Biomonitoring of mycotoxins and exposomics-informed profiling in extreme preterm infants from the NeoVIP cohort.

Extremely premature infants are a highly vulnerable patient group that might be exposed to a vast number of dietary and environmental contaminants, which may impact their health trajectories. This study assessed xenobiotic exposures in 26 extremely premature infants, with a special focus on dietary mycotoxins, using a dual analytical approach combining four sensitive, targeted LC-MS/MS methods and one non-targeted high-resolution mass spectrometry (HR-MS) assay. A total of 187 matched biological samples comprising breast milk (n = 35), plasma (n = 48), urine (n = 52), and stool (n = 52) collec

Integrating multi-omics, EWAS, and reverse network toxicology to explore environmental pollutant risks in erectile dysfunction.

BACKGROUND: Erectile dysfunction (ED) is increasingly prevalent worldwide, arising from complex interactions between genetic susceptibility and environmental exposure. Real-world exposure involves complex chemical mixtures that may induce synergistic toxicity, which traditional methods struggle to elucidate. This study integrates multi-omics data with reverse network toxicology to systematically identify causal molecular targets and environmental pollutants underlying ED risk, thereby clarifying their mechanisms. METHODS: We performed summary data-based Mendelian randomization (SMR) integrati

The xenoestrogens biphenol-A and nonylphenol differentially regulate metalloprotease-mediated shedding of EGFR ligands.

The xenoestrogens bisphenol-A (BPA) and nonylphenol (NP) are endocrine disruptors used in the plastic polymer industry to manufacture different products for human use. Previous studies have suggested a role of these compounds in the shedding of signaling molecules, such as tumor necrosis factor α (TNF-α). The aim of this work was to evaluate the effect of BPA and NP on the sheddase ADAM17 and its newly discovered regulators iRhom1 and iRhom2 in the release of EGFR-ligands. We report that BPA and NP can stimulate the release of the ADAM17-substrates HB-EGF and TGF-α. In cells lacking ADAM17 (Ad

Prenatal Exposure to Nonpersistent Chemicals and Fetal-to-childhood Growth Trajectories.

INTRODUCTION: Prenatal exposure to nonpersistent chemicals, including organophosphate pesticides, phthalates, and bisphenols, is associated with altered fetal and childhood growth. Few studies have examined these associations using longitudinal growth trajectories or considering exposure to chemical mixtures. METHODS: Among 777 participants from the Generation R Study, we used growth mixture models to identify weight and body mass index trajectories using weight and height measures collected from the prenatal period to age 13. We measured exposure biomarkers for organophosphate pesticides, ph

Data integration, analysis, and interpretation of eight academic CLARITY-BPA studies.

"Consortium Linking Academic and Regulatory Insights on BPA Toxicity" (CLARITY-BPA) was a comprehensive "industry-standard" Good Laboratory Practice (GLP)-compliant 2-year chronic exposure study of bisphenol A (BPA) toxicity that was supplemented by hypothesis-driven independent investigator-initiated studies. The investigator-initiated studies were focused on integrating disease-associated, molecular, and physiological endpoints previously found by academic scientists into an industry standard guideline-compliant toxicity study. Thus, the goal of this collaboration was to provide a more compr

Delayed onset of puberty in male offspring from bisphenol A-treated dams is followed by the modulation of gene expression in the hypothalamic-pituitary-testis axis in adulthood.

Bisphenol A (BPA) is a synthetic endocrine-disrupting chemical of high prevalence in the environment, which may affect the function of the hypothalamic-pituitary-testis (HPT) axis in adult rats. The aim of the present study was to evaluate whether exposure to BPA during hypothalamic sexual differentiation at doses below the reproductive no observable adverse effect level of the World Health Organization causes changes in the regulation of the HPT axis. For this, 0.5 or 5mgkg-1 BPA was injected subcutaneously to the mothers from gestational day 18 to postnatal day (PND) 5. In adulthood (PND90),

Gestational and lactational exposure to bisphenol AF in maternal rats increases testosterone levels in 23-day-old male offspring.

During prenatal and postnatal development, exposure to environmental chemicals with estrogenic activity, such as bisphenol AF (BPAF), may result in reproductive disorders. Currently, the mechanisms behind such disorders in male offspring induced by gestational and lactational exposure to BPAF remain poorly understood. Here, female rats from gestational day (GD) 3-19 were exposed to 100 mg BPAF/kg/day by oral gavage. On the day of birth (postnatal day (PD) 0), cross-fostering took place between treated and control litters, and cross-fostered mother rats were given BPAF 100 mg/kg/day during the

Adult exposure to bisphenol A (BPA) in Wistar rats reduces sperm quality with disruption of the hypothalamic-pituitary-testicular axis.

Reproductive physiology involves complex biological processes that can be disrupted by exposure to environmental contaminants. The effects of bisphenol A (BPA) on spermatogenesis and sperm quality is still unclear. The objective of this study was to investigate the reproductive toxicity of BPA at dosages considered to be safe (5 or 25mg BPA/kg/day). We assessed multiple sperm parameters, the relative expression of genes involved in the central regulation of the hypothalamic-pituitary-testicular axis, and the serum concentrations of testosterone, estradiol, LH and FSH. BPA exposure reduced sper

Bisphenol A exposure alters developmental gene expression in the fetal rhesus macaque uterus.

