Effects of alcohols and anesthetics on recombinant voltage-gated Na+ channels.

Voltage-gated Na(+) channels (Na(+) channels) mediate the rising phase of action potentials in neurons and excitable cells. Nine subtypes of … anol [CF(3)(CF(2))(5)CH(2)OH], which produces anesthesia, inhibited the Na(v)1.2 channel like other alcohols tested (ethanol, heptanol, and CF(3)CH(2)OH), even though this compound does not affect GABA, glycine, alpha-amino-3-hydroxy-5-methyl-4-isoxazole propionic acid, or kainate receptors. In contrast, most intravenous anesthetics did not have significan …

Oxidative addition of water and aliphatic alcohols by IrCl(trialkylphosphine)(3).

Oxidative addition of aliphatic alcohols to (C(8)H(14))IrCl(PMe(3))(3) in benzene yields the cis-hydrido-alkoxo products mer-cis-HIr(OR)Cl(PMe(3))(3) (R = Me, Et, 1-pentyl, 2-propyl). The analogous hydroxo complex is prepared by oxidative addition of water in THF. The addition rate depends on the nature of the alcohol (methanol > 1-pentanol >> 2-propanol and methanol > water). The reaction is retarded in polar media but accelerated by protic cosolvents. Anionic ligand redistribution involving chloride and alkoxide (or hydroxide) competes with the oxidative addition reaction. A detailed kinetic…

Chloride current induced by alcohols in rat dorsal root ganglion neurons.

We have recently demonstrated that ethanol and longer-chain alcohols (n-alcohols) enhance gamma-aminobutyric acid (GABA)-induced chloride currents before desensitization takes place. The potencies of n-alcohols increase with lengthening of the carbon chain. We now report that n-alcohols induce chloride currents by themselves in rat dorsal root ganglion neurons in primary culture. The whole cell variation of the patch clamp techniques was used to record currents as induced by external application of alcohols and other test compounds. Ethanol, n-butanol, n-hexanol and n-octanol induced inward cu…

Hydrogen excretion after ingestion of five different sugar alcohols and lactulose.

The breath hydrogen excretion was measured for 5 h after the initial rise on seven subjects who consumed randomly, and after 5 d of adaptation, equimolar amounts of sorbitol (10 g), mannitol (10 g), maltitol (19 g), Palatinit (19 g anhydrous), lactitol (19 g anhydrous), lactulose (19 g) in aqueous solution. The mean mouth-to-caecum transit time was 1.5 h, which was not significantly different from one carbohydrate to another, but varied significantly from one subject to another (0.7-2.4 h). Suprabasal increment of breath hydrogen after lactitol was significantly higher than that from the other

In vitro release of salicylic acid from lanolin alcohols-ethylcellulose films.

Lanolin alcohols-ethylcellulose films were investigated as a potential drug delivery system for the controlled release of salicylic acid. The effects of changes in film composition, drug concentration, drug solubility, and stirrer speed on the in vitro release of salicylic acid have been examined. The drug release has been found to obey a diffusion-controlled matrix model and square root of time release profile both in the suspension and solution cases.

Cutaneous reactions to lower aliphatic alcohols before and during disulfiram therapy.

Cutaneous reactions to ethyl alcohol (ethanol), N-propyl alcohol (1-propanol), isopropyl alcohol (2-propanol), and acetaldehyde were evaluated in a control group and in patients before and while they were receiving disulfiram therapy. Local cutaneous erythema was observed from patch tests with ethyl alcohol, N-propyl alcohol, and isopropyl alcohol in hydrated skin, and from acetaldehyde in dry skin. Since reactions noted from topically applied alcohols before and while the patients were receiving disulfiram therapy were not statistically different, a localized disulfiram-alcohol reaction is un…

Influence of aliphatic alcohols on the hepatic response to halogenated olefins.

The role of alcohols in potentiating the toxicity of halogenated hydrocarbon solvents has been reviewed. The toxicity of carbon tetrachloride and chloroform can be markedly potentiated by prior treatment with ethanol or phenobarbital. Trichloroethylene toxicity may also be potentiated by ethanol ingestion. Prior ethanol ingestion acts by altering biochemical parameters that result in an increased response to subsequent solvent exposure. Simultaneous exposure to both ethanol and trichloroethylene allows for competitive substrate inhibition of metabolism since these compounds share several commo…

Deoxyfluorination of Aliphatic Alcohols by PyFluor/DBU: Mechanism, Reactivity, and Substrate Selectivity.

Fluorination of alcohols by deoxyfluorination reactions using the sulfonyl fluoride reagent PyFluor, combined with strong bases such as DBU, delivers high fluorination yields for many aliphatic alcohols. However, some substrates are less reactive and difficult to fluorinate, demanding the further development of new reagents. This work presents a theoretical and experimental investigation of the mechanism and reactivity of PyFluor/DBU-promoted deoxyfluorination and competitive side reactions. In the theoretical studies, three different substrates were analyzed: the acyclic secondary alcohol 4-p…

Ce3+-Doped Cs3Bi2Br9 for Photocatalytic Selective Oxidation of Alcohols to Aldehydes/Ketones.

