Resveratrol ameliorates hypoxia/ischemia-induced behavioral deficits and brain injury in the neonatal rat brain.

Hypoxia-ischemia (HI) induced injury of the neonatal brain accounts for behavioral deficits concerning mainly neurological reflexes, sensorimotor functions and learning/memory disabilities that may evolve throughout development. The positive biological effects of resveratrol, a natural compound with anti-oxidant/anti-inflammatory properties found mainly in red wine have been indicated recently. Aim of this study was to investigate the delayed outcome of early administration of resveratrol in an experimental model of hypoxic-ischemic encephalopathy, by means of behavioral analysis and late neur…

Altered brain activation in military personnel with one or more traumatic brain injuries following blast.

Explosive blast is a frequent cause of traumatic brain injury (TBI) among personnel deployed to Afghanistan and Iraq. Functional magnetic resonance imaging (fMRI) with an event-related stimulus-response compatibility task was used to compare 15 subjects with mild, chronic blast-related TBI with 15 subjects who had not experienced a TBI or blast exposure during deployment. Six TBI subjects reported multiple injuries. Relative to the control group, TBI subjects had slightly slower responses during fMRI and increased somatic complaints and symptoms of post-traumatic stress disorder (PTSD) and dep…

Posterior fossa brain tissue injury: developmental, neuropsychological, and neurological consequences of brain tumors in children.

The aim of the study was the functional neurodevelopmental assessment of children with posterior fossa tumors, specifically examining whether tumor location in particular cerebellar structures determines particular neuropsychological deficits. The examined group consisted of 34 children treated between 1999 and 2007 at the Division of Pediatric Neurosurgery Silesian University Medical School in Katowice, Poland. Twelve girls and 22 boys between 5 and 21 years of age were examined. The mean age was 12.3 years. There were 21 total and 8 subtotal resections of tumor, and marsupialization was perf

Injury-related production of cysteinyl leukotrienes contributes to brain damage following experimental traumatic brain injury.

The leukotrienes belong to a family of biologically active lipids derived from arachidonate that are often involved in inflammatory responses. In the central nervous system, a group of leukotrienes, known as the cysteinyl leukotrienes, is generated in brain tissue in response to a variety of acute brain injuries. Although the exact clinical significance of this excess production remains unclear, the cysteinyl leukotrienes may contribute to injury-related disruption of the brain-blood barrier and exacerbate secondary injury processes. In the present study, the formation and role of cysteinyl le…

[Deep brain stimulation for essential tremor. Consensus recommendations of the German Deep Brain Stimulation Association].

In Germany, deep brain stimulation (DBS) of the thalamic ventralis intermedius nucleus (VIM) is licensed for treatment of essential tremor in cases unresponsive to pharmacotherapy. Especially a bothersome hand tremor interfering with activities of daily living will improve, whereas head, tongue or vocal tremor shows less response. DBS was proven to be superior to lesional thalamotomy with better functional outcome and less adverse effects. The consensus statement presented here reflects the current recommendations of the German Deep Brain Stimulation Study Group for inclusion and exclusion cri…

Parental exposure to pesticides and childhood brain cancer: U.S. Atlantic coast childhood brain cancer study.

BACKGROUND: The etiology of childhood brain cancer remains largely unknown. However, previous studies have yielded suggestive associations with parental pesticide use. OBJECTIVES: We aimed to evaluate parental exposure to pesticides at home and on the job in relation to the occurrence of brain cancer in children. METHODS: We included 526 one-to-one-matched case-control pairs. Brain cancer cases were diagnosed at < 10 years of age, and were identified from statewide cancer registries of four U.S. Atlantic Coast states. We selected controls by random digit dialing. We conducted computer-assist…

The blood-brain barrier significantly limits eflornithine entry into Trypanosoma brucei brucei infected mouse brain.

Drugs to treat African trypanosomiasis are toxic, expensive and subject to parasite resistance. New drugs are urgently being sought. Although the existing drug, eflornithine, is assumed to reach the brain in high concentrations, little is known about how it crosses the healthy and infected blood-brain barrier. This information is essential for the design of drug combinations and new drugs. This study used novel combinations of animal models to address these omissions. Eflornithine crossed the healthy blood-CNS interfaces poorly, but this could be improved by co-administering suramin, but not n…

Preventing brain injury in newborns with congenital heart disease: brain imaging and innovative trial designs.

BACKGROUND AND PURPOSE: Newborns with congenital heart disease are at high risk for brain injury and adverse neurodevelopmental outcomes. MRI enables the objective determination of the severity of brain injury in critically ill newborns with congenital heart disease. We will rationalize the use of MRI as a surrogate for neurodevelopmental outcome and describe novel randomization techniques that can be used in trials in this population. METHODS: This article describes the evidence for the use of MRI and the link with neurodevelopmental outcome established in newborns. We also discuss the use o…

Antibodies to serotonin attenuate closed head injury induced blood brain barrier disruption and brain pathology.

Closed head injury (CHI) often results in profound brain swelling and instant death of the victims due to compression of the vital centers. However, the neurochemical basis of edema formation in CHI is still obscure. Previous studies from our laboratory show that blockade of serotonin synthesis prior to CHI in a rat model attenuates brain edema, indicating a prominent role for serotonin in head injury. Thus, neutralization of endogenous serotonin activity and/or blocking of its receptors will induce neuroprotection in CHI. Since serotonin has more than 14 receptors and selective serotonin anta…

Cooling the injured brain: how does moderate hypothermia influence the pathophysiology of traumatic brain injury.

