Calcium channel blockers affect angiogenesis by modulating inflammatory factors: a meta-analysis of randomized controlled trials.

INTRODUCTION: Neovascularization is balanced by various factors that stimulate and inhibit angiogenesis, thus ensuring the physiological formation of blood vessels. Pathological angiogenesis also occurs in the development of the disease. Calcium ions play an important role in the human body, and can participate in various functions in cell physiology, such as gene expression, control of the cell cycle, cell motility, apoptosis, and autophagy. Selective activation of transcription factors that can be used for gene transcription, cell proliferation, and migration (have deleted). Studies have fou

Anoctamin-1 Is a Calcium-Dependent Chloride Channel (CaCC) in Müller Cells Involved in Cell Volume Regulation.

Previous functional data showed the presence of Cl- channels in Müller cells, among them Ca2+-dependent Cl- channels (CaCC). An important step to understanding the role of CaCC in Müller cell function is the clarification of their molecular identity. Here, we found protein expression of the CaCC anoctamin-1 (Ano1, formerly TMEM16A) in Müller cells in retinal sagittal sections of mouse and rat using immunohistochemistry and proteomic analysis, as well as in the Müller cell lines ImM10 (murine) and MIO-M1 (human), accompanied by Western blot. Patch-clamp analysis of both cell lines under K+-free

Myometrial mechanotransduction and the timing of parturition: ion-channel pathways linking uterine stretch, calcium signaling, and inflammation.

Mechanical loading is a defining feature of pregnancy, yet its integration with ion-channel physiology in the myometrium remains incompletely synthesized. This Review examines how uterine stretch, overdistension, oxytocin-associated signaling, extracellular-matrix context, and inflammatory cues are translated into Ca²+ entry, membrane excitability, inflammatory amplification, and contraction timing in the pregnant myometrium. It distinguishes candidate direct mechanosensors from mechanically regulated Ca²+/inflammatory coupling pathways and from adjacent excitability regulators, with the stron

TRPV4 channel-mediated calcium signalling in human lens epithelium of cataract patients and healthy lens capsules.

PURPOSE: Transient receptor potential vanilloid 4 (TRPV4) channels are mechanosensitive ion channels implicated in Ca2+ signalling across various tissues, yet their functional role in human lens epithelial cells remains poorly understood. We aimed to investigate the contribution of TRPV4 to mechanically induced Ca2+ signalling in human anterior lens epithelium. METHODS: Human anterior lens epithelial preparations were obtained from postoperative lens capsules, including both cataractous and non-cataractous samples. Using multicellular Ca2+ imaging, we analysed the spatiotemporal properties of

RRAD-reduction reveals efficacy of targeting L-type calcium channel regulation for treatment of heart failure.

AIMS: Heart failure with reduced ejection fraction (HFrEF) is a major health problem. Increasing L-type calcium channel (LTCC) activity deteriorates heart function; however, myocardial RRAD knockout (cRADΔ/Δ) instills tonic modulated LTCC current (ICa,L) that preserves healthy myocardium. Thus, we chose to challenge the dogma that enhanced trigger Ca2+ is maladaptive. The study objective was to test the hypothesis that modulated ICa,L in cRADΔ/Δ mice rescues dilated cardiomyopathy by providing tonic modulated trigger Ca2+. METHODS AND RESULTS: Mouse and human models were tested. The muscle li

Alantolactone mitigates the elevation of blood pressure in mice induced by angiotensin II by inhibiting calcium channel activation.

BACKGROUND: The dried root of Inula helenium L., known as Inulae Radix in Mongolian medicine, is a widely used heat-clearing plant drug with … Molecular docking experiments showed that ATL had a high binding affinity with L-type Voltage-gated Calcium Channels (VGCC), and vascular ring experiments indicated that ATL could significantly inhibit the vascular contractions caused by the agonists of L-type VGCC. In addition, we also observed that ATL had an ameliorative effect on the vascular remodeling induce …

UHPLC-MS/MS method for the simultaneous quantification of five calcium channel antagonists' drugs in human plasma.

One of the principal challenges in managing hypertension is suboptimal medication adherence, which can result in up to 50 % of patients experiencing pseudo-resistant hypertension and inadequate blood pressure control. The intricate nature of antihypertensive regimens, the high cost of therapy, the prevalence of numerous adverse effects and the therapeutic inertia commonly lead to inconsistent drug use and premature discontinuation of treatment. Consequently, nonadherence significantly elevates the risk of cardiovascular events. Drug concentrations in the patient's serum or urine (also known as

Association Between Body Mass Index and the Efficacy of Calcium Channel Blockers for Hypertension in Cardiovascular Disease Patients.

