Absorption of phenoxymethylpenicillin in children. The influence of age, state of disease and pharmaceutical preparation.

The bioavailability of phenoxymethylpenicillin (pc V) was determined in children with upper respiratory infection (age 1 month-5 years) following the administration of different commercial preparations of pc V. Administration of potassium pc V in water solution resulted in a significantly higher plasma concentration than the same dose of potassium pc V in an oil vehicle. There was no difference in absorption between tablet and mixture (the water solution) when potassium pc V was given, but with calcium pc V lower plasma levels were found after the oil mixture form than after the tablets. The r…

[Jejunal mucosa in infants with the syndrome of disorders of intestinal absorption (ultrastructure and morphometry)].

Ultrastructural studies and counts of interepithelial lymphocytes and the density of cellular infiltration of the lamina propria were carried out on 10 biopsies of the small intestine from infants of 4 months to 2 years with the syndrome of disturbed intestinal absorption. The greatest disorders were found to occur in celiac disease and intolerance to cow milk protein and to be manifested in subtotal atrophy of villi and destructive lesions of enterocytes. The authors believe that the marked infiltration of the epithelial layer with lymphocytes and the lamina propria of the mucous membrane wit…

Determination of aluminum in biological samples by atomic absorption spectrophotometry with a graphite furnace.

Aluminum, generally considered non-essential and non-toxic, may accumulate in toxic amounts in the brain in cases of chronic renal failure. We describe a procedure for its analysis in biological fluids by atomic absorption spectrophotometry with a graphite furnace. No sample preparation is required and the procedure is sensitive at the appropriate concentrations. A sample of serum or urine is pipetted into the interior of the graphite tube, where it is sequentially dried, charred, and atomized. Precautions for sample handling are discussed and instrument settings are defined. Precision and acc…

Orogastric determinants of drinking in rats: interaction between absorptive and peripheral controls.

Water intake of water-deprived rats, drinking with absorption prevented by pyloric occlusion, was compared with that of rats drinking normally. Drinking without absorption, under the control of only orogastric factors, was not graded in response to varied deprivation (Experiment 1). Orogastric controls effectively inhibited intake after absorbed water, but not isotonic saline, preloads (Experiment 2). The extent of orogastric inhibition was directly related to absorbed preload volume (Experiment 3). With large absorbed water preloads (9 ml), intakes with and without absorption were equal. Norm…

Magnesium absorption in the caecum of rats related to volatile fatty acids production.

Rats were fed with a semi-synthetic diet with or without raw potato starch. (The level of minerals was the same in the two diets.) Because o … ce in VFA and Mg in the caecum. Nutritional balance study was performed and confirmed the better Mg absorption in rats fed on the raw potato starch diet. The lower parts of the digestive tract seem to play an important role in Mg absorption and further studies on the relations between caecal volume, pH, VFA, microbial activity and Mg absorption are necessary …

The different effects of sodium bicarbonate and aluminium hydroxide on the absorption of indomethacin in man.

The influence of oral sodium bicarbonate and aluminium hydroxide on the absorption of indomethacin has been studied in normal volunteers. While sodium bicarbonate appeared to increase indomethacin absorption, aluminium hydroxide markedly reduced both the rate and extent of absorption. The buccal absorption of indomethacin over the pH range 5-9 was also studied in normal volunteers, and showed that the percentage absorption increased markedly as the pH was reduced. The clinical importance both of pH-partition and of adsorption are discussed in the context of antacid interactions. It is conclude…

Rapid glucose absorption in Arabs in Saudi Arbia compared with that in Africans in Zambia.

Glucose absorption rates in 14 Arabs in Saudi Arabia were determined by a jejunal perfusion technique and compared with those in 18 Africans in Zambia. None of the subjects had clinical evidence of systemic infection, gastrointestinal disease, malnutrition, or diabetes mellitus. The mean absorption rate was significantly higher in the Arabs. This was associated with a higher incidence of subclinical chronic systemic infection in the Africans. Other factors possibly responsible for the difference, including genetic difference, have, however, not been excluded.

Effect of dietary lactose on the absorption of protein, fat and calcium in the postweaning rat.

To assess the effect of dietary lactose on the intestinal absorption of protein, fat and calcium in lactose intolerance due to low levels of intestinal lactase, balance studies were carried out on postweaning rats (initial weight 164 g) for a period of 10 days. The experimental diets contained either 10% or 30% lactose, while in the control diets the lactose was replaced with equivalent amounts of sucrose. The excretions of fecal and urinary nitrogen, calcium and fat are expressed as precent of intake over the 10-day period. The fecal excretion of nitrogen and fat was significantly higher in t…

[Development of an oral retard preparation of verapamil and absorption study in the anesthetized dog].

1. An oral retard preparation of verapamil (Isoptin retard) is described. The sustained release of the active compound from verapamil retard … imilar times of release are found. 4. The in vitro results are supported by tests on the intestinal absorption in the anesthetized dog. In a ligated duodenum section verapamil is, 3 to 8 h after application, markedly more slowly released and absorbed from the retard preparation than from conventional dragées.

Transport into erythrocytes and intestinal absorption of uric acid in hereditary renal hypouricemia.

Uric acid transport into the erythrocytes and uric acid absorption from the intestine in a subject with hereditary defective renal tubular uric acid reabsorption was found to be normal. The findings are discussed in relation to the variability of phenotype expression in other renal tubular transport disorders.

[Absorption and pharmacokinetics of large doses of 1-thyroxine in man (author's transl)].

In recent years suppression tests with a single dose of 3 mg 1-thyroxine (T4) and weekly doses of 1 mg T 4 in the treatment of hypothyroidism have been put to clinical trial and the lack of side-effects of such high doses (5 to 15 X daily requirements) was stressed. Hence, it was decided to study the absorption of 1-T-4 from the gastrointestinal tract and its metabolism in euthyroid patients. Doses from 250 to 2500 mug T 4, mixed with 250 muCi 131I-thyroxine were given to 10 patients. After the thyroid had been blocked with perchlorate, the excretion was followed in faeces and urine for 4 to 5…

Amino acid and peptide absorption in patients with coeliac disease and dermatitis herpetiformis.

A double-lumen perfusion technique has been used to study amino acid and peptide absorption in eight normal control subjects, 13 patients with untreated adult coeliac disease, and 16 patients with dermatitis herpetiformis who had varying morphological abnormalities of the small bowel. All subjects were perfused with isotonic solutions containing 10 mM glycyl-L-alanine and 10 mM glycine + 10 mM L-alanine. Patients with adult coeliac disease had impaired absorption of glycine (p < 0.01) and L-alanine (p < 0.05) from the amino acid solution compared with the control subjects. Amino acid uptake fr…

Mechanism of bicarbonate absorption and its relationship to sodium transport in the human jejunum.

Using a constant perfusion technique, sodium and bicarbonate absorption was studied in human subjects. The following observations were made on sodium absorption from saline solution: (a) the rate of sodium absorption is markedly influenced by bulk water flow, (b) when net water flow is zero, sodium absorption is zero if there are no concentration gradients between plasma and lumen that favor net NaCl diffusion; and (c) the PD between abraded skin and jejunal lumen is near zero when saline is perfused and does not change with partial substitution of sulfate or bicarbonate for chloride. Based on…
