Circadian rhythms in bed rest: Monitoring core body temperature via heat-flux approach is superior to skin surface temperature.

Continuous recordings of core body temperature (CBT) are a well-established approach in describing circadian rhythms. Given the discomfort of invasive CBT measurement techniques, the use of skin temperature recordings has been proposed as a surrogate. More recently, we proposed a heat-flux approach (the so-called Double Sensor) for monitoring CBT. Studies investigating the reliability of the heat-flux approach over a 24-hour period, as well as comparisons with skin temperature recordings, are however lacking. The first aim of the study was therefore to compare rectal, skin, and heat-flux tempe

Endothelial function after 10 days of bed rest in individuals at risk for type 2 diabetes and cardiovascular disease.

Physical inactivity is considered to be deleterious to vascular health, and in particular in first-degree relatives to patients with type 2 … a the Danish Birth Registry. Twenty LBW, 20 CON and 13 FDR were studied before and after 10 days of bed rest. Forearm blood flow (FBF) was measured by venous occlusion plethysmography during brachial intra-arterial infusion of acetylcholine or adenosine at baseline and with superimposed hyperinsulinaemia. Markers of endothelial activation and inflammation …

Prolonged head down bed rest-induced inactivity impairs tonic autonomic regulation while sparing oscillatory cardiovascular rhythms in healthy humans.

BACKGROUND: Physical inactivity represents a major risk for cardiovascular disorders, such as hypertension, myocardial infarction or sudden death; however, underlying mechanisms are not clearly elucidated. Clinical and epidemiological investigations suggest, beyond molecular changes, the possibility of an induced impairment in autonomic cardiovascular regulation. However, this hypothesis has not been tested directly. METHODS: Accordingly, we planned a study with noninvasive, minimally intrusive, techniques on healthy volunteers. Participants were maintained for 90 days strictly in bed, 24 h a

Compression and walking versus bed rest in the treatment of proximal deep venous thrombosis with low molecular weight heparin.

OBJECTIVE: The purpose of this randomized controlled trial was to evaluate the benefits of compression and walking exercises in comparison with bed rest in the acute stage of proximal deep venous thrombosis (DVT). METHODS: Forty-five patients with proximal DVT that was proved with compression ultrasound scan or phlebography were randomized into three groups. Group A consisted of 15 patients who received inelastic compression bandages (Unna boots on the lower leg, adhesive bandages on the thigh), and group B consisted of 15 patients who received thigh-length compression stockings, class II. Gr…

Resting-state connectivity of the bed nucleus of the stria terminalis and the central nucleus of the amygdala in clinical anxiety.

BACKGROUND: The central nucleus of the amygdala and bed nucleus of the stria terminalis are involved primarily in phasic and sustained aversive states. Although both structures have been implicated in pathological anxiety, few studies with a clinical population have specifically focused on them, partly because of their small size. Previous work in our group used high-resolution imaging to map the restingstate functional connectivity of the bed nucleus of the stria terminalis and the central nucleus of the amygdala in healthy subjects at 7 T, confirming and extending structural findings in huma

Validation of an angiographic method for estimating resting blood flow to distal tissue beds in the lower extremities.

PURPOSE: The authors propose to validate an angiographic scoring system that estimates the resting blood flow in the lower extremities in patients with peripheral vascular disease (PVD). MATERIALS AND METHODS: Twenty patients with PVD underwent lower-extremity angiography. For each patient, three readers used the proposed scheme to estimate flow within 23 separate vessel segments from the infrarenal aorta to the pedal vessels. Each reviewer scored each angiogram twice. The scheme includes not only an assessment of stenoses and occlusions but also corrections for collaterals. Flow reductions t

Preserved mitochondrial respiration in presence of oxidative stress and reduced mitochondrial mass after 10-day bed rest in older adults.

Ageing affects mitochondrial integrity in skeletal muscle, and physical inactivity may further exacerbate these changes. Although mitochondrial alterations are documented in ageing and disuse independently, how disuse impacts the mitochondrial phenotype in older populations remains unclear. This work aimed to characterise how physical inactivity impacts mitochondrial function, morphology and gene expression in the skeletal muscle of older adults. Ten healthy older men (65+ years) underwent 10 days of bed rest. Skeletal muscle biopsies were collected before and after bed rest to assess mitochon…

Muscle Weakness and the Irisin-BDNF and Oxidative Stress Axis in the 60-Day Pseudorandomised Controlled AGBRESA Bed Rest Study.

