BACKGROUND: The accumulation and propagation of α-synuclein (α-syn) are hallmark features of Parkinson's disease (PD) and related neurodegenerative disorders. O-GlcNAcylation, an abundant post-translational modification throughout the brain, is regulated by the enzymatic activity of the cycling enzymes O-GlcNAc transferase (OGT) and O-GlcNAcase (OGA) and has been implicated in altering α-syn toxicity. Nevertheless, the interplay between modulating O-GlcNAc cycling and α-syn aggregation and the propagation of amyloid pathology is not well elucidated.
METHODS: To this end, we delivered conforma

BACKGROUND: RNA modifications, collectively known as the epitranscriptome, represent the third layer of gene regulation, influencing gene expression at transcriptional, post-transcriptional, and translational levels. RNA-modifying proteins (RMPs), including writers, erasers, and readers, are responsible for depositing, removing, and recognizing chemical modifications on RNA molecules. These modifications play a crucial role in linking molecular processes to cellular functions. Over the past few decades, a growing body of laboratory evidence, alongside advances in sequencing technologies, has u

As a lactate-derived post-translational modification, lactylation plays a vital role in connecting cellular glycolysis with the regulation of cuproptosis, a unique form of cell death triggered by copper overload. The article explores the growing relationship between lactylation and cuproptosis, highlighting the potential for lactate metabolism to influence cellular responses to copper toxicity. We further discuss the potential roles of lactylation writers, erasers and readers in modulating key cuproptosis-related proteins, such as FDX1, DLAT and CTR1, through the specific actions of molecular

Paclitaxel (PTX) is a core first-line chemotherapeutic agent used to treat a broad spectrum of cancers, which remain leading causes of death worldwide. However, the emergence of intrinsic and acquired resistance critically contributes to chemotherapy failure, thereby promoting tumor recurrence and metastasis and resulting in a poor prognosis. PTX resistance is a complex biological process driven by multiple factors and cross-regulated signaling pathways. It encompasses a wide variety of mechanisms, including aberrations in microtubules and related proteins; changes in mitotic systems and chrom

Chronic kidney disease (CKD) is associated with high cardiovascular risk. CKD patients exhibit a specific lipoprotein pattern termed 'uraemic dyslipidaemia', which is characterized by rather normal low-density lipoprotein cholesterol, low high-density lipoprotein cholesterol, and high triglyceride plasma levels. All three lipoprotein classes are involved in the pathogenesis of CKD-associated cardiovascular diseases (CVDs). Uraemia leads to several modifications of the structure of lipoproteins such as changes of the proteome and the lipidome, post-translational protein modifications (e.g. carb

In Alzheimer's disease and other tauopathies, tau displays several abnormal post-translation modifications such as hyperphosphorylation, truncation, conformation, and oligomerization. Mouse monoclonal antibodies have been raised against such tau modifications for research, diagnostic, and therapeutic purposes. However, many of these primary antibodies are at risk of giving nonspecific signals in common Western blotting procedures. Not because they are unspecific, but because the secondary antibodies used to detect them will also detect the heavy chain of endogenous mouse immunoglobulins (Igs),

Metabolic remodeling is one of the hallmarks of malignant tumors, and post-translational modifications of proteins, such as succinylation, play an important role in the process of metabolic remodeling by regulating the function of metabolic enzymes. However, the role of succinylation modifications of key mitochondrial metabolic enzymes in lung adenocarcinoma (LUAD) remains unclear. Here, using succinylation proteomics, it is demonstrated that the succinylation level of lysine 409 (K409) on dihydrolipoamide S-succinyltransferase (DLST) is significantly increased in LUAD. Further, DLST K409 succ

Gastric Cancer (GC) is a highly prevalent and poorly prognostic gastrointestinal malignancy with low overall treatment efficacy worldwide. Early diagnostic markers and potential therapeutic targets for GC treatment are urgently needed. UFMylation, a novel ubiquitin-like modification is indispensable for numerous fundamental cellular processes. Deficiency in this modification is reported to be associated with several human diseases including cancer. Accumulating evidence suggests that the expression of the key UFMylation components is closely associated with GC cell proliferation, invasion, met

Histone post-translational modifications (PTMs) are crucial to eukaryotic genome regulation, with a range of reported functions and mechanisms of action. Though often studied individually, it has long been recognized that the modifications function by combinatorial synergy or antagonism. Interplay may involve PTMs on the same histone, within the same nucleosome (containing a histone octamer), or between nucleosomes in higher-order chromatin. Given this, the field must distinguish ever greater complexity, and the context in which it is studied, with brevity and precision. The proteoform was int

