Determination of free cyano-cobinamide in swine and rabbit plasma by liquid chromatography tandem mass spectrometry.

In recent years, Cobinamide (Cbi) has shown promise as a therapeutic for cyanide poisoning. There are several forms of Cbi based on the identity of the ligands bound to the cobalt in Cbi and these different forms of Cbi have divergent behavior (e.g., the aquo and hydroxo forms of Cbi readily bind to proteins, limiting their distribution significantly, whereas [Cbi(CN)2] does not). While current analysis techniques only measure total Cbi, methods to elucidate the behavior of 'available' Cbi versus cyanide-complexed Cbi would be valuable for biomedical and pharmacokinetic studies. Therefore, a m

Structural changes of fibrinogen molecule mediated by the N-homocysteinylation reaction.

Homocysteine and its cyclic ester homocysteine thiolactone (HTL) have been involved in the detrimental consequences associated to hyperhomocysteinemia, an independent risk factor for vascular diseases. HTL reacts with protein lysine residues in a process named N-homocysteinylation. The aim of our study was to evaluate the in vitro effects of HTL on the fibrinogen through electrophoretic methods. Fibrinogen was incubated with HTL at different molar ratios and structural changes of the protein were assessed by polyacrylamide gel electrophoresis (PAGE), capillary zone electrophoresis (CZE) and ca

Pharmacological profiling of sigma 1 receptor ligands by novel receptor homomer assays.

The sigma 1 receptor (σ1R) is a structurally unique transmembrane protein that functions as a molecular chaperone in the endoplasmic reticulum (ER), and has been implicated in cancer, neuropathic pain, and psychostimulant abuse. Despite physiological and pharmacological significance, mechanistic underpinnings of structure-function relationships of σ1R are poorly understood, and molecular interactions of selective ligands with σ1R have not been elucidated. The recent crystallographic determination of σ1R as a homo-trimer provides the foundation for mechanistic elucidation at the molecular level

An Uncharacterized Member of the Gls24 Protein Superfamily Is a Putative Sensor of Essential Amino Acid Availability in Streptococcus pneumoniae.

Proteins belonging to the Gls24 superfamily are involved in survival of pathogenic Gram-positive cocci under oligotrophic conditions and other types of stress, by a still unknown molecular mechanism. In Firmicutes, this superfamily includes three different valine-rich orthologal families (Gls24A, B, C) with different potential interactive partners. Whereas the Streptococcus pneumoniae Δgls24A deletion mutant experienced a general long growth delay, the Δgls24B mutant grew as the parental strain in the semisynthetic AGCH medium but failed to grow in the complex Todd-Hewitt medium. Mode of action of nanoparticles against insects.

The employment of nanoparticles obtained through various synthesis routes as novel pesticides recently attracted high research attention. An impressive number of studies have been conducted to test their toxic potential against a wide number of arthropod pests and vectors, with major emphasis on mosquitoes and ticks. However, precise information on the mechanisms of action of nanoparticles against insects and mites are limited, with the noteworthy exception of silica, alumina, silver, and graphene oxide nanoparticles on insects, while no information is available for mites. Here, I summarize cu

Increased Antimicrobial Resistance in a Novel CMY-54 AmpC-Type Enzyme with a GluLeu217-218 Insertion in the Ω-Loop.

During a Spanish surveillance study, a natural variant of a CMY-type β-lactamase related to CMY-2 with a GluLeu217-218 insertion in the Ω-loop (designated CMY-54) was found to increase the minimum inhibitory concentractions to β-lactams in a clinical strain of Escherichia coli. The aim of this study was to characterize CMY-54 by genetic, microbiological, and biochemical analysis. The blaCMY-54 gene is encoded by a plasmid of around 100 kb that hybridizes with K and FIB probes. The genetic context of blaCMY-54 and blaCMY-2 genes was found to be very similar. E. coli expressing CMY-54 under isog

Rapid analysis of aflatoxin M1 in milk using dispersive liquid-liquid microextraction coupled with ultrahigh pressure liquid chromatography tandem mass spectrometry.

A simple, rapid, and sensitive method based on simultaneous protein precipitation and extraction of aflatoxin M1 (AFM1) followed by dispersive liquid-liquid microextraction (DLLME) and ultrahigh pressure liquid chromatography tandem mass spectrometry (UHPLC-MS/MS) analysis was developed for the determination of AFM1 in milk samples. In order to precipitate the proteins and extract AFM1 from milk, a sample pretreatment using acetonitrile and NaCl as the extraction/denaturant solvent and salting-out agent, respectively, was optimised. Subsequently, the acetonitrile (upper) phase, containing AFM1

Lysyl hydroxylase 3 glucosylates galactosylhydroxylysine residues in type I collagen in osteoblast culture.

