The ultrastructural morphology of air embolism: platelet adhesion to the interface and endothelial damage.

The pathogenesis of the ill effects following air embolism cannot be attributed solely to the space occupying and surface tension effects of the air bubbles altering the normal flow of blood through the vasculature. Decompression sickness was induced in rats and the following features of this process observed by electron microscopy in the vessels of the mesentery: imprisonment of blood elements (especially platelets) took place within the various enclosures created by the boundaries set up by different sized air bubbles between the layer of blood and the vessel walls, and the air/blood interfa

New insights into the drying technology of Pleurotus eryngii: Effects on protein structure, properties and product flavor analysis.

Drying techniques have been widely used in food macromolecule modification, sterilization, and other fields. However, comparisons about the effects of different heat treatments on the structural and functional properties of proteins have yet to be elucidated. In this study, we compared the effects of boiling water, microwave, hot air, and infrared pretreatment on the structural and functional properties of P. eryngii (Pleurotus eryngii) proteins (PEP), with the addition of low-temperature hot air drying (Fresh-H) and vacuum freeze drying (VFD) as controls. The flavor of apricot mushroom produc

Identification of shared and divergent molecular pathways linking ambient air pollutants to mortality and major chronic diseases through proteomic profiling.

Ambient air pollutants have been associated with increased risks of mortality and chronic diseases; however, population-based evidence on their shared versus pollutant- and outcome-specific molecular mechanisms remains unclear. Leveraging proteomics data from 48,645 participants in the UK Biobank, we characterized proteomic signatures related to overall and individual air pollutants, including particulate matter (PM2.5 and PM10), nitrogen dioxide (NO2) and nitrogen oxides (NOx), and examined their associations with risks of mortality and major chronic diseases. Subsequently, we investigated po

Factors improving dry fractionation of Bambara groundnut aiming at production of protein-rich ingredients with elevated nutritional and technological quality.

Bambara groundnut is a nutritious African grain legume with potential to be refined into a high-protein ingredient. This work aimed to develop protein-rich ingredients from Bambara via dry processing. Due to Bambara's high fat content, two dry fractionation approaches were developed. In the first one, Bambara was defatted using supercritical CO2 extraction, milled and air classified producing ingredients with 48 % protein, achieving 74 % of total protein separation efficiency. Without defatting, the efficiency stood at 16.7 %. A second attempt, incorporating a flow aid during air classificatio

Impact of Surfactants on Silk Fibroin Self-Assembly at the Air-Water Interface.

Silk fibroin (SF)-based materials attract significant interest because of their biocompability and great diversity of possible morphologies. One of the approaches to obtain SF materials is the use of an air-water or oil-water interface as a template for protein self-assembly. Surfactants can change the surface properties of adsorbed SF layers by promoting or preventing the formation of SF fiber networks. This study focuses on the influence of two typical ionic surfactants, cationic cetyltrimethylammonium bromide (CTAB) and anionic sodium dodecyl sulfate (SDS), on the dynamic properties of SF l

Genetic disorders of surfactant homeostasis.

Adaptation to air breathing at birth requires the precise orchestration of cellular processes to initiate fluid clearance, enhance pulmonary blood flow, and to synthesize and secrete pulmonary surfactant needed to reduce surface tension at the air-liquid interface in the alveoli. Genetic programs regulating the synthesis of the surfactant proteins and lipids required for the production and function of pulmonary surfactant are highly conserved across vertebrates, and include proteins that regulate the synthesis and packaging of pulmonary surfactant proteins and lipids. Surfactant proteins B and

Co-existing "spear-and-shield" air filter: Anchoring proteinaceous pathogen and self-sterilized nanocoating for combating viral pandemic.

Infectious pollutants bioaerosols can threaten human public health. In particular, the indoor environment provides a unique exposure situation to induce infection through airborne transmission like SARS-CoV-2. To prevent the infection from spreading, personal protective equipment or indoor air purification is necessary. However, it has been discovered that the conventional filter can become contaminated by pathogen-containing aerosols, meaning that advanced filtering and self-sterilization systems are required. Here, we fabricate a multilayered nanocoating around the fabric using laponite (LAP

Nanoparticulate air pollution disrupts proteostasis in Caenorhabditis elegans.

The proteostasis network comprises the biochemical pathways that together maintain and regulate proper protein synthesis, transport, folding, and degradation. Many progressive neurodegenerative diseases, such as Huntington's disease (HD) and Alzheimer's disease (AD), are characterized by an age-dependent failure of the proteostasis network to sustain the health of the proteome, resulting in protein misfolding, aggregation, and, often, neurotoxicity. Although important advances have been made in recent years to identify genetic risk factors for neurodegenerative diseases, we still know relative

AIRE is induced in oral squamous cell carcinoma and promotes cancer gene expression.

Autoimmune regulator (AIRE) is a transcriptional regulator that is primarily expressed in medullary epithelial cells, where it induces tissue-specific antigen expression. Under pathological conditions, AIRE expression is induced in epidermal cells and promotes skin tumor development. This study aimed to clarify the role of AIRE in the pathogenesis of oral squamous cell carcinoma (OSCC). AIRE expression was evaluated in six OSCC cell lines and in OSCC tissue specimens. Expression of STAT1, ICAM1, CXCL10, CXCL11, and MMP9 was elevated in 293A cells stably expressing AIRE, and conversely, was dec

Repeated Gonadotropin Administration Suppresses T Cell Development in the Mouse Thymus.