Bisphenol A (BPA) exposure results in numerous developmental and functional abnormalities in reproductive organs in rodent models, but limited data are available regarding BPA effects in the primate uterus. To determine if maternal oral BPA exposure affects fetal uterine development in a non-human primate model, pregnant rhesus macaques carrying female fetuses were exposed orally to 400 µg/kg BPA or vehicle control daily from gestation day (GD) 50-100 or GD100-165. Fetal uteri were collected at the completion of treatment (GD100 or GD165); tissue histology, cell proliferation, and expression o

Urinary bisphenol A concentrations are associated with abnormal liver function in the elderly: a repeated panel study.

BACKGROUND: Bisphenol A (4,40-isopropylidenediphenol, BPA) is known to adversely affect various organs. The liver is reported to be affected by BPA in animal studies. However, there are few studies in humans on the effects of BPA on the liver. Therefore, we evaluated the relationship between urinary BPA levels and liver function in elderly subjects using repeated measurements. METHODS: From 2008 to 2010, a total of 560 elderly subjects residing in Seoul were each evaluated up to three times. At the first visit, demographic data, environmental exposure and lifestyle information were obtained f

Prediction of human genes and diseases targeted by xenobiotics using predictive toxicogenomic-derived models (PTDMs).

New technologies for systems-level determinants of human exposure to drugs, industrial chemicals, pesticides, and other environmental agents provide an invaluable opportunity to extend the understanding of human health and potential environmental hazards. We report here the development of a new computational-systems toxicology framework, called predictive toxicogenomics-derived models (PTDMs). PTDMs integrate three networks of chemical-gene interactions (CGIs), chemical-disease associations (CDAs) and gene-disease associations (GDAs) to infer chemical hazard profiles, identify exposure data ga

Determination of bisphenol a in urine from mother-child pairs-results from the duisburg birth cohort study, Germany.

Bisphenol A (BPA) may alter endocrine functions, and human exposure is widespread. In the Duisburg birth cohort study the influence of several contaminants was examined on the neuropsychological and pubertal development of children. This study reports the biomonitoring results on BPA within the 6-yr follow-up study (children's age 6-8 yr). Total BPA and free (unconjugated) BPA concentrations in 208 urine samples of 104 mother-child pairs were measured. For quality control, total BPA was analyzed by two independent laboratories. BPA was detected in all urine samples, while free BPA was observed

Trends in environmental chemical concentrations in the Canadian population: Biomonitoring data from the Canadian Health Measures Survey 2007-2017.

Ten years of nationally representative biomonitoring data collected between 2007 and 2017 are available from the Canadian Health Measures Survey (CHMS). These data establish baseline environmental chemical concentrations in the general population. Here we sought to evaluate temporal trends in environmental chemical exposures in the Canadian population by quantifying changes in biomarker concentrations measured in the first five two-year cycles of the CHMS. We identified 39 chemicals that were measured in blood or urine in at least three cycles and had detection rates over 50% in the Canadian p

Bisphenol S-Induced Neurobehavioral Impairment Is Characterized by c-Jun Activation and Distinct Dysregulation of Nrf2 and BDNF in the Zebrafish Brain.

Bisphenol S (BPS), a widely used substitute for bisphenol A (BPA), is increasingly detected in consumer products and aquatic environments, yet its neurotoxic potential remains insufficiently understood. Building on our previous findings that BPS induces anxiety-like behaviour and oxidative damage in zebrafish, the present study investigates the mechanistic basis of BPS-mediated neurotoxicity with a focus on stress-activated kinase signalling, antioxidant defences, and neurotrophic regulation. Adult zebrafish were exposed to a sub-lethal concentration of BPS (63.93 µM) for 7, 14, and 21 days, f

Implications of early-life BPS-induced thyroid disruption on adult female reproductive disorders.

Reproductive disorders have become a growing public health concern, with early-life environmental exposures increasingly recognized as critical determinants of long-term reproductive outcomes. Among emerging endocrine disruptors, bisphenol S (BPS) raises concern due to its potential to interfere with hormone signaling during sensitive developmental windows. This study examined whether prenatal BPS exposure affects reproductive outcomes in adulthood through maternal thyroid disruption. Pregnant rats were exposed to BPS and the thyroid hormone receptor (THR) antagonist (AT 1-850). Key parameters

In vitro remineralization of severely compromised bonded dentin.

Biomimetic remineralization is potentially useful for the remineralization of incompletely resin-infiltrated collagen matrices created by etch-and-rinse adhesives. In this study, we tested the hypothesis that structurally altered dentin collagen cannot be remineralized to the same hierarchical order and dimension seen in structurally intact dentin collagen. The remineralization medium consisted of a set Portland cement/simulated body fluid system containing polycarboxylic acid and polyvinylphosphonic acid as biomimetic analogs. Remineralization of air-dried, collapsed hybrid layers was apparen

Bisphenol A-induced NF-κB activation enhances proliferation and migration of renal cell carcinoma cells via SLC7A11 upregulation and redox remodeling.

Bisphenol A (BPA) is a ubiquitous endocrine-disrupting chemical, yet its influence on renal cell carcinoma (RCC) remains unclear. We examined whether low-dose BPA alters redox regulation linked to ferroptosis susceptibility in 786-O and ACHN cells. After concentration screening, 100 nM BPA was used for mechanistic experiments. BPA enhanced proliferation and migration, increased the glutathione redox ratio (GSH/GSSG), reduced reactive oxygen species (ROS) signals, and preserved mitochondrial membrane potential. Public cancer dataset analyses prioritized SLC7A11, which was upregulated by BPA at
