[Stem cell-based treatment of neurologic diseases].

Therapeutic strategies based on stem cells are being increasingly used to treat a wide range of neurological diseases. Although these strategies were initially designed to replace dead cells in injured tissue, the potential of stem cells to migrate, secrete trophic factors, and immunomodulate allows their therapeutic use as a vehicle for gene therapy, as in Parkinson's disease, or as immunomodulators and neuroprotectors in diseases such as multiple sclerosis. This review will focus on current clinical and experimental evidence on the treatment of neurological disorders with strategies based on

Cardiac tissue engineering for the treatment of hypoplastic left heart syndrome (HLHS).

Hypoplastic left heart syndrome (HLHS) is a deadly congenital heart disease that arises when the left ventricle and outflow tract fail to develop appropriately, inhibiting the adequate perfusion of the rest of the body. Historically, this disease has been treated via a series of surgeries that allows the heart to use a single ventricle. These surgeries are often a palliative measure, and heart transplantation is the only definitive therapy that exists for this condition. It has been hypothesized that stem cell-based regenerative therapies could have a role in promoting cardiac tissue regenerat

Adipose-derived stem cells seeded in Pluronic F-127 hydrogel promotes diabetic wound healing.

BACKGROUND: Chronic nonhealing wound is a multifactorial complication of diabetes that results specifically as a consequence of impaired angiogenesis and currently lacks in effective treatments. Although a stem cell-based therapy may provide a novel treatment to augment diabetic wound healing, inferior cell survival at the diabetic skin wound is one of the key causes that are responsible for the low efficacy of the stem cell therapy. MATERIALS AND METHODS: In this work, we used an injectable, biocompatible, and thermosensitive hydrogel Pluronic F-127 to encapsulate allogeneic nondiabetic adip

Melatonin-A Potent Therapeutic for Stroke and Stroke-Related Dementia.

Secreted by the pineal gland to regulate the circadian rhythm, melatonin is a powerful antioxidant that has been used to combat oxidative stress in the central nervous system. Melatonin-based therapies have been shown to provide neuroprotective effects in the setting of ischemic stroke by mitigating neuroinflammation and accelerating brain tissue restoration. Melatonin treatment includes injection of exogenous melatonin, pineal gland grafting and melatonin-mediated stem cell therapy. This review will discuss the current preclinical and clinical studies investigating melatonin-based therapeutic

Neural and mesenchymal stem cells in animal models of Huntington's disease: past experiences and future challenges.

Huntington's disease (HD) is an inherited disease that causes progressive nerve cell degeneration. It is triggered by a mutation in the HTT gene that strongly influences functional abilities and usually results in movement, cognitive and psychiatric disorders. HD is incurable, although treatments are available to help manage symptoms and to delay the physical, mental and behavioral declines associated with the condition. Stem cells are the essential building blocks of life, and play a crucial role in the genesis and development of all higher organisms. Ablative surgical procedures and fetal ti

Nanomedicine-Based Therapeutic Strategies for Next-Generation Cancer Immunotherapy.

Cancer immunotherapy has reshaped cancer treatment by enabling the immune system to recognize and eliminate tumor cells. Nevertheless, its clinical translation remains limited by suboptimal response rates, poor penetration into deep tumor tissues, and insufficient therapeutic efficacy. Cancer immunotherapy alone is often inadequate as a sole treatment modality. Nanomedicine-based therapeutic strategies, including photothermal therapy, photodynamic therapy, sonodynamic therapy, nanozyme-based therapy, and cancer vaccines, have achieved considerable recognition for cancer treatment. Upon integra

Super-stable two-dimensional radical conjugated metal-organic frameworks for efficient NIR-II photothermal conversion.

Radical-functionalized chromophores hold promise as noninvasive NIR-II agents due to their narrow HOMO-SOMO gaps, but inherent instability from radical quenching has limited their practical applications. Here, we report a molecularly engineered class of two-dimensional conjugated metal-organic frameworks (2D c-MOFs) that simultaneously integrate stable radicals (>24 months), extended π-delocalization, and metal-ligand coordination. This synergistic architecture achieves an ultrabroad NIR-II absorption peak extending beyond 1400 nm with extinction coefficients approaching ~106 M-1 cm-1, which o

Role of mesenchymal stem cells in meniscal repair.