Neither any neuroprotective drug has been shown to be beneficial in improving the outcome of severe traumatic brain injury (TBI) nor has any prophylactically-induced moderate hypothermia shown any beneficial effect on outcome in severe TBI, despite the optimism generated by preclinical studies. This contrasts with the paradox that hypothermia still is the most powerful neuroprotective method in experimental models because of its ability to influence the multiple biochemical cascades that are set in motion after TBI. The aim of this short review is to highlight the most recent developments conc…

Diffuse brain injury induces local expression of Na+/myo-inositol cotransporter in the rat brain.

We studied expression of an osmoprotective gene, sodium/myo-inositol cotransporter (SMIT) in Marmarou's animal model for human diffuse brain injury by in situ hybridization and immunohistochemistry. In rats with diffuse brain injury, transient upregulation of SMIT mRNA was exclusively observed in the lateral area of pyramidal tract in lower brainstem. The expression was induced at 1 h after injury, peaked at 24 h, and returned to almost control levels at 48 h. Upregulated expression was found mainly in small glia-like cells. By immunohistochemistry using antibodies to phosphorylated mitogen-ac…

Hepatoblastoma metastatic to brain: prolonged survival after multiple surgical resections of a solitary brain lesion.

PURPOSE: Pulmonary metastases of hepatoblastoma confined to the lung have been cured using therapy that included radical surgical resection. We report the case of a child with a hepatoblastoma metastatic to brain that was successfully treated with multiple surgical resections, irradiation, and chemotherapy. The case demonstrates that such an approach, employing aggressive surgery, can produce durable remission of an extrapulmonary metastasis with hepatoblastoma. PATIENTS AND METHODS: A 17-month-old girl presented with a hepatoblastoma that remained unresectable after chemotherapy and irradiat…

Specific degradation of subendothelial matrix proteoglycans by brain-metastatic melanoma and brain endothelial cell heparanases.

One of the many features of the malignant phenotype, in vitro and in vivo, is elevated heparanase production and activity. Using in vitro model systems, we examined the capacity of murine (B16B15b) and human (70W) brain-metastatic melanoma cells to degrade the subendothelial matrix produced by endothelial cell monolayer cultures. B16B15b and 70W melanoma cells solubilized sulfated matrix proteoglycans at levels significantly higher than their parental lines (B16F1, MeWo). Sulfated matrix proteoglycans were rich in heparan sulfate (HSPGs), with minor amounts of chondroitin and dermatan sulfates…

Brain natriuretic peptide stimulates particulate guanylate cyclase activity in selected areas of the rat brain.

The effect of porcine brain natriuretic peptide (pBNP) on cyclic guanosine monophosphate (cGMP) production was investigated in localized rat brain areas by radioimmunoassay procedure. Porcine BNP activated particulate guanylate cyclase in the median eminence, subfornical organ, choroid plexus, olfactory bulb, paraventricular nucleus and pineal gland in a concentration-dependent fashion and its action was comparable to that of rat atrial natriuretic peptide (alpha-ANP), with ED50 values ranging from 5 to 7 x 10(-7) M for both peptides. Our results suggest that the activation of a specific recep…

Correlation of blood-brain barrier function and HT7 protein distribution in chick brain circumventricular organs.

The HT7 protein defined by a monoclonal antibody is a specific marker for chick brain endothelial cells (EMBO J., 5 (1986) 3179-3183). In this study, we have investigated the expression of this protein in the brain circumventricular organs which lack a blood-brain barrier. Using immunohistochemical techniques we found that the protein was absent from the vascular system of the pituitary, median eminence, subfornical organ, pineal gland, the organum vasculosum lamina terminalis and the layer of sinusoid blood vessels of the area postrema. In some regions of the median eminence and, more strikin…

Right brain, left brain; left face, right face: hemisphericity and the expression of facial emotion.

Research on the asymmetry of facial expressions is reviewed in terms of neuroanatomy, qualitative and quantitative measures of asymmetrical expression, method of expression elicitation (spontaneous, posed, imagery-based), type of expression (happy, sad, etc.), and expression rating procedures. Neuroanatomical evidence indicated that contralateral control of facial musculature exists only for the lower face and that different motor pathways are responsible for spontaneous versus posed expressions. Empirical research on the differential assignment of qualitative trait attributes to facial region

Serial observations of brain stem function by auditory brain stem responses in central transtentorial herniation.

Changes in auditory brain stem responses were serially investigated in a patient with downward transtentorial herniation due to acute obstructive hydrocephalus to correlate the neurological signs with the results of computerized tomography. Neurological deterioration correlated highly with disruption of auditory brain stem responses, especially with that of the components of waves V to VII. A noninvasive technique, the measurement of auditory brain stem responses is thought to be useful in detecting the severity of downward transtentorial herniation and in estimating the recovery of brain stem…

Antibodies against viruses and structural brain components in oligoclonal IgG obtained from multiple sclerosis brain.

Immunoglobulin G (IgG) was isolated from three multiple sclerosis (MS) and two control brains by Protein A Sepharose affinity chromatography and was characterized by thin-layer polyacrylamide gel isoelectric focusing (PAG IEF) and antiserum immunofixation. The three MS brains contained oligoclonal IgG. Immunofixation with measles, herpes simplex, varicella, rubella, mumps and cytomegalovirus as antigens and autoradiography revealed that some of the oligoclonal IgG bands separated by PAG IEF contained antibodies against herpes simplex in one, measles in two and varicella virus in all three MS b…
