqa. A non-probability convenience sampling method was used to recruit 166

Blood biomarker changes and relationships after low dose oral ketamine treatment for post-traumatic stress disorder (PTSD).

RATIONAL: Ketamine has been investigated as a treatment alternative for PTSD for the last 20 years, yet there have been limited reports of biological changes or biomarker characterisation related to treatment. OBJECTIVES: To address this significant gap, this study analysed blood samples from 25 participants with PTSD who took part in an open-label 6-week trial of low dose oral ketamine treatment. METHODS: Serum and plasma samples were quantified before and after ketamine treatment for brain-derived neurotrophic factor (BDNF), vascular endothelial growth factor A (VEGF-A), serotonin, FK506 bin

Network-level structural alterations distinguish persistent from remitted post-traumatic stress disorder: a morphometric covariance approach.

Background: Post-traumatic stress disorder (PTSD) exhibits heterogeneous clinical trajectories: while many affected individuals achieve remission, a substantial proportion develop a chronic, persistent form. The neuroanatomical mechanisms distinguishing these divergent outcomes remain poorly defined. This study aimed to elucidate the structural network organisation of persistent PTSD using structural covariance network analysis. Methods: Participants included trauma-unexposed healthy controls (HC; n = 112), individuals with remitted PTSD (n = 32), and those with persistent PTSD (n = 78). Associations between complex post-traumatic stress disorder and intimate relationship quality: the parallel mediating roles of mentalization and playfulness.

BACKGROUND: The association between Complex Post-Traumatic Stress Disorder (CPTSD) and intimate relationship quality remains underexplored, particularly regarding protective factors that may buffer against trauma-related impairments. METHODS: A total of 3,250 participants completed self-report surveys assessing CPTSD symptoms, intimate relationship quality, mentalization, and playfulness, with 12.8% (N = 416) meeting criteria for probable CPTSD. A dynamic network heatmap and Mantel-based network analysis revealed distinct connectivity patterns. Structural equation modeling (SEM) was used to e

A preliminary clinical risk model for probable post-traumatic stress disorder symptoms among intensive care unit survivors.

BACKGROUND: Post-traumatic stress disorder (PTSD) is a disabling sequela among survivors of critical illness, yet early identification of high-risk individuals during hospitalization remains challenging. We aimed to develop a clinically interpretable prediction model for probable PTSD symptoms among ICU survivors using routinely collected clinical data. METHODS: In this single-center retrospective cohort study, adult ICU survivors were assessed for probable PTSD symptoms after discharge using the PC-PTSD-5 questionnaire. A multivariable logistic regression model was developed with predictor s

Dynamic immune and metabolic dysregulation in women with post-traumatic stress disorder: Longitudinal transcriptomic insights following sexual assault.

Post-traumatic stress disorder (PTSD) is a chronic and debilitating psychiatric condition, and sexual assault represents the leading risk factor for PTSD in women, with up to 50% of survivors developing the disorder. However, most transcriptomic studies have focused on male military veterans, limiting insight into the molecular mechanisms underlying PTSD in women. We conducted a longitudinal RNA sequencing study of blood samples from 65 women with PTSD and 65 healthy controls (HC). One-year follow-up assessments were performed in 35 PTSD participants after completion of treatment with either s

Post-traumatic stress disorder impairs CD8+ T cell-dependent anti-tumor immune responses by downregulating thiamine.

Post-traumatic stress disorder (PTSD) is a severe and chronic psychiatric condition. However, whether PTSD influences tumor immunity remains poorly understood. In this study, we demonstrated that PTSD promotes the growth of subcutaneous transplant tumors in mice, an effect that can be reversed by anti-CD8 antibody treatment. Furthermore, we found that PTSD downregulates serum thiamine levels, which mediates the tumor-promoting effects observed in PTSD mice. In vitro, thiamine enhances CD8+ T cell activation and cytotoxic function. Mechanistically, thiamine augments tricarboxylic acid (TCA) cyc

Socioeconomic deprivation and maternal mental health in developing countries: a multicenter study of postpartum depression and post-traumatic stress disorder.

Despite growing research on maternal mental health, evidence from low- and middle-income countries, particularly in Africa, remains limited and fragmented. We provide context-specific evidence in the post-COVID era and address an important gap in the literature on socioeconomic vulnerability and maternal mental health. This study investigates the effect of socioeconomic deprivation on postpartum depression and post-traumatic stress disorder among postpartum women using validated assessment tools. Maternal mental health outcomes are assessed using the Edinburgh Postnatal Depression Scale (EPDS)

Large-Scale Data Analysis of Post-Traumatic Stress Disorder (PTSD) Through GWAS Fine-Mapping and Systems Biology.

Background/Objectives: Post-Traumatic Stress Disorder (PTSD) is a complex psychiatric condition with a strong polygenic and stress-related biological basis. Although Genome-Wide Association Studies (GWAS) have acknowledged abundant risk variants, translating these findings into biologically meaningful candidates remains challenging. This study introduces an integrative computational approach from raw file preparation by python-coded application into downstream in-depth silico analyses designed to systematically refine GWAS signals for PTSD using fine-mapping, linkage disequilibrium (LD), and h

Childhood trauma and systemic lupus erythematosus activity in Tunisian patients: exploring the mediating role of post-traumatic stress disorder.

BACKGROUND: Systemic lupus erythematosus (SLE) is a chronic autoimmune disease influenced by multiple factors. Growing evidence suggests that adverse childhood experiences (ACEs) and post-traumatic stress disorder (PTSD) may contribute to immune dysregulation and increased disease activity. However, these associations remain poorly explored in North Africa and the Middle East. OBJECTIVE: This study aimed to investigate the mediating role of PTSD in the path from ACEs to SLE activity. METHODS: This cross-sectional study involved adult patients with SLE treated at the Internal Medicine Departm

Examining the epigenetic transmission of risk for chronic pain associated with paternal post-traumatic stress disorder: a focus on veteran populations.

Chronic pain is a public health problem that significantly reduces quality of life. Although the aetiology is often unknown, recent evidence suggests that susceptibility can be transmitted intergenerationally, from parent to child. Post-traumatic stress disorder (PTSD) is a debilitating psychological disorder, often associated with chronic pain, that has high prevalence rates in military personnel and Veterans. Therefore, we aimed to characterise the epigenetic mechanisms by which paternal trauma, such as PTSD, is transmitted across generations to confer risk in the next generation, specifical

Interprofessional collaboration across sectors for unemployed refugees with post-traumatic stress disorder in Denmark: a panacea to person-centered care?

Many refugees suffer from post-traumatic stress disorder (PTSD), influenced by traumatic experiences and post-migration stressors, including unemployment. This complexity calls for person-centered care (PCC) and interprofessional collaboration across sectors. In this qualitative study we aimed to understand what refugees and professionals from two sectors value about participating in cross-sector network meetings to coordinate and agree on shared plans for PTSD treatment and assessing employability. We conducted interviews with 24 unemployed refugees, 10 physicians, and 20 municipal employment