BACKGROUND: Hypertension, or high blood pressure, is a major risk factor for cardiovascular diseases (CVDs) worldwide. Variations in body mass index (BMI) may influence the efficacy of calcium channel blockers (CCBs) by affecting drug metabolism, vascular resistance, and inflammatory responses associated with adipose tissue. OBJECTIVE: This study aims to evaluate the association between BMI and the short-term efficacy of CCBs in managing hypertension among patients with CVDs over a six-month follow-up period. METHODOLOGY: This prospective observational study was conducted at the Department o

Calcium Channel α2δ Ligands Mirogabalin, Pregabalin, and Gabapentin: Advancements in Diabetic Peripheral Neuropathic Pain Therapeutics.

Diabetic peripheral neuropathic pain (DPNP) is becoming increasingly prevalent as the global burden of diabetes continues to rise. DPNP manifests moderate-to-severe pain with burning, shooting, and tingling sensations, which increase clinical and economic burden and reduce the quality of life (QoL). α2δ ligands were developed on the basis of their mechanism of action involving the modulation of voltage-gated calcium channels (VGCCs). These ligands bind to the α2δ subunit of VGCCs, which reduces calcium influx and subsequently decreases the release of excitatory neurotransmitters. A majority of…

Inappropriate prescription of loop diuretics for calcium channel blocker-related peripheral edema: Lessons from physiology.

Calcium Channel Blockers (CCB) are commonly used antihypertensive agents, with peripheral edema being a common side effect, reported in up to 70 % of cases. A common pitfall among physicians is that they consider this fluid overload, with some prescribing loop diuretics. Nonetheless, this might lead to several complications, such as electrolyte abnormalities and acute kidney injury, among others. In this article, we describe the main mechanisms of the vasodilatory and diuretic-resistant edema of CCB and suggest several strategies that might aid physicians in reducing this side effect.

The effect of the calcium channel blocker nimodipine on hippocampal BDNF/Ach levels in rats with experimental cognitive impairment.

OBJECTIVE: Alzheimer's disease (AD) occurs in approximately 10% to 30% of individuals aged 65 or older worldwide. Novel therapeutic agents therefore need to be discovered in addition to traditional medications. Nimodipine appears to possess the potential to reverse cognitive impairment-induced dysfunction in learning and memory through its regulatory effect on the brain-derived neurotrophic factor (BDNF), acetylcholine (Ach), and acetylcholinesterase (AChE) pathway in the hippocampus and prefrontal cortex. METHODS: Twenty-four male Sprague Dawley rats weighing 380 ± 10 g were used for behavio

Exploring calcium channel blocker as a candidate drug for Pseudomonas aeruginosa through network pharmacology and experimental validation.

Calcium channel blocker (CCB) is known to possess antibacterial effects. We aimed to apply network pharmacology (docking and protein-protein interaction [PPI] analyses) to predict the potential targets and mechanisms of CCB against Pseudomonas aeruginosa (PA) as well as to verify the effect of these drugs. The chemical structures of three CCBs were obtained through the Drug Bank platform. The potential channel proteins, efflux pump proteins and ion channel proteins of CCB against bacteria were derived from the literature. These proteins were collected through the PDB and Uniprot platform. The

Calcium channel blockers are associated with improved survival in pancreatic cancer patients undergoing neoadjuvant chemotherapy and resection.

BACKGROUND: Repurposing existing drugs for use in oncology is more efficient, cost-effective and safe than novel drug discovery. Calcium signalling is increasingly recognised to have a key role in chemoresistance. This study assessed the impact of calcium channel blockers (CCB) in pancreatic cancer. METHODS: Retrospective population study of patients undergoing resection (curative intent) of pancreatic ductal adenocarcinoma (SEER-Medicare, 2007-2017). Cox models were built to assess the impact on overall survival. As laboratory studies suggest a chemosensitising effect, the impact of CCB was

Calcium channel blockers for preventing cardiomyopathy due to iron overload in people with transfusion-dependent beta thalassaemia.

BACKGROUND: Beta-thalassaemia is an inherited blood disorder that reduces the production of haemoglobin. The most severe form requires recurrent blood transfusions, which can lead to iron overload. Cardiovascular dysfunction caused by iron overload is the leading cause of morbidity and mortality in people with transfusion-dependent beta-thalassaemia. Iron chelation therapy has reduced the severity of systemic iron overload, but removal of iron from the myocardium requires a very proactive preventive strategy. There is evidence that calcium channel blockers may reduce myocardial iron deposition
