Compound action potential and single acoustic nerve fibres activity generation: an equivalent neuron approach.

The peripheral acoustic system can be easily subdivided into three distinct subsystems: a mechanical one (middle ear and basilar membrane), the set of mechanical-to-neural transducers (the inner and outer hair cells) and the neural part. The functional behaviour of the external and middle ear is well known, but the processes concerned with the transduction of mechanical motion into neural activity are still under investigation. Actually the mechanism of transmission of information concerning acoustic stimuli to the central nervous pathways is not clearly known. On the basis of physiological kn…

Comparison of round window and cochleostomy approaches with a prototype hearing preservation electrode.

INTRODUCTION: Preservation of residual hearing in cochlear implant recipients has been demonstrated to be possible and provides the potentia … l scala tympani cochleostomy. The cochleostomy was sited inferior to the RWM to avoid trauma to the basilar membrane and spiral ligament. Specimens were embedded and fixed with acrylic resin and the cochleae then examined histologically at 200-mum intervals using a grinding and polishing technique. RESULTS: Full insertion of the electrode was achieved without sig …

Voltage-gated K+ channel dysfunction in myocytes from a dog model of subarachnoid hemorrhage.

Delayed cerebral vasospasm after subarachnoid hemorrhage is primarily due to sustained contraction of arterial smooth muscle cells. Its pathogenesis remains unclear. The degree of arterial constriction is regulated by membrane potential that in turn is determined predominately by K+ conductance (GK). Here, we identified the main voltage-gated K+ (Kv) channels contributing to outward delayed rectifier currents in dog basilar artery smooth muscle as Kv2 class through a combination of electrophysiological and pharmacological methods. Kv2 current density was nearly halved in vasospastic myocytes a

Cochlear partition anatomy and motion in humans differ from the classic view of mammals.

Mammals detect sound through mechanosensitive cells of the cochlear organ of Corti that rest on the basilar membrane (BM). Motions of the BM and organ of Corti have been studied at the cochlear base in various laboratory animals, and the assumption has been that the cochleas of all mammals work similarly. In the classic view, the BM attaches to a stationary osseous spiral lamina (OSL), the tectorial membrane (TM) attaches to the limbus above the stationary OSL, and the BM is the major moving element, with a peak displacement near its center. Here, we measured the motion and studied the anatomy…

Intracytoplasmic retention of type VII collagen and dominant dystrophic epidermolysis bullosa: reversal of defect following cessation of or marked improvement in disease activity.

It has been recently shown that the presence of perinuclear "stellate bodies" within the epidermis in patients with a form of dominant dystrophic epidermolysis bullosa named "transient bullous dermolysis of the newborn" corresponds to collections of type VII collagen. To determine the temporal relationship of this unique immunohistochemical defect with course of clinical disease activity, we have longitudinally studied the expression of two epitopes of type VII collagen (LH 7:2; L3d) in nine patients in four such kindreds by immunofluorescence and immunoelectron microscopic technique. In every

Finite Element Modeling of Organ of Cortis Throughout the Cochlea During Blast Wave Transmission.

INTRODUCTION: Exposure to blast overpressures in the line of duty causes many service members and veterans to suffer inner ear injuries and the resulting hearing disabilities. The response to blast waves within the cochlea, particularly by the sensory hair cells within the organ of Corti (OC), is challenging to measure experimentally. Recently, 3D finite element (FE) models of the human ear at macroscale and OC models at microscale have been established to examine the blast transmission throughout the ear and the OC response at the middle turn of the basilar membrane (BM). This multiscale mode…

A quantitative model of the "effective" signal processing in the auditory system. I. Model structure.

This paper describes a quantitative model for signal processing in the auditory system. The model combines a series of preprocessing stages with an optimal detector as the decision device. The present paper gives a description of the various preprocessing stages and of the implementation of the optimal detector. The output of the preprocessing stages is a time-varying activity pattern to which "internal noise" is added. In the decision process, a stored temporal representation of the signal to be detected (template) is compared with the actual activity pattern. The comparison amounts to calcul

Cochlear hair cell and vascular changes in the guinea pig following high level pure-tone exposures.