BACKGROUND: Muscle atrophy and weakness are among the most detrimental consequences of disuse, microgravity, hospitalisation and ageing. Oxidative modifications of myofibrillar proteins generated by oxidative stress may contribute to the reduced force- and power-generating capacity of skeletal muscles. As part of the 60-day AGBRESA bed rest (BR) study, we studied (1) how microgravity-induced disuse affected markers of systemic and muscle oxidative stress, (2) how these related to muscle function and (3) to what extent artificial gravity (AG) attenuated these changes. Since the myokine irisin m…

Aerobic Exercise and Subsequent Venoconstrictive Thigh Cuffs Preserve Cardiorespiratory Fitness but Not Muscle Function after 30 Days of Bed Rest.

PURPOSE: During spaceflight, countermeasures are crucial to counteract cardiorespiratory and musculoskeletal deconditioning, similar to effects seen after prolonged immobilization. Within a bed rest study simulating the effects of microgravity, we investigated the effectiveness of submaximal aerobic exercise and postexercise venoconstrictive thigh cuffs (VTC) for maintaining cardiorespiratory fitness and muscle function. METHODS: Twenty-four males and females were randomly allocated to a countermeasure ( n = 12, age = 34 ± 9 yr, body mass index = 22.9 ± 2.9 kg·m -2 ) or control group ( n = 12…

Altered cerebral perfusion in response to chronic mild hypercapnia and head-down tilt Bed rest as an analog for Spaceflight.

PURPOSE: Following prolonged stays on the International Space Station (ISS), some astronauts exhibit visual acuity changes, ophthalmological findings, and mildly elevated intracranial pressures as part of a novel process called spaceflight-associated neuro-ocular syndrome (SANS). To determine the pathophysiology of SANS, NASA conducted a multi-investigator study in which 11 healthy participants underwent head-down tilt bed rest, mimicking microgravity-induced cephalad fluid shifts, combined with elevated ambient CO2 levels similar to those on the ISS (HDT+CO2). As part of that study, we examin…

A single day of bed rest, irrespective of energy balance, does not affect skeletal muscle gene expression or insulin sensitivity.

NEW FINDINGS: What is the central question of this study? What are the initial metabolic and molecular events that underpin bed rest-induced skeletal muscle deconditioning, and what is the contribution of energy balance? What is the main finding and its importance? A single day of bed rest, irrespective of energy balance, did not lead to overt changes in skeletal muscle gene expression or insulin sensitivity. More than 1 day of physical inactivity is required to observe the insulin resistance and robust skeletal muscle transcriptional responses associated with bed rest and consequent alteratio…

Exercise during artificial gravity preserves cardiorespiratory fitness but not orthostatic tolerance following 60 days of head-down bed rest (BRACE).

Exercise is a critical countermeasure to prevent cardiovascular deconditioning during spaceflight; however, exercise does not protect astronauts from postflight orthostatic intolerance. Artificial gravity (AG) by short-arm centrifugation can attenuate reductions in orthostatic tolerance following prolonged head-down bed rest (HDBR), but AG does not protect cardiorespiratory fitness. The European Space Agency hypothesized that exercise and AG countermeasures could be applied simultaneously to protect both cardiorespiratory fitness and orthostatic tolerance following prolonged HDBR. Twenty-four …

Sex differences between postmenopausal women and similar-age men in response to orthostatic stress following 2 wk of bed rest.

Reduced orthostatic tolerance is common following periods of bed rest that are associated with illness or surgery, putting individuals at higher risk for syncope and falls following hospitalization. Following menopause, mechanisms of female cardiovascular regulation change, which may be associated with sex-specific responses to orthostatic stress following bed rest. The purpose of our experiment was to investigate sex differences between healthy postmenopausal women and similar-age men (age: 55-65 yr) for their orthostatic tolerance and cerebrovascular responses to standing following bed rest.…
