Background/Objectives: In situ vaccines that directly release endogenous tumor antigens in situ to elicit anti-tumor immune responses without exogenous antigen preparation have emerged as a promising cancer immunotherapy strategy, due to their enhanced safety by local immunization that minimizes systematically adverse reactions. However, the anti-tumor efficacy of most in situ vaccines is affected by their limited access to tumors in distant sites and the toxicity of the adjuvants contained. Methods: To overcome these shortcomings, the present study explored the feasibility of utilizing extrac

BACKGROUND: In China, lung cancer stands as the leading cause of cancer-related deaths, often resulting in brain metastases (BM) that severely compromise patients' quality of life and reduce survival outcomes. The delivery of drugs to the brain is further complicated by the blood-brain barrier (BBB). To address this, we developed EGFR single-chain fragment variable (scFv)-modified macrophage membrane liposomes (scFv-MML) encapsulating LPCAT1 siRNA (scFv-MML@LPCAT1si) as a targeted therapy for non-small cell lung cancer (NSCLC) BM.
METHODS: EGFR scFv-pcDNA3.1(-) plasmids were transfected into

Oral tumors are not shaped by biochemical signals alone; they also evolve within mechanically active and thermally fluctuating microenvironments that critically regulate malignant progression, immune remodeling, and therapeutic resistance. Here, we develop a dynamic bioorthogonal organoid platform that couples a 3D-printed photothermal elastic scaffold with an interpenetrating hydrogel network derived from decellularized oral mucosa extracellular matrix, enabling the reconstruction of in vivo-like structural support, matrix-derived biochemical cues, spatial cellular compartmentalization, and d

Glycyrrhiza uralensis polysaccharides (GUPS) are considered safe and effective natural medicine due to their extensive pharmacological activity and low toxicity. However, their large molecular weight (approximately 29 kDa) and high polarity restrict intestinal absorption, resulting in poor oral bioavailability. Here, GUPS-PLGA nanoparticles (GUPS-PLGA NPs, GPN) were prepared and their intestinal absorption characteristics and immunostimulatory activity were evaluated both in vitro and in vivo. Notably, GPN with a high encapsulation rate (63.18%) was prepared by response surface methodology. Fu

Hydrogels containing stem cells are a promising strategy to improve spinal cord injury (SCI) repair. Recent advances in tissue engineering have made cell-containing hydrogels more effective for achieving better regeneration outcomes. One such advancement is the incorporation of decellularized extracellular matrix (ECM) into hydrogels to develop a cell-compatible environment that mimics the mechanical and biological properties of native tissue ECM. Herein, a unique bioactive injectable hydrogel based on decellularized nerve ECM, carboxymethyl chitosan (CMC) and oxidized alginate (OA) was fabric

Immunotherapy has revolutionized the landscape of cancer treatment; however, most current strategies target only isolated steps of the cancer-immunity cycle and therefore struggle to achieve durable clinical success. Central to immune surveillance, the cGAS-STING pathway orchestrates innate and adaptive responses by promoting dendritic cell maturation, cytotoxic T lymphocyte infiltration, and durable immunological memory. However, chemotherapeutic activation of this axis remains elusive due to a paucity of safe, efficacious, and translatable inducers of tumor-derived double-stranded DNA (dsDNA

Myocardial infarction (MI) is a major ischemic heart condition that leads to the loss of billions of cardiomyocytes, triggering adverse ventricular remodeling and fibrotic tissue formation, which can ultimately result in heart failure. Current treatments fail to address cardiomyocyte loss, prompting interest in cardiac regeneration strategies. Although cell-based therapies have gained attention, they face limitations such as poor cell survival and engraftment. Acellular nanoformulations represent a promising alternative. In this study, protein-free bioinspired lipid nanoparticles mimicking low

Stem cell-derived β-cells (SC-BCs) represent a potential source for curing diabetes. To date, in vitro generated SC-BCs display an immature phenotype and lack important features in comparison to their bona-fide counterparts. Transplantation into a living animal promotes SC-BCs maturation, indicating that components of the in vivo microenvironment trigger final SC-BCs development. Here, we investigated whether cues of the pancreas specific extracellular matrix (ECM) can improve the differentiation of human induced pluripotent stem cells (hiPSCs) towards β-cells in vitro. To this aim, a pancreas