Gonadotropins, such as follicle-stimulating hormone (FSH) and human chorionic gonadotropin (hCG), are widely used to induce ovarian hyperovulation during in vitro fertilization and embryo transfer (IVF-ET) for the treatment of infertility. However, the effects of repeated administration of these gonadotropins on immune function, particularly on T cell development in the thymus, remain poorly understood. This study investigated the effects of repeated administration of pregnant mare serum gonadotropin (PMSG) and hCG on thymic T cell development in mice. Histological analysis revealed structural

Hepatic autoantigens in patients with autoimmune polyendocrinopathy-candidiasis-ectodermal dystrophy.

BACKGROUND & AIMS: Autoimmune polyendocrinopathy-candidiasis-ectodermal dystrophy (APECED) is caused by mutations of both copies of the autoimmune regulator (AIRE) gene. It is characterized by susceptibility to mucocutaneous candidiasis and multiple autoimmune lesions. A serious disease component is hepatitis. To identify diagnostic autoantibodies for APECED hepatitis, sera from 64 patients with APECED were screened for autoantibodies established in the diagnosis of idiopathic autoimmune hepatitis, and for autoantibodies against 10 cytochrome P450s. METHODS: Screening methods were indirect im

Study of Monoclonal Antibody Aggregation at the Air-Liquid Interface under Flow by ATR-FTIR Spectroscopic Imaging.

Throughout bioprocessing, transportation, and storage, therapeutic monoclonal antibodies (mAbs) experience stress conditions that may cause protein unfolding and/or chemical modifications. Such structural changes may lead to the formation of aggregates, which reduce mAb potency and may cause harmful immunogenic responses in patients. Therefore, aggregates need to be detected and removed or ideally prevented from forming. Air-liquid interfaces, which arise during various stages of bioprocessing, are one of the stress factors causing mAb aggregation. In this study, the behavior of an immunoglobu

Aire.

Mutations in the transcriptional regulator, Aire, cause APECED, a polyglandular autoimmune disease with monogenic transmission. Animal models of APECED have revealed that Aire plays an important role in T cell tolerance induction in the thymus, mainly by promoting ectopic expression of a large repertoire of transcripts encoding proteins normally restricted to differentiated organs residing in the periphery. The absence of Aire results in impaired clonal deletion of self-reactive thymocytes, which escape into the periphery and attack a variety of organs. In addition, Aire is a proapoptotic fact

Air pollution-regulated E-cadherin mediates contact inhibition of proliferation via the hippo signaling pathways in emphysema.

Air pollution has been linked to emphysema in chronic obstruction pulmonary disease (COPD). However, the underlying mechanisms in the development of emphysema due to air pollution remain unclear. The objective of this study was to investigate the role of components of the Hippo signaling pathway for E-cadherin-mediated contact inhibition of proliferation in the lungs after air pollution exposure. E-Cadherin-mediated contact inhibition of proliferation via the Hippo signaling pathway was investigated in Sprague-Dawley (SD) rats whole-body exposed to air pollution, and in alveolar epithelial A54

From Inhalation to Neurodegeneration: Air Pollution as a Modifiable Risk Factor for Alzheimer's Disease.

Air pollution, a growing concern for public health, has been linked to various respiratory and cardiovascular diseases. Emerging evidence also suggests a link between exposure to air pollutants and neurodegenerative diseases, particularly Alzheimer's disease (AD). This review explores the composition and sources of air pollutants, including particulate matter, gases, persistent organic pollutants, and heavy metals. The pathophysiology of AD is briefly discussed, highlighting the role of beta-amyloid plaques, neurofibrillary tangles, and genetic factors. This article also examines how air pollu

The Role of the Autoimmune Regulator Gene in the Control of MHC II Antigen-Processing and Presentation by Medullary Thymic Epithelial Cells.

The autoimmune regulator (AIRE) gene deficiency in humans causes the autoimmune polyglandular syndrome type 1 (APS-1). AIRE contributes to self-tolerance and prevents autoimmunity via the regulation of tissue-restricted antigen (TRA) expression in medullary thymic epithelial cells (mTECs). Using a murine Aire-deficient mTEC line (Aire-/-), we previously reported that Aire regulates cell adhesion genes, affecting homotypic mTEC/mTEC and heterotypic mTEC/thymocyte interaction in vitro. Transcriptional analysis comparing AireWT with Aire-/- mTECs indicates that Aire modulates antigen processing a

Proteomic signatures of ambient air pollution and risk of type 2 diabetes.

Despite accumulating evidence linking air pollution to type 2 diabetes (T2D), the underlying mechanisms remain largely unexplored. We aimed to profile proteomic signatures associated with air pollution and examine their relationship to T2D. We conducted proteome-wide association studies on 2911 plasma proteins among 49,134 UK Biobank participants. Exposures to fine particulate matter (PM2.5), nitrogen dioxide (NO2), sulfur dioxide (SO2), and benzene were estimated based on residential addresses. Proteomic signatures and their corresponding scores for each air pollutant were identified using li