Lysyl hydroxylase 3 (LH3), encoded by Plod3, is the multifunctional collagen-modifying enzyme possessing LH, hydroxylysine galactosyltransferase (GT), and galactosylhydroxylysine-glucosyltransferase (GGT) activities. Although an alteration in type I collagen glycosylation has been implicated in several osteogenic disorders, the role of LH3 in bone physiology has never been investigated. To elucidate the function of LH3 in bone type I collagen modifications, we used a short hairpin RNA technology in a mouse osteoblastic cell line, MC3T3-E1; generated single cell-derived clones stably suppressin

The Hsp60-(p.V98I) mutation associated with hereditary spastic paraplegia SPG13 compromises chaperonin function both in vitro and in vivo.

We have previously reported the association of a mutation (c.292G > A/p.V98I) in the human HSPD1 gene that encodes the mitochondrial Hsp60 chaperonin with a dominantly inherited form of hereditary spastic paraplegia. Here, we show that the purified Hsp60-(p.V98I) chaperonin displays decreased ATPase activity and exhibits a strongly reduced capacity to promote folding of denatured malate dehydrogenase in vitro. To test its in vivo functions, we engineered a bacterial model system that lacks the endogenous chaperonin genes and harbors two plasmids carrying differentially inducible operons with h

Human placental lipid induces melanogenesis by increasing the expression of tyrosinase and its related proteins in vitro.

Lipids, particularly sphingolipids, are emerging as novel regulators of cellular activity. A placental total lipid fraction (PTLF), the total lipid prepared from an hydroalcoholic extract of fresh term human placenta, was previously shown to have a pigment-inducing activity in an animal model. The PTLF contains sphingolipids which stimulate DNA synthesis and melanin formation with marked morphological changes in B16F10 melanoma cells. In order to identify the mechanism underlying the increased melanin synthesis, B16F10 cells were treated with PTLF to assess the catalytic activities of tyrosina

V antigen-polyhistidine fusion peptide: binding to LcrH and active immunity against plague.

The structural gene for V antigen (lcrV) is known to be encoded within the lcrGVH-yopBD operon of the approximately 70-kb low-calcium-response or Lcr plasmid of Yersinia pestis. This 37-kDa monomeric peptide was reported to provide active immunity in mice, suppress inflammatory cytokines, and regulate expression of the low calcium response (Lcr+). Here we describe pVHB62, encoding a polyhistidine-V antigen fusion peptide (Vh) and linked LcrH. Vh underwent degradation from both the C terminus and N terminus during classical chromatographic fractionation but remained intact within two compartmen

Identification of a mutation in the gene encoding the alpha subunit of the stimulatory G protein of adenylyl cyclase in McCune-Albright syndrome.

McCune-Albright syndrome (MAS) is characterized by polyostotic fibrous dysplasia, café-au-lait lesions, and a variety of endocrine disorders, including precocious puberty, hyperthyroidism, hypercortisolism, growth hormone excess, and hyperprolactinemia. The diverse metabolic abnormalities seen in MAS share the involvement of cells that respond to extracellular signals through activation of the hormone-sensitive adenylyl cyclase system (EC 4.6.1.1). Mutations that lead to constitutive activation of Gs alpha, the guanine nucleotide-binding regulatory protein that stimulates adenylyl cyclase acti

Effect of light on the transfer of sugars from sugar nucleotides to rod outer segment membranes of control and dystrophic rats.

The transfer of N-acetyl-D-glucosamine (GlcNAc), D-mannose (Man), D-galactose (Gal) and L-fucose (Fuc) from their nucleotide complexes to isolated rod outer segment (ROS) membranes obtained from dark-adapted 21 +/- 2 days old dystrophic (RCS) and control (RCS-rdy+) rat retinas, was studied under light or dark conditions of incubation. It was found that all of these sugars were transferred to ROS membranes in the dark. Under these conditions there was significantly less (p less than 0.001) Gal transferred to dystrophic than to control membranes. Exposure to light affected the transfer of Gal an

ON THE SURFACE COMPOSITION OF NORMAL AND SENSITIZED MAMMALIAN BLOOD CELLS.