Epigenetic modifications in histones are crucial for proper sperm physiology, egg activation and reproductive development of males. The objectives of this study were to determine the conservation and interactomes of histone three (H3) and ascertain the expression dynamics of acetylated and methylated H3 lysine 27 (H3K27ac and H3K27me3) in spermatozoa from Holstein bulls with different fertility. Methods in immunocytochemistry and flow cytometry were used to evaluate the expression dynamics of H3K27ac and H3K27me3 in spermatozoa from 10 bulls with different in vivo fertility. BACKGROUND: Protein arginine methyltransferase 9 (PRMT9) is dysregulated in various malignancies, particularly in lung adenocarcinoma (LUAD). This study aims to systematically investigate the expression patterns, biological functions, and underlying molecular mechanisms of PRMT9 in LUAD pathogenesis.
METHODS: PRMT9 expression was evaluated in paired clinical LUAD specimens and adjacent normal tissues, as well as in normal alveolar epithelial cells versus established lung cancer cell lines. Genetic silencing of PRMT9 was performed in A549 and H1568 cell models to assess its functional impact.

RNA metabolic dysregulation is a key pathological mechanism underlying the onset and progression of Alzheimer's disease (AD), involving multiple aspects such as abnormal RNA splicing, loss of function in RNA-binding proteins, dysregulation of non-coding RNAs, and impaired nuclear-cytoplasmic transport. In recent years, the emergence of epigenome research has revealed the critical role of RNA chemical modifications in regulating RNA metabolism at the post-transcriptional level. N4-acetylcytidine (ac4C) is the only known RNA acetylation modification in eukaryotes and is specifically catalyzed by

Proteolytic processing of precursor proteins liberates bioactive fragments, yet the systematic discovery of such peptides remains challenging. Here, we establish a fragment-mining strategy that combines in silico prediction with chemical tailoring to generate self-assembling antimicrobial peptides directly from protein sequences, including previously unannotated proteins. Self-assembly proved integral to activity: amphiphilic modification promoted nanonet formation and efficient bacterial membrane disruption, while rational glycosylation reprogrammed physicochemical properties to confer select

Acute myeloid leukemia (AML) is a heterogeneous and aggressive malignancy with limited therapeutic options and high relapse rates. Despite advances in genomic profiling, many genetic aberrations remain untargetable, and current risk stratification models often fail to predict treatment responses. Proteomics offers a complementary approach by directly measuring protein abundance, post-translational modifications, and protein-protein interactions, providing mechanistic insights into drug resistance, disease progression, and therapeutic vulnerabilities. In this review, we explore the emerging rol

Aberrant aggregation of β-amyloid (Aβ) peptides into insoluble fibrils is recognized as one of the hallmarks of Alzheimer's disease (AD). Among the post-translational modifications influencing Aβ behavior, nitration and nitrosation by nitric oxide (NO) derivatives play a key role, though their impact on aggregation and toxicity remains unclear. This contribution explores the effects on Aβ aggregation induced by an NO photodonor (NOPD) releasing two NO molecules via a stepwise mechanism under the control of visible blue light. Significant reduction of protein aggregation is observed when a cons

Bacterial pathogens often make use of post-translational modifications to manipulate host cells. Legionella pneumophila, the causative agent of Legionnaires disease, secretes the enzyme AnkX that uses cytidine diphosphate-choline to post-translationally modify the human small G-Protein Rab1 with a phosphocholine moiety at Ser76. Later in the infection, the Legionella enzyme Lem3 acts as a dephosphocholinase, hydrolytically removing the phosphocholine. While the molecular mechanism for Rab1 phosphocholination by AnkX has recently been resolved, structural insights into the activity of Lem3 rema

The SLC25A20 transporter, also known as carnitine acyl-carnitine carrier (CAC), catalyzes the transport of short, medium and long carbon chain acyl-carnitines across the mitochondrial inner membrane in exchange for carnitine. The 30-year story of the protein responsible for this function started with its purification from rat liver mitochondria. Even though its 3D structure is not yet available, CAC is one of the most deeply characterized transport proteins of the inner mitochondrial membrane. Other than functional, kinetic and mechanistic data, post-translational modifications regulating the

Lysine malonylation (Kmal) is a new post-translational modification (PTM), which has been reported in several prokaryotic and eukaryotic species. Although Kmal can regulate many and diverse biological processes in various organisms, knowledge about this important PTM in the apicomplexan parasite Toxoplasma gondii is limited. In this study, we performed the first global profiling of malonylated proteins in T. gondii tachyzoites using affinity enrichment and Liquid chromatography-tandem mass spectrometry (LC-MS/MS) analysis. Three experiments performed in tandem revealed 294, 345, 352 Kmal sites

Huntington's disease is a dreadful, incurable disorder. It springs from the autosomal dominant mutation in the first exon of the HTT gene, which encodes for the huntingtin protein (HTT) and results in progressive neurodegeneration. Thus far, all the attempted approaches to tackle the mutant HTT-induced toxicity causing this disease have failed. The mutant protein comes with the aberrantly expanded poly-glutamine tract. It is primarily to blame for the build-up of β-amyloid-like HTT aggregates, deleterious once broadened beyond the critical ∼35-37 repeats threshold. Recent experimental findings
