Peptidergic mechanisms in feeding behaviour.

Hyperphagia was induced in mice by p.o. administration of different types of CNS depressant drugs, like chlordiazepoxide 25 mg/kg diazepam 2.5 mg/kg, cyproheptadine 2 mg/kg and phenobarbitone 25 mg/kg. Such hyperphagia was abolished by pretreatment with naloxone 0.1 mg/kg sc. Naloxone per se at this dose produced no significant effect on the food intake. This is suggestive of the role of peptidergic mechanisms in the feeding behaviour in mice.

No evidence for presynaptic opioid receptors on cholinergic, but presence of kappa-receptors on dopaminergic neurons in the rabbit caudate nucleus: involvement of endogenous opioids.

The effects of various opioid receptor agonists and antagonists were studied in rabbit caudate nucleus slices preincubated with either [3H]dopamine or [3H]choline, superfused with medium (containing in most experiments the D2 receptor antagonist domperidone) and subjected to electrical field stimulation. The stimulation-evoked [3H]overflow from slices prelabeled with [3H]dopamine (evoked [3H]dopamine release) was significantly reduced by preferential kappa-opioid receptor agonists, like U-50,488 H, but not by mu- or delta-opioid receptor selective drugs. Opioid receptor antagonists shifted the

Opioids depress in vitro human monocyte chemotaxis.

Intravenous drug abusers (IDA) normally show functional defects in monocyte activity, in particular their chemotactic response. The aim of the present work was to study the action of several opioids on monocyte chemotaxis. To do so, monocytes from healthy individuals were incubated with heroin and morphine at three different concentrations (10(-5) M, 10(-6) M and 10(-7) M), with the finding of a significant depression in monocyte chemotaxis in all cases. This alteration could be due to a receptor effect or, conversely, to a non-specific effect. Accordingly, in the second phase of the study, mo

Effects of opioid agonists on sympathetic and parasympathetic transmission to the dog heart.

Effects of leu- and met-enkephalin, pentazocine and morphine on negative or positive chronotropic response to vagal nerve stimulation or cardiac sympathetic nerve stimulation were examined in anesthetized dogs in order to determine whether opioid receptors modulate vagal and sympathetic transmission. Leu- and met-enkephalin (10-100 micrograms/kg i.v.) and pentazocine (100-1000 micrograms/kg i.v.) inhibited bradycardic response to vagal nerve stimulation (1-4 Hz) in a dose-dependent manner. Morphine (300 and 1000 micrograms/kg i.v.) did not affect vagal bradycardia. The inhibitory effect of leu

Inescapable shock, neurotransmitters, and addiction to trauma: toward a psychobiology of post traumatic stress.

Chronic post traumatic stress has been described as a "physioneurosis" (Kardiner 1941), that is, a mental disorder with both psychological and physiological components. The behavioral sequelae of inescapable shock in animals and of massive psychic trauma in people show a striking parallel. Inescapable shock in animals leads to both transient catecholamine depletion and subsequent stress-induced analgesia. We postulate that the numbing and catatenoid reactions following trauma in humans correspond to the central nervous system (CNS) catecholamine depletion that follows inescapable shock in anim

Comparison of the ability of opioid analgesics to increase endogenous opioid-like activity in the cerebrospinal fluid of rabbits.

We have previously demonstrated that the acute administration of morphine increases the level of endogenous substances, which have antinociceptive activity, in cerebrospinal fluid (CSF). The present study was conducted to determine whether other opioid analgesics exert a similar effect. CSF was withdrawn from the cisterna magna of anesthetized rabbits before and after s.c. injections of meperidine, pentazocine, levorphanol and methadone, and was bioassayed for opioid-like activity in the mouse tail-flick and phenylquinone writhing tests. The opioid-like activity of CSF taken 60 min after meper

Delta versus mu receptors: cardiovascular and respiratory effects of opiate agonists microinjected into nucleus tractus solitarius of cats.