Noise is thought to exert metabolic and/or mechanical stress on sensory and vascular tissues of the cochlea, the relative influence of the stressors being influenced by the intensity of the noise. Guinea pigs exposed to either of two pure-tone frequencies, 1.33 or 3.85 kHz for 6 hours at intensity levels ranging from 102 dB to 120 dB SPL, were studied for pathological changes in two spiral lamina vessels--the vessel of the basilar membrane (VSBM) and the vessel of the tympanic lip (VSTL). In general, animals sustaining mild to severe degrees of hair cell destruction one month after noise expos…

Adrenergic stimulation of ciliary process epithelium causes surface membrane internalization.

An ultrastructural change induced in the nonpigmented epithelium (NPE) of the ciliary processes by adrenergic stimulation in the albino rabbit was studied. Thirty min after topical treatment with 2% isoproterenol, an extensive intracellular membranous network, previously reported to be smooth endoplasmic reticulum, was revealed by electron microscopy. It was postulated that this network originated from the plasma membrane. Using cationized ferritin (CF) as an ultrastructural tracer, freshly isolated anterior segments were incubated in buffer containing 10(-5) M isoproterenol and 0.2% CF. As ea

Moving Past Tonsil Position: Craniocervical Junction Crowding Shapes Cerebrospinal Fluid Effective Motility in Chiari I Malformation.

Chiari I malformation (CM-I) is conventionally defined by cerebellar tonsil position, yet tonsil position is an indirect surrogate for the anatomic obstruction that impairs cerebrospinal fluid (CSF) flow across the craniocervical junction (CCJ). We hypothesized that CCJ crowding, quantified as subarachnoid space narrowing at the foramen magnum and C1, would explain CSF flow impairment more directly than tonsil position. Using non-invasive low b-value diffusion-weighted MRI (low-b dMRI), we quantified effective CSF motility, indexed by mean pseudo-diffusivity (MΨ), across the upper cervical spi

Vertebral and Basilar Artery Dissection in a Patient With Alport Syndrome.

Basilar artery occlusion (BAO) is a rare cause of stroke associated with significant morbidity and mortality. It is most frequently thromboembolic in nature, but may be caused by vertebral artery dissection. We present a case of BAO in a 36-year-old woman with Alport syndrome. She was treated with emergent thrombectomy via the right vertebral artery with return to baseline neurological status. Her clinical status deteriorated later the same day and she was found to have re-occlusion. Repeat thrombectomy was complicated by persistent re-occlusion requiring 7 passes to achieve reperfusion. Unfor

[Numerical analysis of the influence of otitis media on the hearing compensation performance of round-window stimulation].

In order to study the influence of tympanic membrane lesion and ossicular erosion caused by otitis media on the hearing compensation performance of round-window stimulation, a human ear finite element model including cochlear asymmetric structure was established by computed tomography (CT) technique and reverse engineering technique. The reliability of the model was verified by comparing with the published experimental data. Based on this model, the tympanic membrane lesion and ossicular erosion caused by otitis media were simulated by changing the corresponding tissue structure. Besides, thes

Microanatomy of the human tunnel of Corti structures and cochlear partition-tonotopic variations and transcellular signaling.

Auditory sensitivity and frequency resolution depend on the optimal transfer of sound-induced vibrations from the basilar membrane (BM) to the inner hair cells (IHCs), the principal auditory receptors. There remains a paucity of information on how this is accomplished along the frequency range in the human cochlea. Most of the current knowledge is derived either from animal experiments or human tissue processed after death, offering limited structural preservation and optical resolution. In our study, we analyzed the cytoarchitecture of the human cochlear partition at different frequency locat…
