Shared genetic etiology underlying Alzheimer's disease and major depressive disorder.

Patients with late-onset Alzheimer's disease (LOAD) frequently manifest comorbid neuropsychiatric symptoms with depression and anxiety being most frequent, and individuals with major depressive disorder (MDD) have an increased prevalence of LOAD. This suggests shared etiologies and intersecting pathways between LOAD and MDD. We performed pleiotropy analyses using LOAD and MDD GWAS data sets from the International Genomics of Alzheimer's Project (IGAP) and the Psychiatric Genomics Consortium (PGC), respectively. We found a moderate enrichment for SNPs associated with LOAD across increasingly st

C57BL/6 substrain differences in inflammatory and neuropathic nociception and genetic mapping of a major quantitative trait locus underlying acute thermal nociception.

Sensitivity to different pain modalities has a genetic basis that remains largely unknown. Employing closely related inbred mouse substrains can facilitate gene mapping of nociceptive behaviors in preclinical pain models. We previously reported enhanced sensitivity to acute thermal nociception in C57BL/6J (B6J) versus C57BL/6N (B6N) substrains. Here, we expanded on nociceptive phenotypes and observed an increase in formalin-induced inflammatory nociceptive behaviors and paw diameter in B6J versus B6N mice (Charles River Laboratories). No strain differences were observed in mechanical or therma

The interplay of canonical and noncanonical Wnt signaling in metabolic syndrome.

Metabolic syndrome is a cluster of inherited metabolic traits, which centers around obesity and insulin resistance and is a major contributor to the growing prevalence of cardiovascular disease. The factors that underlie the association of metabolic traits in this syndrome are poorly understood due to disease heterogeneity and complexity. Genetic studies of kindreds with severe manifestation of metabolic syndrome have led to the identification of casual rare mutations in the LDL receptor-related protein 6, which serves as a co-receptor with frizzled protein receptors for Wnt signaling ligands.

Association of GTF2I and GTF2IRD1 polymorphisms with systemic lupus erythematosus in a Chinese Han population.

PURPOSE: Increasing evidence suggests that gut microbiota are associated with the risk of kidney stone disease; however, whether a causal relationship exists remains unclear. We used Mendelian randomization to assess the potential causal relationship between gut microbiota and kidney stone disease risk. MATERIALS AND METHODS: Genetic instrumental variables for gut microbiota were identified from a genome-wide association study of 18,340 participants. Summary statistics for kidney stone disease were derived from genome-wide association studies, including 284,598 cases and 480,873 controls. The

Perinatal depression and omega-3 fatty acids: a Mendelian randomisation study.

BACKGROUND: There have been numerous studies investigating the association between omega-3 fatty acids (FAs) and depression, with mixed findings. We propose an approach which is largely free from issues such as confounding or reverse causality, to investigate this relationship using observational data from a pregnancy cohort. METHODS: The Avon Longitudinal Study of Parents and Children (ALSPAC) cohort collected information on FA levels from antenatal blood samples and depressive symptoms at several time points during pregnancy and the postnatal period. Conventional epidemiological analyses we

Genetically Predicted Gut Microbiota and Lymphoma Risk: A Mendelian Randomization Study.

INTRODUCTION: Growing evidence links gut microbiota (GM) to hematological malignancies; however, its role in lymphoma remains unclear. This study aimed to investigate the potential causal relationships between genetically predicted gut microbial taxa and lymphoma subtypes using a Mendelian randomization (MR) framework. METHODS: Using genome-wide association study (GWAS) summary data for 211 gut microbial taxa and 10 lymphoma subtypes, we performed bidirectional Mendelian randomization (MR) and sensitivity analyses to assess causality. Reverse MR was also used to evaluate reverse causation. St

Mendelian randomization identifies circulating miRNAs as causal mediators of gastric cancer susceptibility and survival outcomes.

Gastric cancer (GC) remains a leading cause of cancer mortality, yet the causal roles of microRNAs (miRNAs) in its pathogenesis are poorly characterized. While observational studies implicate miRNAs in GC progression, confounding biases and tissue-specific limitations hinder causal inference and clinical translation. We conducted a 2-sample Mendelian randomization (MR) analysis using genetic instruments derived from plasma miRNA expression quantitative trait loci (eQTLs). Summary-level data for miRNA-eQTLs were obtained from a study by Huan et al. (involving 5239 individuals and 280 miRNAs), w

Constraints on the evolution of a doublesex target gene arising from doublesex's pleiotropic deployment.

"Regulatory evolution," that is, changes in a gene's expression pattern through changes at its regulatory sequence, rather than changes at the coding sequence of the gene or changes of the upstream transcription factors, has been increasingly recognized as a pervasive evolution mechanism. Many somatic sexually dimorphic features of Drosophila melanogaster are the results of gene expression regulated by the doublesex (dsx) gene, which encodes sex-specific transcription factors (DSX(F) in females and DSX(M) in males). Rapid changes in such sexually dimorphic features are likely a result of chang

Type 2 Diabetes Reduces IFN-α2 and Compromises Antiviral Defense: Evidence From Mendelian Randomization and H1N1-Infected Diabetic Mice.