The cardiovascular and respiratory responses to relatively specific mu or delta agonists microinjected (0.5 microliter/kg) into the region of the nucleus of tractus solitarius (NTS) were examined in anesthetized cats. Blood pressure, heart rate, and respiratory rate were monitored for 30 min after the microinjection of opioid compounds or saline vehicle. The delta agonist, (D-Ala2,D-Leu5)-enkephalin (10-100 nmol/kg) elicited dose-dependent decreases in blood pressure, heart rate, and respiratory rate which were naloxone reversible. In contrast the mu agonists, morphine (10-54 nmol/kg) and morp

A met-enkephalin analog inhibits adrenocorticotropin secretion by cultured pituitary cells from a patient with Nelson's syndrome.

ACTH excretion by cultured nonenzymatically dispersed pituitary tumor cells from a patient with Nelson's syndrome was studied. Hormone release was suppressed by 74 +/- 6% by the addition of 1 microM of the met-enkephalin analog FK 33824, while naloxone (1 microM) stimulated ACTH release by 70 +/- 5%. Somatostatin, dexamethasone, bromocriptine, and cyproheptadine in a concentration of 1 microM each inhibited ACTH release by 25 +/- 2%, 35 +/- 2%, 52 +/- 2%, and 61 +/- 4%, respectively, while lysine vasopressin (0.1 microM) and dibutyryl cAMP (5 mM) stimulated ACTH release by 112 +/- 8% and 220 +

The orexigenic effect of kyotorphin in chicks involves hypothalamus and brainstem activity and opioid receptors.

Kyotorphin (KTP), first isolated in the bovine brain and now having been identified in a variety of species, is known most extensively for its analgesic-like properties. KTP indirectly stimulates opioid receptors by releasing methionine enkephalin (met-enkephalin). Stimulation of opioid receptors is linked to hunger perception. In the present study, we sought to elucidate the effect of KTP on food intake in the neonatal chick. Intracerebroventricular injection of 0.6, 3.0 and 12 nmol KTP increased feeding up to 60 min post-injection. KTP treated chicks increased pecking efficiency and decrease

Effects of naltrexone on electrocutaneous pain in patients with hypertension compared to normotensive individuals.

An opioid mechanism may help explain hypertensive hypoalgesia. A double-blind placebo-controlled design compared the effects of opioid blockade (naltrexone) and placebo on electrocutaneous pain threshold, pain tolerance, and retrospective McGill Pain Questionnaire ratings in 35 unmedicated patients with essential hypertension and 28 normotensive individuals. The hypertensives experienced less pain than normotensives during the assessment of their pain tolerance; however, this manifestation of hypertensive hypoalgesia was not moderated by naltrexone. These findings fail to support the hypothesi

Habituation to test procedure modulates the involvement of dopamine D2- but not D1-receptors in ethanol-induced locomotor stimulation in mice.

RATIONALE: Novelty associated with behavioral testing has been shown to enhance psychostimulant- and morphine-induced locomotor stimulation. Evidence has demonstrated that novelty increases dopamine (DA) activity, and habituation to a novel environment reduces such activation. However, it is not clear whether novelty modulates ethanol-induced behavioral stimulation and whether DA plays a role in this effect. OBJECTIVES: The present work sought to demonstrate a role of habituation to test procedure as a factor that could modulate the involvement of DA in ethanol-induced locomotor stimulation.

Structure-activity relationship, conformation and pharmacology studies of morphiceptin analogues--selective mu-opioid receptor ligands.

Morphiceptin (Tyr-Pro-Phe-Pro-NH(2)) is one of the most selective agonists for the mu-opioid receptor. In this review structure-activity relationships of morphiceptin analogues and studies resulting in defining low energy conformations are discussed. Finally, new developments in the control of tumour growth and cell proliferation by morphiceptin analogues are surveyed, which open future perspectives in the diagnosis and treatment of various cancers.

Behavior of rats during chronic activation and blockade of the neostriatal opiate system.

The effects of daily microinjections (MI), over of three weeks, bilaterally into the rostral striatum, of morphine, promedol, native leu-enkephalin and its synthetic tetrapeptide analogs were studied in experiments on rats. Naloxone was used as an antagonist. An active avoidance conditioned reflex was developed preliminary in a shuttle box. A decrease in the accuracy of the realization and an increase in the latent period of the reflex were observed after the first MI of morphine and enkephalins. The effect of the most stable aminated ornithine-containing tetrapeptides proved to be the stronge

Morphine cross-reacts with somatostatin receptor SSTR2 in the T47D human breast cancer cell line and decreases cell growth.