Meniscus integrity is the key for joint health of the knee. Therefore, the main goal of every meniscus treatment should be the maintenance of as much meniscus tissue as possible.Repair of meniscus tears can be achieved by meniscus suture. However, in a recently published meta-analysis, the long-term outcome of meniscus repair showed a mean failure rate of 24%.In a preclinical trial, locally applied mesenchymal stem cells produced differentiated meniscus-like tissue in meniscus tears indicating that mesenchymal-based cells, harvested from the bone marrow, enhance meniscus healing in critical-si

Disease-derived circulating extracellular vesicle preconditioning: A promising strategy for precision mesenchymal stem cell therapy.

Mesenchymal stem cell (MSC)-based therapies have emerged as promising methods for regenerative medicine; however, how to precisely enhance their tissue repair effects is still a major question in the field. Circulating extracellular vesicles (EVs) from diseased states carry diverse pathological information and affect the functions of recipient cells. Based on this unique property, we report that disease-derived circulating EV (disease-EV) preconditioning is a potent strategy for precisely enhancing the tissue repair potency of MSCs in diverse disease models. Briefly, plasma EVs from lung or ki

Successful treatment of Scedosporium apiospermum soft tissue abscess with caspofungin and voriconazole in a severely immunocompromised patient with acute myeloid leukemia.

We report the case of a 53-year-old female patient with refractory acute myeloid leukemia developing a necrotic, soft tissue abscess on the right forearm caused by Scedosporium apiospermum during prolonged severe neutropenia (absolute white blood cell count <500/μL for 49 days). In the context of the severely immunocompromised state of the patient and her need for allogeneic hematopoietic stem cell transplantation (HSCT), surgical treatment options were not favored. Therefore, combined antifungal therapy with voriconazole and caspofungin was started, based on the results of the in vitro testin

Future Perspectives of Wharton's Jelly Mesenchymal Stem Cells and their Soluble Factors in Radioprotection.

Acute radiation syndrome (ARS) is also known as triple syndrome, which develops after whole-body radiation exposure. During unforeseen exposures, these syndromes are set in depending on the dose of radiation. Cell-based therapy, especially using stem cells and their soluble factors, is gaining wide attention in the field of regenerative medicine to treat various diseases, including degenerative diseases. Stem cells attract prime attention because of their profound inherent tissue repair capability and regeneration potential. Further, stem cell therapy can be one of the promising strategies for

Development of poly(ethylene glycol) hydrogels for salivary gland tissue engineering applications.

More than 40,000 patients are diagnosed with head and neck cancers annually in the United States with the vast majority receiving radiation therapy. Salivary glands are irreparably damaged by radiation therapy resulting in xerostomia, which severely affects patient quality of life. Cell-based therapies have shown some promise in mouse models of radiation-induced xerostomia, but they suffer from insufficient and inconsistent gland regeneration and accompanying secretory function. To aid in the development of regenerative therapies, poly(ethylene glycol) hydrogels were investigated for the encap

Exploring the Potential of Stem Cell-Based Therapy for Aesthetic and Plastic Surgery.

Over the last decade, stem cell-associated therapies are widely used because of their potential in self-renewable and multipotent differentiation ability. Stem cells have become more attractive for aesthetic uses and plastic surgery, including scar reduction, breast augmentation, facial contouring, hand rejuvenation, and anti-aging. The current preclinical and clinical studies of stem cells on aesthetic uses also showed promising outcomes. Adipose-derived stem cells are commonly used for fat grafting that demonstrated scar improvement, anti-aging, skin rejuvenation properties, etc. While stem

Therapeutic Potential of Stem Cells in the Treatment and Management of Immunomodulatory Disorders.

INTRODUCTION: Immunomodulatory disorders, such as autoimmune diseases, inflammatory conditions, and viral infections, stem from immune system dysregulation and often resist conventional therapies. Stem cells, particularly mesenchymal (MSCs) and hematopoietic stem cells (HSCs), possess immunomodulatory and regenerative properties, offering a promising therapeutic alternative. METHODOLOGY: A systematic literature review was conducted using databases, including PubMed, Scopus, Web of Science, and Google Scholar, for studies published between 1996 and 2025. A total of 287 articles were screened,

Cytomegalovirus-specific cytokine-induced killer cells: concurrent targeting of leukemia and cytomegalovirus.

BACKGROUND AIMS: Human cytomegalovirus (CMV) infection and reactivation is a leading complication of allogeneic hematopoietic stem cell transplantation (HSCT). In addition to drug treatment, the adoptive transfer of virus-specific T cells to restore cellular immunity has become a standard therapy after allogeneic HSCT. We recently demonstrated potent anti-leukemic activity of interleukin (IL)-15-activated cytokine-induced killer (CIK) cells. With the use of the same expansion protocol, we asked whether concurrent CMV antigen-pulsing might generate CIK cells with anti-leukemic and anti-CMV acti