The interfacial tension method has been applied to the study of the surface composition of mammalian blood cells and to certain other particles. Unsensitized erythrocytes and stromata possess only a small margin of stability in the interface and pass readily into the oil phase. Specifically sensitized erythrocytes and stromata possess much greater stability in the interface and pass into the oil only with considerable mechanical aid; characteristic deformations of the erythrocyte surface or the interface or both often result. With special immune sera prepared by Landsteiner and van der Scheer

Denaturing gradient gel electrophoresis analysis of the tissue nonspecific alkaline phosphatase isoenzyme gene in hypophosphatasia.

Hypophosphatasia, a heritable form of rickets/osteomalacia, was first described in 1948. The biochemical hallmark, subnormal alkaline phosphatase (ALP) activity in serum, reflects a generalized disturbance involving the tissue-nonspecific isoenzyme of ALP (TNSALP). Deactivating mutations in the gene that encodes TNSALP have been reported in patients worldwide. Nevertheless, hypophosphatasia manifests an extraordinary range of clinical severity spanning death in utero to merely premature loss of adult teeth. There is no known medical treatment. To delineate the molecular pathology which explain

[Mild solubilization, purification, and immunoreactivity characterization of VP1 inclusion body protein of serotype O foot-and-mouth disease virus based on sodium lauroyl sarcosinate].

To efficiently prepare the capsid protein VP1 of serotype O foot-and-mouth disease virus (FMDV) with favorable immunoreactivity and to address the challenges that VP1 is prone to form inclusion bodies in the Escherichia coli expression system and is difficult to refold, this study developed a novel process based on sodium lauroyl sarcosinate (LSN) for the mild solubilization and chromatographic purification of FMDV VP1 inclusion body protein. First, the recombinant SUMO-VP1 protein was expressed in E. coli by induction, and Western blotting results revealed that the recombinant SUMO-VP1 protei

Comparative purification of a Salmonella bacteriophage for protein profiling.

Salmonella enterica bacteriophages are increasingly studied for bacterial biocontrol and applied microbiology. However, reliable downstream characterization depends strongly on how the phage material is purified and concentrated. This study compared five preparation strategies for a Salmonella bacteriophage: sucrose density gradient centrifugation, cesium chloride gradient centrifugation, ultrafiltration followed by lyophilization, ultrafiltration followed by ammonium sulfate precipitation, and ultrafiltration followed by trichloroacetic acid precipitation. The preparations were assessed for i

Aminoglycosides suppress the protein folding activity of the molecular chaperone HSC70: implication of a structure-activity relationship.

BACKGROUND: The mechanism of aminoglycoside (AG)-induced nephrotoxicity has not been fully elucidated. We previously reported that gentamicin suppresses the functions of HSC70, a heat shock cognate protein, which is a cytosolic molecular chaperone in mammalian cells. However, the relationship between HSC70 dysfunction and nephrotoxicity has not been clarified yet. METHODS: The effects of 11 AGs, including gentamicin, on the function, protein refolding and substrate binding of HSC70 were examined in vitro. The protein refolding activity of HSC70 was examined by enzymatic activity restoration o

The Amount of MUC5B mucin in cervical mucus peaks at midcycle.

The physical character and amount of mucus secreted by the endocervix changes dramatically during the menstrual cycle to facilitate sperm migration at the time of midcycle ovulation. Mucins are highly glycosylated, high-molecular-weight proteins, which are the major structural components of the protective mucus gel covering all wet-surfaced epithelia, including that of the endocervix. We have previously demonstrated that the endocervical epithelium expresses messenger RNA (mRNA) of three of the large gel-forming mucins, designated MUC5AC, MUC5B, and MUC6, with mRNA of MUC5B predominating. Evaluation of lactic acid as a novel fixative for histological and neuroanatomical applications.

Formaldehyde-based fixatives, particularly neutral-buffered formalin (NBF), are widely used in histology owing to their strong protein cross-linking capacity and reliable preservation of cytoarchitecture. However, formaldehyde is toxic and carcinogenic, motivating the search for safer alternatives. Here, we systematically evaluated lactic acid (LA) as a potential formaldehyde-free fixative for murine brain tissues. Brains were fixed either by immersion or by transcardial perfusion followed by immersion using LA at varying concentrations, pH-adjusted LA, or phosphate-buffered saline (PBS), and