UNLABELLED: Type 2 diabetes (T2D) impairs antiviral immunity; however, the causal link between T2D and interferon-α2 (IFN-α2) deficiency remains unclear. This study used genome-wide association study-based Mendelian randomization (MR) to investigate this relationship and validated the findings in an H1N1-infected diabetic mouse model. MR analysis of 26 single nucleotide polymorphisms showed a significant negative association between T2D and IFN-α2 levels (inverse variance weighted odds ratio 0.667; P = 0.000116) without heterogeneity or pleiotropy. In vivo experiments confirmed that db/db mice

Identifying the genetic link between type 1 diabetes and autoimmune liver diseases.

INTRODUCTION: It appears that type 1 diabetes mellitus (T1DM) and autoimmune liver diseases (AILDs) are associated, but there is no evidence linking them causally or in a specific direction. This study aims to evaluate the causal relationship between T1DM and AILDs. MATERIAL AND METHODS: We performed a two-sample Mendelian randomization (MR) analysis. Following thorough evaluation, the dataset from the genome-wide association study was utilized to identify potential candidate single nucleotide polymorphisms. Inverse variance weighting (IVW) served as the primary analytical method, complemente

Causal Associations Between Pre-Pregnancy Diabetes Mellitus and Pre-Eclampsia Risk: Insights from a Mendelian Randomization Study.

BACKGROUND AND OBJECTIVES: Pre-eclampsia (PE) is a serious pregnancy complication defined by the onset of hypertension and multi-organ dysfunction occurring after 20 weeks of gestation. Studies have indicated the correlation between diabetes mellitus (DM) and PE, but the causal relationship remains unclear. MATERIALS AND METHODS: The two-sample Mendelian randomization (MR) approach, including the inverse variance weighted random effects (IVW-RE) model and the traditional sensitivity model, was employed to assess the causal effects of pre-pregnancy type 1 diabetes (T1D) and type 2 diabetes (T2

Genetically predicted CD28 on CD4+ CD45RA+ T cells mediate the association between rheumatoid arthritis and blood leukocytes mitochondrial DNA copy number: A mediation Mendelian randomization study.

Rheumatoid arthritis (RA) is a chronic autoimmune disorder affecting approximately 1% of adults globally, characterized by synovial inflammation and cartilage destruction. A connection between RA and mitochondrial DNA copy number (mtDNA-CN) has been found in observational studies, but the causal relationship remains unresolved. This study is the first to integrate bidirectional Mendelian randomization (MR) and mediation analysis to explore whether RA causally affects mtDNA-CN and to identify immune cell phenotypes, particularly CD28 on CD4 + CD45RA + T cells, that mediate this relationship. Rheumatoid arthritis and the risk of fracture: A Mendelian randomization study.

Observational epidemiological studies indicate a higher fracture incidence in rheumatoid arthritis (RA) patients compared to the general population. However, the causal relationship between RA and fracture risk, particularly traumatic and osteoporotic fractures, is not well established. We performed Mendelian randomization (MR) analysis to evaluate the causal relationship between RA and fracture risk. We performed a MR analysis using summary statistics from genome-wide association studies to investigate the causal association between RA and the risk of traumatic fractures at 9 sites and 3 type

Effects of circulating inflammatory proteins on osteoporosis and fractures: evidence from genetic correlation and Mendelian randomization study.

OBJECTIVE: There is a controversy in studies of circulating inflammatory proteins (CIPs) in association with osteoporosis (OP) and fractures, and it is unclear if these two conditions are causally related. This study used MR analyses to investigate the causal associations between 91 CIPs and OP and 9 types of fractures. METHODS: Genetic variants data for CIPs, OP, and fractures were obtained from the publicly available genome-wide association studies (GWAS) database. We used inverse variance weighted (IVW) as the primary analysis, pleiotropy, and heterogeneity tests to analyze the validity an

Association between habitual coffee consumption and multiple disease outcomes: A Mendelian randomisation phenome-wide association study in the UK Biobank.

BACKGROUND: Coffee is the most commonly consumed beverage in the world after water, however the debate as to whether coffee consumption is beneficial or detrimental to health continues. Current evidence of the link between coffee and health outcomes is predominately observational, thus subject to methodological issues such a confounding and reverse causation. METHODS: This Mendelian randomisation phenome-wide association study (MR-PheWAS) used information from up to 333,214 participants of White-British ancestry in the UK Biobank to examine the causal association between genetically instrumen

Site-Resolved Plasma-Protein Architecture of Infection Susceptibility: A Cis-pQTL Mendelian Randomization and Colocalization Study.

Background/Objectives: Infection susceptibility and systemic infection severity arise from distinct tissue barriers and host-response compartments, but plasma-protein genetic studies often treat infection as a broad phenotype. We aimed to define a site-resolved host plasma-protein architecture of infection susceptibility across urinary, lower-respiratory, systemic, and skin/soft-tissue infection outcomes. Methods: We performed proteome-wide two-sample cis-protein quantitative trait locus (cis-pQTL) Mendelian randomization (MR) using UK Biobank Pharma Proteomics Project (UKB-PPP) instruments an

Causal links between personality disorders and schizophrenia: A Mendelian randomization study.

Although observational studies have suggested associations between personality disorders and schizophrenia, the causality of these relationships remains unclear. Determining whether personality disorders causally contribute to schizophrenia could inform early identification and preventive efforts. We performed two-sample Mendelian randomization (MR) analysis using large-scale Genome-wide Association Study data from populations of European ancestry. Because no single nucleotide polymorphism for personality disorders reached the conventional genome-wide significance threshold (P < 5 × 10-8), we