Using plant extracts as photosensitizers in photodynamic therapy (PDT) represents a significant green approach toward sustainability. This study investigates beetroot juice (BRJ), betanin, and their liposomal formulations (Lip-BRJ, Lip-Bet) as photosensitizers in cancer PDT. BRJ was prepared, and its betanin content was quantified via HPLC. The p-nitrosodimethylaniline (RNO)/imidazole technique monitored the singlet oxygen formation. BRJ and betanin decreased the RNO absorbance at 440 nm by 12% and 9% after 45 min of irradiation, respectively. Furthermore, betanin interaction with Bcl-2 protei

The safe administration of thrombolytic agents is a challenge for the treatment of acute thrombosis. Lipid-based nanoparticle drug delivery technologies present opportunities to overcome the existing clinical limitations and deliver thrombolytic therapy with enhanced therapeutic outcomes and safety. Herein, lipid cubosomes are examined as nanocarriers for the encapsulation of thrombolytic drugs. The lipid cubosomes are loaded with the thrombolytic drug urokinase-type plasminogen activator (uPA) and coated with a low-fouling peptide that is incorporated within a metal-phenolic network (MPN). A tumor in the anterior mediastinum was found in an adult male German Shorthaired Pointer. The dog presented with hypercalcemia that varied with the removal and recurrence of the tumor. The neoplasm was characterized by an encapsulated multilobed grey-white soft mass with stroma, and the recurrent tumor infiltrated veins and the heart. The tumor consisted of groups of clear cells with well outlined cytoplasmic boundaries separated by thin vascular stroma. These cells had few organelles, scattered clumps of glycogen, clear cytosol and prominent infolding and interdigitating plasma membranes and

Glaucoma, a major cause of irreversible blindness worldwide, is associated with elevated intraocular pressure (IOP) and progressive loss of retinal ganglion cells (RGCs) that undergo apoptosis. A mechanism for RGCs injury involves impairment of neurotrophic support and exogenous supply of neurotrophic factors has been shown to be beneficial. However, neurotrophic factors can have widespread effects on neuronal tissues, thus targeting neurotrophic support to injured neurons may be a better neuroprotective strategy. In this study, we have encapsulated LM22A-4, a small neurotrophic factor mimetic

Cultured epidermal autografts (CEA) reportedly have better cosmesis than traditional autografts and are especially useful in the absence of healthy skin for grafting. To support cell growth and prevent desiccation during cultivation, CEAs require platelet-rich plasma (PRP), which is typically applied daily for 2 weeks. However, repeated withdrawal of blood may be unfeasible in certain patients, and importantly, the optimal PRP concentration for cell growth remains undefined. Identifying lower, yet effective, concentrations may reduce the need for extensive blood collection. In this in vitro ob

The infiltration and cytotoxicity of chimeric antigen receptor (CAR)-T cells are crucial for effective elimination of solid tumors. While metallo-immunotherapy is a promising strategy that can activate the antitumor immunity, its role in promoting CAR-T cell therapy remains elusive. The first single-element nanomaterial based on chromium nanoparticles (Cr NPs) for cancer photo-metallo-immunotherapy has been reported previously. Herein, an extended study using biodegradable polydopamine as a versatile carrier for these nanoparticles, enabling synergistic CAR-T cell therapy, is reported. The res

Replacement of β cells is only a curative approach for type 1 diabetes (T1D) patients to avoid the threat of iatrogenic hypoglycemia. In this pursuit, islet allotransplantation under Edmonton's protocol emerged as a medical miracle to attain hypoglycemia-free insulin independence in T1D. Shortage of allo-islet donors and post-transplantation (post-tx) islet loss are still unmet hurdles for the widespread application of this therapeutic regimen. The long-term survival and effective insulin independence in preclinical studies have strongly suggested pig islets to cure overt hyperglycemia. Import