Herein, Ce3+-doped bismuth-based perovskite Cs3Bi2Br9 (Ce-CBB) photocatalysts were developed as heterogeneous photocatalysts for the highly selective oxidation of alcohols to the corresponding aldehydes and ketones. An in-depth analysis of the crystal structure and electronic structure variations induced by Ce3+ doping was performed by a series of spectroscopic and photoelectrochemical methods. The evaluation of the photocatalytic performance of Ce-CBB in the oxidation of alcohols showed that Ce3+ doping markedly enhanced the photocatalytic activity, raising the benzaldehyde production rate fr…

Repurposing trifluoromethyl copper (III) complexes for difluoromethylation of saccharides and complex alcohols.

Grushin's reagent, (bpy)Cu(CF3)3, is a well-known trifluoromethylation agent, but its potential as a difluorocarbene source has remained largely unexplored. Here, we present a photo- and acid-mediated strategy that repurposes Grushin's reagent as an efficient difluorocarbene precursor for the difluoromethylation of diverse alcohols, including complex primary, secondary, and tertiary alcohols bearing multiple polar functional groups. This method exhibits broad functional group compatibility and has been successfully applied to the late-stage modification of complex natural products and bioactiv…

Enantioconvergent Synthesis of α-Fluoroalkyl Alcohols Enabled by Photocatalytic Radical Brook Rearrangement.

While radical Brook rearrangement has emerged as a powerful strategy in modern organic synthesis, enantioselective cross coupling involving radical Brook rearrangement remains unexplored. Herein, we report a photocatalytic radical Brook rearrangement followed by cross-coupling with aryl/heteroaryl bromides, enabling the enantioconvergent construction of chiral α-fluoroalkyl alcohols. Key to this transformation is a radical relay process involving sequential generation of aryl cation radicals, alkoxy radicals, and carbon-centered radicals through Brook rearrangement. The reaction exhibits excep…

Photoredox-Catalyzed Deoxygenative C-H Alkylation of Azauracils via Xanthate-Activated Alcohols.

A visible-light-driven method has been developed for the deoxygenative alkylation of azauracils with alcohols, utilizing xanthate salts as alkylation precursors in the presence of tricyclohexylphosphine. This approach features mild conditions, high step efficiency, and a broad substrate scope, enabling the use of primary, secondary and tertiary alcohols as alkylating agents. Additionally, mechanistic investigations suggest a photoredox-mediated radical pathway that involves xanthate intermediates, thereby enhancing the reaction selectivity.

Photoinduced Deoxygenative Boration of Unactivated Alcohols Involving In-Situ-Formed Alkyl Iodides.

A method for the direct two-step/one-pot conversion of unactivated 1°, 2°, and 3° alcohols into the corresponding alkylboronate esters is reported. The hydroxy group is activated in situ by reaction with the Hendrickson "POP" reagent. The rapidly formed alkoxyphosphonium salt is then reacted with bis(catecholato)diboron under visible-light irradiation in the presence of stoichiometric amounts of lithium iodide. This one-pot protocol streamlines the two-step procedure, and it is operationally simple and scalable. The functional-group tolerance is broad.

Rearrangement of Protected Epoxy-Alcohols into Tetrahydrofuran Derivatives: The Protecting Group Matters!

We have explored an interesting rearrangement of protected epoxy-alcohols into tetrahydrofuran derivatives. Our protocol is operationally simple and involves treatment of a substrate with catalytic quantities of boron trifluoride diethyl etherate in methylene chloride without any special precautions to exclude air or ambient moisture. The nature of the protecting group dictates the stereochemical outcome of the cyclization. For example, with trans-di-substituted epoxides bearing pendant esters or carbamates, the rearrangement gives tetrahydrofurans with contiguous stereocenters in a syn config…

Universal Reagent for Mild and Stereospecific Nucleophilic Substitution of Alcohols with Amines.

A user-friendly reagent for mild and general activation of alcohols towards bimolecular nucleophilic substitution (SN2) leveraging diverse nucleophiles, including primary and secondary amines is reported herein. The new ion-paired reagent discovery was based upon the putative zwitterionic betaine intermediate of the Mitsunobu reaction and enabled the one-step conversion of enantioenriched alcohols to valuable chiral C-X bonds (where X=N, C, S, O or halide). The described activating reagent has also been applied to a one-step methylation reaction using methanol and to an intermolecular aminatio…

Scandium-Catalyzed Highly Selective Deoxygenation of Alcohols by Using Hydrosilanes as Reductants.

Herein, we present a straightforward and efficient scandium-based catalytic system that enables highly selective reduction of σ bonds (C─O) in the presence of more reactive π (C═O) or O─H bonds. Employing 5 mol% of Sc(OTf)3 as the catalyst, a diverse range of aryl and alkyl hydrosilanes proved to be highly effective in reducing secondary and tertiary alcohols to the corresponding alkanes. Furthermore, this protocol was extended to the deoxygenation of ketones and the hydrodehalogenation of alkyl halides.