In a previous study, we found that morphine decreases, in a dose-dependent manner, the cell growth of T47D human breast cancer cells, despite the lack of mu opioid receptors and an interaction of morphine with other opioid sites. We have therefore examined a possible interaction of morphine with other membrane receptor systems of the cell. The present study describes for the first time an interaction between mu-acting opioid drugs and the somatostatinergic system. We have found that [125I]Tyr11-somatostatin binds with high affinity to T47D cells. Analysis of the binding data showed the presenc

Involvement of amino acids, opioids, nitric oxide, and NMDA receptors in learning and memory consolidation in crickets.

The effect of injections of selected amino acids and of N-methyl-D-aspartate (NMDA); morphine; and NMDA, nitric oxide (NO), and opioid inhibitors given before a maze-learning was investigated. Thirsty crickets (Pteronemobius sp) were trained to turn only to one side of a symmetrical Y-shaped maze using reinforcements with water. The insects retained the learned task 24 h later. N2 anoxia applied immediately after training produced retrograde amnesia. Injections of alanine (Ala), arginine (Arg), glutamine (Gln), morphine, or NMDA prior to training blocked the amnesic action of anoxia. Naloxone,

Cholinergic and opioid involvement in release of growth hormone during exercise and recovery.

Cholinergic and opioid pathways have been implicated as mediators of the increased growth hormone (GH) release observed during exercise. This study compared the GH responses induced by a moderate-intensity exercise bout during treatment with placebo (Plac), the opioid receptor antagonist naltrexone (Nalt), the indirect cholinergic agonist pyridostigmine (PD), or a combination of the two drugs (P + N). Ten active males served as subjects (age, 25.1 +/- 0.6 yr; wt, 79.7 +/- 2.5 kg; % body fat, 14.9 +/- 1.4; peak oxygen consumption, 46.2 +/- 2.7 ml.kg-1 x min-1). Blood samples were drawn at 5-min

Opioid and prostaglandin mechanisms involved in the effects of GABAergic drugs on body temperature.

1. Intraperitoneal (i.p.) injection to restrained rats of GABA (250-1000 mg/kg) or the GABAA-receptor agonist muscimol (0.05-1 mg/kg) induced a dose-dependent decrease in body temperature (BT). 2. Intraperitoneal injection of low doses of the GABAB-receptor agonist (+/-)-baclofen (1-10 mg/kg) did not significantly affect BT. However, baclofen, at high doses (30 mg/kg), produced an increase in BT. 3. Pretreatment with either bicuculline (3 mg/kg) or naloxone (1 mg/kg) did not significantly modify the hypothermic response observed with GABA or muscimol, except for the high dose of GABA (1000 mg/

Cardiovascular effects of opioid antagonist naloxone in rostral ventrolateral medulla of rabbits.

We have examined the influence of endogenous opioids on the basal and reflex control of arterial blood pressure in the pressor region of the rostral ventrolateral medulla (RVLM) of chloralose-anesthetized rabbits. We tested basal effects both in intact animals and after hypotensive hemorrhage. Bilateral administration of the opiod antagonist naloxone (20 nmol, 100 nl) directly into the RVLM induced a gradual and prolonged increase in mean arterial pressure (MAP) (+17 +/- 2 mmHg). This was preceded by a brief and mild depressor effect (-9 +/- 3 mmHg), which was attributable to a transient reduc

Prodynorphin peptide distribution in the forebrain of the Syrian hamster and rat: a comparative study with antisera against dynorphin A, dynorphin B, and the C-terminus of the prodynorphin precursor molecule.

The neuroanatomical distribution of the prodynorphin precursor molecule in the forebrain of the male Syrian hamster (Mesocricetus auratus) has been studied with a novel antiserum directed against the C-terminus of the leumorphin [dynorphin B (1-29)] peptide product. C-peptide staining in sections from colchicine-treated hamsters is compared to staining in sections from untreated animals. In addition, the pattern of C-peptide immunostaining in hamster brain is compared to that in the rat brain. Finally, the C-peptide immunolabeling patterns in hamsters and rats are compared to those obtained wi
