Acetylcholinesterase and luteinizing hormone-releasing hormone distinguish separate populations of terminal nerve neurons.

The terminal nerve is composed of a morphologically heterogeneous population of unipolar, bipolar and multipolar neurons located in the nasal and intracranial cavities of vertebrates. The question has arisen as to whether these neurons are neurochemically heterogeneous and therefore possibly functionally different as well. Among the substances localized in the terminal nerve are acetylcholinesterase and luteinizing hormone-releasing hormone-like immunoreactive material. We have developed a double-label procedure, combining immunocytochemistry and enzyme histochemistry to determine whether thes

Innervation of the posterior fossa dura of the cat.

This study was designed to identify the location of neurons giving rise to fibers innervating the posterior fossa dura in the cat using horseradish peroxidase (HRP, Sigma, Type VI). Investigations since the 19th century have implicated innervation by cranial nerves V, VII, IX, X and XII and the upper cervical nerves, C1-3. The meninges of the posterior fossa of 14 cats was exposed using one of three surgical approaches: (1) a suboccipital craniectomy and C1 laminectomy, (2) a parieto-occipital craniectomy with removal of the occipital lobe and bony tentorium exposing the meninges over the cere

Mechanisms underlying intracisternal TRH-induced stimulation of gastric acid secretion in rats.

The neurohumoral pathways mediating intracisternal TRH-induced stimulation of gastric acid secretion were investigated. In urethane-anesthetized rats, with gastric and intrajugular cannulas, TRH or the analog [N-Val2]-TRH (1 microgram) injected intracisternally increased gastric acid output for 90 min. Serum gastrin levels were not elevated significantly. Under these conditions the TRH analog, unlike TRH, was devoid of thyrotropin-releasing activity as measured by serum TSH levels. In pylorus-ligated rats, gastrin values were not modified 2 h after peptide injection whereas gastric acid output

Thiamine deficiency and nervous system function disturbances.

Thiamine is important for oxidative metabolism, and B1 deficiency is thought to give rise to polyneuropathies. A group of male Wistar rats (n = 15) received a vitamin B1 deficient diet (group-a), and the pair fed control group (n = 20, group-b) received a normal diet with no vitamin deficiency. A second control group (group-c) was fed unrestrictedly with a standard diet (n = 19). All animals were examined for 25 weeks. The sensory nerve conduction velocity, the compound radicular, spinal and brain stem responses and the SEP were derived for tail and hind paw stimulation. The examination was re

Immunohistochemical study of subclasses of olfactory nerve fibers and their projections to the olfactory bulb in the rabbit.

The organization of the olfactory nerve projection to the olfactory bulb was studied immunohistochemically in the rabbit by using monoclonal antibodies (MAbs). Out of 42 MAbs raised against the homogenate of the olfactory bulb, two types of MAbs that strongly stained the olfactory nerve fibers (axons of olfactory receptor cells) were selected and their staining patterns were analysed in detail. MAbs of one type (represented by MAb R2D5) specifically labeled all olfactory receptor cells in the nasal epithelium and all olfactory nerve fibers and their terminal portions in the bulb. The other typ

Central projections of gustatory nerves in the rat.

The central distributions of gustatory and non-gustatory branches of cranial nerves V, VII, IX, and X were examined after application of horseradish peroxidase to the cut nerve. The nerves conveying gustatory information, chorda tympani (CT), greater superficial petrosal (GSP), lingual-tonsilar branch of IX (LT-IX), superior laryngeal branch of X (SL), distributed primarily to the lateral division of the nucleus of the solitary tract (NST) from its rostral pole to the obex. The CT and GSP distributions were coextensive and terminated most densely in the rostral pole of NST. The LT-IX distribut

Fine afferent fibers from viscera do not terminate in the substantia gelatinosa of the thoracic spinal cord.

Transganglionic transport of horseradish peroxidase (HRP) has been used to study the anatomy of the central projection of somatic and visceral afferent fibers to the thoracic spinal cord of the cat. A dense concentration of somatic afferent fibers and terminals was found in laminae I and II of the dorsal horn and more scattered terminals were present in laminae III, IV and V and in Clarke's column. In contrast, visceral afferent fibers and terminals were found only in lamina I or reaching lamina V via a small bundle of fibers located in the lateral border of the dorsal horn. These results indi

Reflex suppression of renin release by ventricular receptors with vagal afferents.

Intracoronary injection of veratrum alkaloids is known to stimulate ventricular receptors with vagal afferents resulting in bradycardia and hypotension. This study examined the role of veratrum-sensitive ventricular receptors in the control of renin secretion in eight chloralose-anesthetized dogs. Hemorrhage of 10% caused renin release to increase significantly from a control of 778 to 1,971 ng/min. Arterial pressure and renal blood flow did not change significantly. The augmented release of renin during volume depletion was virtually abolished by injection of cryptenamine (2 microgram/kg) int

The hypocretins and their role in narcolepsy.

A series of discoveries spanning the last decade have uncovered a new neurotransmitter - hypocretin - and its role in energy metabolism, arousal, and addiction. Also, notably, a lack of hypocretin function has been unequivocally associated with the sleep disorder narcolepsy. Here we review these findings and discuss how they will influence future treatments of narcolepsy and other arousal and hyperarousal disorders. We introduce the concept of the hypocretin peptides and receptors and discuss the neuroanatomy and neurophysiology of the hypocretin system. A gain of function through pharmacololo

Potential mechanisms in chemical intolerance and related conditions.

