A database of polymorphisms in the von Willebrand factor gene and pseudogene. For the Consortium on von Willebrand Factor Mutations and Polymorphisms and the Subcommittee on von Willebrand Factor of the Scientific and Standardization Committee of the International Society on Thrombosis and Haemostasis.

Nucleotide sequence polymorphisms in the von Willebrand factor (vWF) gene are useful for genetic studies in von Willebrand disease (vWD). This database describes 33 known vWF polymorphisms distributed throughout the vWF gene. DNA sequence information is available for 21 of these sites. The most informative system is a tetranucleotide repeat polymorphism in vWF intron 40. Sixteen of these polymorphisms are within vWF exons, and approximately half of them also alter the encoded amino acid sequence. Many occur close to mutations that cause vWD. The high prevalence of vWF polymorphisms must be con

Distinct features of three clinical subtypes in 533 patients with primary hypertrophic osteoarthropathy.

BACKGROUND: Primary hypertrophic osteoarthropathy (PHO) is a rare genetic disorder classified into clinical subtypes and genetic subtypes. Previous clinical studies have primarily focused on case reports and family analyses, largely characterizing the genetic subtypes. However, there remains a long-standing gap in understanding the characteristics of the different clinical subtypes of PHO. This study aimed to determine the distribution of the three clinical subtypes of PHO and compare their clinical characteristics using a large global sample. METHODS: A systematic literature search was condu

IDDB: a comprehensive resource featuring genes, variants and characteristics associated with infertility.

Infertility is a complex multifactorial disease that affects up to 10% of couples across the world. However, many mechanisms of infertility remain unclear due to the lack of studies based on systematic knowledge, leading to ineffective treatment and/or transmission of genetic defects to offspring. Here, we developed an infertility disease database to provide a comprehensive resource featuring various factors involved in infertility. Features in the current IDDB version were manually curated as follows: (i) a total of 307 infertility-associated genes in human and 1348 genes associated with repr

Estimating familial and genetic contributions to variability in human testicular function: a pilot twin study.

The biological basis for the remarkably wide variation in sperm output between and within men remains unclear. Although some contributing factors have been identified, the familial and genetic contributions to variation in human sperm output have been little studied, although such sources of variation are known to be significant in experimental animals. In order to identify such familial and genetic factors in a classical twin study design, we recruited monozygotic and dizygotic twins aiming to study the degree of resemblance between genetically identical and non-identical twins in sperm outpu

Identification of Shared Gene Signatures Associated with Alzheimer's Disease and COVID-19 through Bioinformatics Analysis.

INTRODUCTION: Some studies have shown a link between Alzheimer's disease (AD) and COVID-19. This includes a Mendelian randomization study, which suggests that Alzheimer's disease and COVID-19 may be causally linked in terms of pathogenic mechanisms. However, there are fewer studies related to the two in terms of common pathogenic genes and immune infiltration. We conducted this study to identify key genes in COVID-19 linked to Alzheimer's disease, assess their relevance to immune cell profiles, and explore potential novel biomarkers. METHODS: The RNA datasets GSE157103 and GSE125583 for COVID

WiNGS-API: a federated genome/phenome data sharing platform enabling gene discovery and variant classification for rare diseases.

BACKGROUND: Genomic research on rare diseases requires large-scale data sharing to identify pathogenic variants. Traditionally, this is achieved through centralized databases that require the physical transfer of genomic data to third-party servers. However, this approach presents significant challenges under data protection frameworks like the General Data Protection Regulation (GDPR), which considers genomic data inherently identifiable and imposes strict limitations on data sharing. RESULTS: To address these limitations, WiNGS-API introduces a federated, automated, and privacy-compliant pla

Multidisciplinary analysis of the prognosis and biological function of NUBPL in gastric cancer.

BACKGROUND: Researchers are currently concentrating on molecular markers and potential therapeutic targets associated with gastric cancer in light of recent developments in precision medicine and molecular biology. Disulfidptosis was first proposed in 2023 as a novel programmed cell death mode associated with the cytoskeleton. Disulfidptosis-related proteins are essential for the preservation of protein stability and abnormal expression of disulfidptosis-related genes may be linked to cancer development and drug resistance. MATERIALS AND METHOD: The gastric cancer transcriptomic data were ret

New Diagnostic and Therapeutic Targets for Spinal Cord Injury: GRN Gene.

Spinal cord injury (SCI) is a severe disabling disease due to the poor self-healing of the central nervous system. Studies showed that many N6-methyladenosine (m6A) RNA methylation profiles are hypomethylated after SCI, which are related to neural regeneration and different m6A marker genes. In addition, immune cell infiltration may significantly affect the development and progression of SCI. Therefore, we attempted to identify the correlation between SCI-related biomarkers and m6A methylation regulators in order to classify them. To this end, we collected two gene expression profile datasets

Functional Insights Through Gene Ontology, Disease Ontology, and KEGG Pathway Enrichment.

Understanding gene function and pathways is a key goal in genomics. Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) provide frameworks to annotate genes based on their roles, cellular locations, and functions. These classifications help researchers analyze complex systems, identify drug targets, and investigate gene-disease connections. GO and KEGG annotations are central to enrichment analysis, which tests for overrepresented pathways in gene sets, often comparing disease to healthy states. Enrichment analysis methods include overrepresentation analysis (ORA) for gene li

The validity of immunotherapy against palate mucoepidermoid carcinoma based on data enrichment from oral cancer database.

BACKGROUND: Palate mucoepidermoid carcinoma (PMEC) is an infrequent but tough subtype of oral carcinoma. Due to the lack of independent and comprehensive database for PMEC, the selection or screening of targeted protein on PMEC might be inaccurate. However, several databases of gene expression based on oral cancers having been constructed and uploaded. With the help of combining data enrichment and lab experiments, the target protein choosing against PMEC could be available. METHOD: We firstly searched multiple databases, which being relevant with oral cancers, and compared the gene expressio