The symptom of chemical intolerance may occur in isolation, but often occurs in conjunction with other chronic symptoms such as pain, fatigue, memory disturbances, etc. This frequent clustering of symptoms in individuals has led to the definition of several chronic multisymptom syndromes, such as multiple chemical sensitivity, fibromyalgia, chronic fatigue syndrome, and Gulf War illnesses. The aggregate research into these syndromes has suggested some unifying mechanisms that contribute to symptomatology. Multiple lines of evidence suggest that there is aberrant function of numerous efferent n

Listening in Reverse: A Case for Retrograde Signaling During Auditory Inner Hair Cell Ribbon Synapse Maturation.

Cochlear inner hair cells (IHCs) conduct rapid, sustained, and temporally precise transmission of acoustic information onto spiral ganglion neurons (SGNs). For this purpose, IHCs utilize highly specialized sensory synapses. During development, these so-called 'ribbon' synapses and their postsynaptic counterparts undergo extensive structural and functional refinement-a process driven by pre-sensory spontaneous activity. The underlying molecular mechanisms remain incompletely understood to date. Interestingly, little is known about the involvement of retrograde feedback provided by postsynaptic

Neuroimmune interactions in cardiovascular homeostasis and disease.

Cardiovascular homeostasis is an adaptive process shaped by the interplay between the nervous system and the heart. Emerging evidence demonstrates that immune cells integrate within and across these signaling pathways to modulate inflammatory responses to injury. This neuroimmune interface has a fundamental role in cardiovascular homeostasis and, when dysregulated, may contribute to the pathogenesis of several cardiovascular diseases. Despite its importance, research on the neuroimmune mechanisms in cardiovascular disease is scarce, possibly because it stands upon an intersection of two tradit

Clinical application of respiratory-gated auricular vagal afferent nerve stimulation.

Vagus nerve stimulation (VNS) has garnered significant attention as a promising bioelectronic therapy. In recent years, respiratory-gated auricular vagal afferent nerve stimulation (RAVANS), a novel non-invasive vagus nerve stimulation technique, has emerged. RAVANS integrates respiration with transcutaneous auricular vagus nerve stimulation (taVNS) and shares a similar mechanism of action to traditional VNS. Similar to conventional Vagus Nerve Stimulation (VNS), RAVANS may mitigate brain injury through three primary pathways: reducing neuronal apoptosis, modulating neurotransmitter release, a

Glomerular microcirculation: Implications for diabetes, preeclampsia, and kidney injury.

This review outlines the features of tandem regulation of glomerular microcirculation by autoregulatory mechanisms and intraglomerular redistribution of blood flow. Multiple points of cooperation exist between autoregulatory and distributional mechanisms. Mutual interactions between myogenic and tubuloglomerular feedback (TGF) mechanisms regulating the inflow are briefly discussed. In addition to this, TGF operation involving purinergic, autocoid, and NO signaling affects, however, not only afferent arteriolar tone, but mesangial cell tone as well. The latter reversibly reconfigures the distri

Renal microvascular constriction to membrane depolarization and other stimuli: pivotal role for rho-kinase.

Contraction of vascular smooth muscle is determined not only by levels of intracellular free calcium but also by the sensitivity of its contractile apparatus. A potential modulator of the latter is rho-kinase. We addressed the question of a possible central role for rho-kinase in the regulation of periglomerular microvascular tone. In the rat hydronephrotic kidney model, diameter changes of distal interlobular arteries, afferent and efferent arterioles were measured using three distinctly different stimuli: intravascular pressure changes, angiotensin II (AngII) and membrane depolarization, whi

Developing zebrafish utilize taste-signaling pathways for oxygen chemoreception.

A fundamental requirement for all animals is to sense and respond to changes in environmental O2 availability. Low O2 (hypoxia) typically stimulates breathing, a universal and critical response termed the hypoxic ventilatory response (HVR). In this study, we test the hypothesis that taste-signaling pathways are used for O2 sensing and activation of the HVR. We show that Merkel-like cells (MLCs), which are part of the taste-bud complex, function as O2 chemoreceptor cells in larval zebrafish and that transduction of the O2 signal uses taste-signaling pathways. Specifically, MLCs responded to hyp

Effect of bladder outlet obstruction on micturition reflex pathways in the rat.

The effect of outlet obstruction on neural pathways to the bladder was examined six weeks after urethral ligation in rats. Ligated rats exhibited a seven-fold increase in bladder weight, a six-fold increase in volume, and significant increases (p less than 0.001) in micturition pressure, residual volume and compliance measured under urethane anesthesia. Unanesthetized ligated rats in comparison to control rats voided smaller volumes of urine more frequently (0.7 vs 1.1 voids/hr., p less than 0.001). Inappropriate secretion of antidiuretic hormone in a patient with chronic inflammatory demyelinating polyneuropathy.

A 54-year-old man suffered from a relapse of chronic inflammatory demyelinating polyneuropathy (CIDP), and developed quadriplegia and somnolence requiring mechanical ventilation for respiratory failure. Serum Na concentration remained at low levels during the clinical course, and a diagnosis of inappropriate secretion of antidiuretic hormone (SIADH) was made. The present case had not only acute aggravation of CIDP with autonomic dysfunction but also intracranial hypertension caused by increased CSF protein (maximum level, 1,315 mg/dl). It seemed likely that injury of the afferent fibers of the
